    Notes: Abstract Aims/hypothesis. The vascularisation of newly transplanted islets originates from the recipients. Because islets transplanted into a diabetic do less well than those transplanted into a euglycaemic environment, we examined the hypothesis that gene expression of angiogenic factors in grafts is delayed in diabetes. These factors include hepatocyte growth factor (HGF) and its receptor c-MET, and urokinase plasminogen activator (uPA) and its receptor uPAR, basic fibroblast growth factor (bFGF), TGF-α and TGFβ-1.¶Methods. Isolated rat islets were studied in vitro under normoxic and hypoxic culture conditions and gene expression was determined with semi-quantitative multiplex RT-PCR. We found that HGF but not c-MET expression was induced by hypoxia in vitro. Using syngeneic Lewis rats, gene expression was also studied in grafts on days 1, 3, 5, 7 and 14 after transplantation.¶Results. In grafts of normoglycaemic rats, HGF expression was enhanced on day 3 and maintained whereas expression of c-MET fell and remained down until day 14. Expression of uPA was up at day 3 and remained high; expression of uPAR was also up at day 3 but then fell to control levels at day 14. Expression of bFGF, TGF-α and TGFβ-1 persisted throughout. Vimentin, a marker of fibroblasts, had increased expression at day 1 which was further enhanced in subsequent days. In the grafts of diabetic recipients the expression of HGF, uPA and uPAR were delayed, being clearly expressed at day 5 rather than day 3. Vimentin expression was similarly delayed.¶Conclusion/interpretation. This apparent delay in angiogenesis provides a potential mechanism for the less favourable outcomes of islets transplanted into diabetic recipients. [Diabetologia (2000) 43: 763–772]

    Notes: Abstract Aims/hypothesis. To examine the functional, metabolic and structural abnormalities in peripheral nerve in the spontaneously Type II (non-insulin-dependent) diabetic BBZDR/Wor rat and compare these data with those in the Type I (insulin-dependent) diabetic BB/Wor rat.¶Methods. Animals were examined at 6 and 14 months of diabetes. Nerve conduction velocity was measured longitudinally. Nerve polyols were analysed using gas liquid chromatography and Na/K+-ATPase activity was measured enzymatically. Light and electron microscopic techniques were used for nerve morphometry.¶Results. Diabetic BBZDR/Wor rats showed a slowly progressive nerve conduction defect that reached 17 % (p 〈 0.01) at 14 months. There was a decrease in Na+/K+-ATPase of 35 % (p 〈 0.05). Structurally, there were mild myelinated fibre atrophy (p 〈 0.05), mild or absent changes of the node of Ranvier, but significant (p 〈 0.001) segmental demyelination and Wallerian degeneration. These findings point to a more severe nerve conduction defect, severe myelinated fibre atrophy and profound nodal changes in Type I spontaneously diabetic BB/Wor rats maintained at the same hyperglycaemic concentrations.¶Conclusion/interpretation. We conclude that other factors, beside hyperglycaemia, are involved in the pathogenesis of the more severe Type I diabetic neuropathy which possibly involve insulin and C-peptide deficiencies.     Notes: Abstract. Creating a computer model from an existing part is a common problem in reverse engineering. The part might be scanned with a device like the laser range scanner, or points might be measured on its surface with a mechanical probe. Sometimes, not only the spatial location of points, but also some associated physical property can be measured. The problem of automatically reconstructing from this data a topologically consistent and geometrically accurate model of the object and of the sampled scalar field is the subject of this paper. The proposed algorithm can deal with connected, orientable manifolds of unrestricted topological type, given a sufficiently dense and uniform sampling of the object's surface. It is capable of automatically reconstructing both the model and a scalar field over its surface. It uses Delaunay triangulations, Voronoi diagrams, and α-shapes for efficiency of computation and theoretical soundness. It generates a representation of the surface and the field based on Bernstein—Bézier polynomials, with the surface modeled by implicit patches (A-patches), that are guaranteed to be smooth and single-sheeted.

