    Notizen: Abstract Percutaneous transluminal angioplasty of aortoiliac and femoropopliteal atherosclerotic lesions can provide long-lasting hemodynamic improvement. High-dose aspirin is commonly prescribed as reocclusion prophylaxis, but low doses would be preferable because of fewer adverse effects. We performed a double-blind, randomized, controlled clinical trial in patients with peripheral vascular disease with lesions appropriate for angioplasty. We compared the efficacy and side effects of two doses of aspirin (50 mg vs. 900 mg daily) during a period of 12 months after angioplasty. A total of 359 patients were evaluated: 175 were randomly assigned to treatment with 900 mg aspirin daily and 184 to 50 mg aspirin a day. Thirty-nine patients developed restenosis at the angioplasty site; the cumulative percentage of event-free survival after 1 year (patency rate) was 85% in the 900-mg group and 84% in the 50-mg group. An equivalence test showed the two groups equivalent with respect to restenosis rates (P = 0.003 for an equivalence region of 〈 10% difference). Nine patients (5%) in the 900-mg group had serious gastrointestinal side effects (peptic ulcer, eight; erosive gastritis requiring transfusion, one) compared to two (peptic ulcer) in the 50-mg group (P = 0.03). The results of our study show that a dose of 50 mg aspirin a day is as effective as one of 900 mg for the prevention of restenoses after lower limb angioplasty, and that severe gastrointestinal side effects are less frequent.

    Notizen: Abstract The in situ process—laser ablation in combination with thermal evaporation of Tl2O—has turned out to be a preparation method for single-phase and epitaxial TlBa2Ca2Cu3O9 (1223) thin films with T c values up to 109 K. It was found by several groups that a partial substitution of Tl by Bi simplifies the phase development of the 1223 compound in the usual two-step process. We have investigated the influence of the Bi doping on the in situ growth. X-ray measurements show that the films consisted mainly of the 1223 compound. In 300-nm thin films there was no evidence of a Bi amount in the crystal structure, but thinner films (80 nm) show a small amount of Bi. We concluded that Bi doping supports the phase development of the 1223 compound only in an early stage of the film growth. The Bi-doped films have higher T c values up to 114 K, higher j c values up to 6 × 105 A/cm2 (77 K, 0 T), and lower surface resistances of 56 mΩ (77 K, 87 GHz) than the undoped films.

    Notizen: Abstract Thin film properties of technologically important materials (Si, GaAs, SiO2, WSix) have been measured by using a novel technique that combines secondary ion mass spectrometry (SIMS) and laser interferometry. The simultaneous measurement of optical phase and reflectance as well as SIMS species during ion sputtering yielded optical constants, sputtering rates and composition of thin films with high depth resolution. A model based on the principle of multiple reflection within a multilayer structure, which considered also transformation of the film composition in depth and time during sputtering, was fitted to the reflectance and phase data. This model was applied to reveal the transformation of silicon by sputtering with O 2 + ions. Special attention was paid to the preequilibrium phase of the sputter process (amorphization, oxidation, and volume expansion). To demonstrate the analytical potential of our method the multilayer system WSix/poly-Si/SiO2/Si was investigated. The physical parameters and the stoichiometry of tungsten suicide were determined for annealed as well as deposited films. A highly sensitive technique that makes use of a Fabry-Perot etalon integrated with a Michelson type interferometer is proposed. This two-stage interferometer has the potential to profile a sample surface with subangstroem resolution.

    Notizen: Abstract During analysis with SIMS (secondary ion mass spectroscopy) a HeNe laser beam was focussed on the sample surface. While sputtering Si with oxygen ions, the laser irradiation produced a strong increase of the target current and the SIMS intensities as well. This was found for lightly p-doped Si only, whereas no effect was observed for highly p-doped or n-doped Si. To explain this we assume that a depletion layer exists under the surface oxide layer and free charged carriers are created therein by laser excitation. The laser induced effects observed in the SIMS intensity or the target current can be used for measuring the profile of an ion beam or for measuring the alignment of an ion beam at a laser marked target. In addition, laser irradiation combined with SIMS allows one to measure qualitatively both the profile of the doping impurity and its electrically active part.

    Notizen: Abstract A sensitive interferometric technique has been applied for studying the thermal displacement of thin film heated by electron or ion beams. The steady state displacement has been measured and we discuss the dependence on material properties and film thickness showing that this method has a potential for in-situ monitoring of thin-film deposition or etching. Transient effects are studied in a thin quartz plate and the propagation velocity of thermal waves is measured.

