    Notes: Summary Na+/K+-ATPase related strophanthidin sensitive 3-O-methylfluorescein-phosphatase activity, [3H]ouabain binding and expression of Na+/K+-ATPase subunit isoforms were measured in the left ventricle of the heart of normal and streptozotocin-diabetic rats with and without insulin treatment. Compared to control animals, the enzyme activity was 0.75 ± 0.09 and 0.62 ± 0.06 times lower in rats diabetic for 2 and for 4 weeks, respectively. This was associated with a proportional decrease of the [3H]ouabain binding sites. Immunoblots indicated a 0.76 ± 0.08 and 0.61 ± 0.08-fold decrease of alpha1, a 0.68 ± 0.09 and 0.41 ± 0.04-fold decrease of alpha2 subunit in 2- and 4-week diabetic rats, respectively relative to controls. Beta1 subunit decreased proportionally 0.71 ± 0.07 and 0.38 ± 0.06-fold, and beta2 decreased 0.75 ± 0.08 and 0.31 ± 0.06-fold, respectively. Northern blot analysis revealed a significant reduction in mRNA level of Na+/K+-ATPase subunit isoforms after 2 and 4 weeks of diabetes (for alpha1 66.2 ± 8.2 and 55.9 ± 7.8 % of controls for alpha2 91.7 ± 12.1 and 41.1 ± 7.1 % of controls and for beta subunit 93.4 ± 11.1 and 49.8 ± 6.8 % of controls, respectively). Although, mRNA levels of isoform reverted to even higher levels than the control values after insulin treatment, insulin caused only a partial recovery of enzyme activity, [3H]ouabain binding capacity and protein expression. We have obtained evidence that in cardiac left ventricle there are more than one type of Na+/K+-ATPase alpha and beta subunit isoforms which are affected in diabetes and by insulin treatment. The time course of diabetes induced changes and the degree of involvement suggest that the Na+/K+-ATPase isoforms are altered individually. [Diabetologia (1997) 40: 1255–1262]

    Notes: Summary Identified neurons in the dorsal lateral geniculate nucleus (dLGN) of the rat were studied using a Golgi/EM procedure in combination with post-embedding GABA immunocytochemistry. The results of this investigation identify clearly the GABA-immunoreactive nature of the two morphologically] distinct neuronal populations found within the rat dLGN — thalamocortical relay cells are GABA-immunonegative whilst the putative ‘interneurons’ are GABA-immunopositive. This immunocytochemical evidence strongly suggests that ‘interneurons’ synthesize and use GABA to mediate their local inhibitory synaptic mechanisms within the dLGN of the rat.

    Notes: Summary Residual neurons following chronic surgical disconnection from the cortex were studied by light and electron microscopy in the dorsal Lateral Geniculate Nucleus (LGN) of adult cats. One year after operation the volume of the decorticated LGN had shrunken to approximately half that of the control LGN. The number of nerve cells decreased at the same time, to 13–15% of the control, with relatively higher cell loss in A and A1 laminae than in lamina C. The Golgi, Golgi-EM and EM analysis of the residual neurons revealed that they contain two distinct nerve cell types. (1) 55% of all surviving neurons were identified as geniculo-cortical relay cells, while (2) 45% of the persisting nerve cells were interneurons. These data suggest that in the normal LGN of adult cat 7% (or more) of all nerve cells are local interneurons. Finally, those factors which might contribute to the unexpected survival of many relay neurons to the axotomy-caused retrograde degeneration, are considered and discussed.

    Notes: Summary The corona radiata containing all geniculo-cortical and cortico-geniculate fibers was transected in adult cats. One year after the operation synaptic architecture of laminae A and A1 of the decorticated LGN was investigated utilizing electron-microscopy and Golgi-EM. It was found that synaptic glomeruli became embedded in heavily gliotic neuropile, which contained the dendrites of surviving relay cells in addition to retinal axons and interneuronal processes. In contrast to the intact LGN, where the relay cell dendrites are exclusively postsynaptic, many morphologically identified relay neuronal dendrites of the decorticated LGN also contained large, round synaptic vesicles associated with distinct presynaptic sites. In 60% of all glomeruli, the large presynaptic dendrites of relay neurons occupied a central position and were postsynaptic to retinal axons and interneuronal dendrites/axons, but were presynaptic to other relay dendrites or interneuronal dendritic profiles. Occasional dendro-dendritic synapses between relay cell processes could be observed in the extraglomerular neuropile. Relay cell presynaptic dendrites formed unequivocal Gray I-type synapses, in accordance with the supposed excitatory character of relay cell output. Since sprouting of extrinsic or intrinsic axons could not be observed after decortication, it is proposed that the partial synaptic reorganization of the LGN was primarily due to the compensatory “axonization” of relay cell dendrites. It is further suggested that the electrophysiologically demonstrated expansion of retinal receptive fields in the chronically decorticated LGN was accomplished by the formation of dendro-dendritic synapses between relay cells and interneurons.