    Notes: Abstract Synorogenic veins from the Proterozoic Eastern Mount Isa Fold Belt contain three different types of fluid inclusions: CO2-rich, aqueous two-phase and rare multiphase. Inclusions of CO2 without a visible H2O phase are particularly common. The close association of CO2-rich inclusions with aqueous two-phase, and possibly multiphase inclusions suggests that phase separation of low- to -moderate salinity CO2-rich hydrothermal fluids led to the selective entrapment of the CO2. Microthermometric results indicate that CO2-rich inclusions homogenize between –15.5 and +29.9 °C which corresponds to densities of 0.99 to 0.60 g.cm−3. The homogenization temperatures of the associated aqueous two-phase inclusions are 127–397 °C, with salinities of 0.5 to 18.1 wt.% NaCl equivalent. The rarely observed multiphase inclusions homogenize between 250 and 350 °C, and have salinities ranging from 34.6 to 41.5 wt.% NaCl equivalent. Evidence used to support the presence of fluid immiscibility in this study is mainly derived from observations of coexisting H2O-rich and CO2-rich inclusions in groups and along the same trail. In addition, these two presumably unmixed fluids are also found on adjacent fractures where monophase CO2-rich inclusions are closely related to H2O-rich inclusions. Similar CO2-rich inclusions are widespread in mineral deposits in this region, which are simply metal-enriched synorogenic veins. Therefore, we argue that fluid immiscibility caused volatile species such as CO2 and H2S to be lost from liquid, thus triggering ore deposition by increasing the fluid pH and decreasing the availability of complexing ligands.

    Notes: Abstract The Zn-Pb-Ag deposit contained in the metasediments of middle Proterozoic age at Dugald River forms one of a number of significant zones of sulphide mineralization within the Eastern Fold Belt of the Mount Isa Inlier. It is characterized by its high Zn + Pb grade, high Zn/(Zn+Pb) ratio and strong structural controls with the present resource standing at 38 million tons averaging 13% Zn, 2.1% Pb and 42 g/t Ag. Microstructural timing relationships and a variety of microscopic ore textures indicate that the deposit resulted from replacement and partial infill of carbonaceous and pyritic host rocks by hydrothermal ore-forming fluids during the D4 deformation event. This genesis is contrary to earlier syngenetic-based interpretations, but accords with the discordant nature and structural controls on emplacement of the mineralization. Key timing criteria include (1) truncation of S2 and/or S3 by the late ore minerals, (2) replacement textures in undeformed mineral paragenesis, (3) slightly preferred dimensional orientation and undulating extinction of quartz and muscovite that are intergrown but which crystallized earlier than sulphide minerals in veins. The presence of these microstructural relationships throughout the deposit and the complete absence of any syngenetic stratiform precursor to zinc-lead-silver mineralization indicates that the zinc, lead and silver at Dugald River were epigenetically introduced rather than just having undergone syngenetic deposition during sedimentation and remobilization during deformation. The regional distribution of the mineralization in a specific stratigraphic zone is most likely due to the partitioning of deformation between different rock types. This caused the weaker lithology to accommodate significantly higher strain than adjacent more competent units, resulting in fracturing and localization of the syntectonic mineralization.

    Notes: Abstract. Dihydropyrimidinase from Pseudomonas stutzeri ATCC 17588 was purified 100-fold and characterized. It was found that dihydrouracil, dihydrothymine and hydantoin could serve as substrates for the partially purified enzyme. The K m values for dihydrouracil, dihydrothymine and hydantoin were determined to be 19.6 µM, 21.3 µM and 36.4 µM, respectively, while their respective V max values were 0.836 µmol/min, 0.666 µmol/min and 2.21 µmol/min. Between pH 7.5 and 9.0, enzyme activity was shown to be maximal. The optimum temperature for enzyme activity was 45 °C. Using gel filtration, the molecular weight of the enzyme was calculated to be approximately 115 000 Da. Metal ions were found to influence the level of enzyme activity. Dihydropyrimidinase activity was stimulated by magnesium ions and inhibited by either zinc or copper ions.