Saxitoxin (STX) exclusively and reversibly blocked Na channels, the effect being fully described by a one-to-one reaction between STX and a receptor at the channel with an equilibrium dissociation constant,K S ,of 1.4 nM; the mean offset rate constant,k 2S ,was 1.76×10−2 sec−1 (16±1°C; pH 7.2), 1.7 times the value for tetrodotoxin (TTX). 3. At pH 5.6,k 2S was increased by a factor of 1.33 while the equilibrium STX effect was decreased in a way suggesting competition between STX and protons. 4. After pretreatment of nodes with 3.1 nM TTX the extra block on adding 9.0 nM STX as well as its relief on taking out STX of the TTX-STX mixture revealed transients in the time course of receptor occupation. 5. These non-monotonic time courses are incompatible with the idea of two independent blocking sites (for STX and TTX) per channel but could be quantitatively fitted by analog-computed curves assuming competition between STX and TTX for the same site.

Voltage clamp experiments were done on single myelinated nerve fibres of the frog,Rana esculenta. 2. 53 μM 4-aminopyridine (4-AP) reducedI K to about one-fifth if tested with infrequent (1/min) and short (10 ms) depolarizing pulses; the onset time constant under these circumstances was ca. 160 s (14–15° C). After prolonged treatment the effect was virtually irreversible. 3. At equilibrium with 4-AP, increasing the frequency of short pulses removed part of the block, the block removal accelerating with inceasing pulse duration and frequency. 4. In 53 μM 4-AP unblocking of K channels during long (0.8 s) depolarizing pulses proceeded with a time constant, τ r , of ca. 0.2 s. Restoration of block at the resting potential proceeded with a much larger time constant, τ′ r , of ca. 1 min. 5. The stationary fraction,r ∞, of K channels conducting in 53 μM 4-AP was 0.66, 0.41, and 0.24 at V=120, 50, and 0 mV, respectively. 6. In a series of experiments with [4-AP] varying between 13.3 and 848 μM, τ r decreased from 0.25 to 0.10 s (V=130 mV, ca. 17°C) whiler ∞ followed the empirical relation 1/r ∞=1+c t +c v exp(−0.77 EF/RT) with E=V-70 mV.c t andc v are dimensionless quantities that increase with [4-AP] and reflect the voltage-independent and voltage-dependent component, respectively, of block. 7. Block of K channels and partial removal are also observed with inwardI K at raised [K+]0. Removal proceeds on depolarization even ifI K is additionally but temporarily suppressed by tetraethylammonium. Hence neither direction nor amplitude ofI K but only the pulse potential seems to determine the extent of block for a given [4-AP].

The mean half times of onset and offset of procaine block were 3.7 and 28 s, respectively. In the presence of 1.4 nM saxitoxin (STX) the corresponding times were virtually the same, 3.1 and 27 s. 3. Similarly, the time course of partial relief from procaine block that is obtained by increasing the frequency of the prepulse-test pulse pairs from 1–10 Hz was unaffected in the presence of STX. 4. Comparison of the equilibrium effects of procaine concentrations ranging from 0.03–1.0 mM suggest a one-to-one drug-receptor reaction. The fraction of Na channels blocked at equilibrium with 1.0 mM procaine, 1.4 nM STX, and 1.0 mM procaine + 1.4 nM STX was 0.81, 0.49, and 0.90, respectively. This result and the kinetic behaviour fully agree with the idea of two separate and independent receptors for procaine and STX.

    Notizen: Zusammenfassung Es besteht großes Interesse daran, zu klären, in welchen Fällen von Bluthochdruck die vasopressorische Substanz Angiotensin II (Hypertensin II) in vermehrter Menge im Organismus vorliegt. Dieser Wirkstoff kann heute synthetisch hergestellt werden; es sind aber bisher noch keine Laboratoriumsmethoden zu dessen quantitativem Nachweis aus Patientenblut bekanntgeworden. Daher wurde zunächst eine Methode entwickelt, die es ermöglicht, Angiotensin aus Blut mit guter Ausbeute zurückzugewinnen. Der beschriebene Weg umfaßt vier verschiedene Arbeitsgänge: Extraktion und Ultrafiltration; Säulenchromatographie; Elution und Präcipitation. Die gewonnene Substanz wird biologisch ausgewertet.
