    Notes: Abstract The mutation of diploid human lymphoblasts by methylnitronitrosoguanidine (MNNG) was measured over the range of 0–45 ng of MNNG/ml of medium. We found a 12-day lag in the phenotypic expression of 6-thioguanine resistance; the occurrence of this lag was independent of MNNG concentration. We hypothesize that the unexpectedly long lag period reflects a requirement for the loss of previously existing molecules of the enzyme hypoxanthine-guanine phosphoribosyltransferase (HGPRT) after mutation at the HGPRT locus.

    Notes: Six enzymes associated with the activities of the nucleus (thymidine kinase), mitochondria (succinate dehydrogenase), lysosomes (acid phosphatase), peroxisomes (catalase), (cytosol lactate dehydrogenase), and the intermembranal mitochondrial space (alkaline DNase) were assayed at 2 hr intervals over the division cycle of repetitively resynchronized HeLa cells. The results indicated a high degree of reproductibility for cells synchronized by the method of perpetual resynchronization and may be of direct use to those interested in subcellular organellogenesis.

    Notes: Unbalanced growth has been studied in HeLa cell cultures maintained in perpetual division synchrony by periodic inhibition of DNA synthesis. Observation of eight independent cellular parameters indicates that once synchrony has been established the degree of unbalanced growth in later cycles is not as great as is observed with the conventional “double-block” technique. This significant diminution of unbalanced growth appears to be related to the significantly shorter periods of inhibition of DNA synthesis used in the resynchronization process.

    Notes: A variety of diploid human fibroblast lines have been successfully grown to high densities (〉106 cell/ml) on recently developed microcarriers. Interferon induction using poly I·poly C and a superinduction procedure resulted in yields greater than 10,000 units/ml with one cell line. A direct comparison of microcarrier cultures to roller bottle cultures showed equivalent interferon yields on a per cell basis and some apparent differences relating to optimum inducer concentrations and kinetics of interferon accumulation.

    Notes: HeLa S3 cells in suspension culture are resynchronized in each successive generation by exposure to 0.25mM thymidine. The resynchronization approach appears to be superior to single- and double-block techniques because the period of effective inhibition of DNA synthesis is significantly shortened. Gram quantities of highly synchronous viable cells have been produced on a daily basis.
