    Notes: Background Antikeratin (AK) autoantibodies, circulating antibodies against epidermal keratins, have been detected in all normal human sera. However, direct evidence on the biological significance of AK autoantibodies is still lacking. Objectives To purify AK autoantibodies from human serum and to make a preliminary study of their biological effects on human keratinocytes. Methods We first extracted keratin polypeptides from human stratum corneum and analysed their purity using sodium dodecyl sulphate–polyacrylamide gel electrophoresis (SDS–PAGE). Next, a keratin affinity column was prepared with the extracted keratins, and AK autoantibodies were purified from pooled normal human serum. Antibodies obtained were identified with SDS–PAGE, enzyme-linked immunosorbent assay, immunoperoxidase staining, immunoelectron microscopy and Western blotting. The biological effect of AK autoantibodies on cultured human keratinocytes was studied using a DNA synthesis assay, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) colorimetric determination and cell cycle analysis. Results On average, 1·83 ± 0·24 mg of antibodies could be purified from 10 mL of pooled human serum. High-titre IgG (about 1 : 70) and low-titre IgM (about 1 : 30) AK autoantibodies were obtained. The DNA synthesis assay and MTT colorimetric determination demonstrated that AK autoantibodies have a significant dose-dependent inhibitory effect on cultured keratinocytes. Correlation coefficients in the two experiments were − 0·583 and − 0·797, respectively. Cell cycle analysis indicated that a small dose of AK autoantibodies leads to inhibition of proliferation of cultured keratinocytes, whereas a large dose of AK autoantibodies causes a visible hypodiploid peak, suggesting apoptosis of keratinocytes. Conclusions The present research lays a solid foundation for further investigation into the biological significance of natural AK autoantibodies.

    Notes: From June to July 1988, larvae of Mystus macropterus (Bleeker) were obtained by artificial propagation of spawners collected from the Jialing River, China. Larvae grown in water temperatures ranging from 26 to 29°C were fully developed at approximately 20 days. The newly-hatched larvae measured 6.0–7.5 mm t.l., exceeding the dimensions of any known newly-hatched larvae of freshwater catfishes indigenous to China. Five days after hatching, when the larvae were 11.4 mm t.l. and the prolarval stage was complete, exogenous feeding commenced. Twelve days after hatching, at 18.4 mm t.l., the yolk sac disappeared, organogenesis was almost complete and the juvenile period began. At 20 days post-hatching, at 23.0 mm t.l., the lateral line system had formed and the juveniles resembled the adults with respect to all external features, which signalled the end of thejuvenile period. Comparison of the larval development in eight species ofeconomic freshwater catfishes shows that the most significant aspect of the larval development of M. macropterus is the sustained (6–7 days) mixed nourishment period and the early differentiation of organs, which enhances the survival rate of the larvae.

    Notes: Abstract This paper deals with the limit analysis of rigid perefectly plastic body under the combined action of initial constant loadings and proportional loadings. Using the von Mises yielding condition and the finite element technique, we present an efficient algorithm based on the mathematical programming formula of the classical kinematic theorem of plasticity theory. This algorithm includes an iterative procedure which produces a monotonically decrescent sequence converging to an upper bound of the real limit load. The results of some numerical examples are presented and show the stable convergency of the new algorithm.

    Notes: Summary Explicit finite-difference procedures are proposed to simulate the elastodynamic response of layered media. To avoid large numerical dispersion and dissipation, different grid sizes are used for different material layers so that the CFL number in each layer can be set to one in the limit. The master-slave technique is applied to deal with the mismatch of grids along the material interface. The interface can be either bonded or unbonded. The dynamic contact between two layers along an unbonded interface is also considered. Numerical examples are presented.
