    Notizen: Cysticercosis is infestation of human tissues by Cysticercus cellulosae, the encysted larva of Taenia solium. An endemic disease in tropical countries, especially in Latin America, an increase is expected in its frequency in Europe because of increasing shifts in population. We report the case of a Spanish woman who had lived for some time in Bolivia and Peru who presented at our Department with a tumoral growth on the mucosa of her lower lip. Clinically, this was interpreted as a mucocele (salivary mucoid ‘cyst’); but histological study of the excised piece revealed, surprisingly, that it was a single–element cysticercosis.

    Notizen: : Electron beam irradiated sliced cantaloupe was tested for 21 d of storage for total aerobic microbial counts, texture, color, and different sensorial parameters as a function of irradiation doses 0, 0.7, and 1.4 kGy and the wash treatments, 1 and 200 ppm chlorine applied to the melons before cutting. Irradiation resulted in a reduction in the total aerobic microbial counts with increasing doses. Melons washed only with water before cutting had total aerobic bacterial counts of 4.0, 2.0, and 0.8 log colony-forming units (CFU)/g on day 0 at irradiation doses of 0, 0.7, and 1.4 kGy, respectively. Across all doses of irradiation, counts were consistently lower for cantaloupe pieces obtained from melons that had been subjected to chlorine rinse in comparison with those washed with water without chlorine. Melons washed with chlorine before cutting had total aerobic bacterial counts of 2.7, 0.7, and 0.5 log CFU/g on day 0 at irradiation doses of 0, 0.7, and 1.4 kGy, respectively. Objective color analysis indicated no significant effect of irradiation on the color of cantaloupe. Texture measured as compression force was lower only for cantaloupe irradiated at 1.4 kGy. Irradiation did not affect descriptive attribute flavor and texture sensory attributes of cantaloupe pieces. Decontamination of whole cantaloupes before cutting using chlorine wash may be combined with low-dose irradiation for shelf-life extension of sliced cantaloupe.

    Notizen: Multinucleate cell angiohistiocytoma (MCAH) was first described by Smith and Wilson-Jones in 1985. It is an uncommon entity but probably underdiagnosed because of lack of recognition by clinicians and pathologists. We report a 47-year-old man with asymptomatic grouped violaceous papules on the dorsum of the hands for 3 years. The histopathological and immunopathological features of our case revealed characteristics of MCAH similar to the initial description of Smith and Wilson-Jones and other reports.

    Notizen: Idiotypes are molecular clues used to explore the specificity and diversity of immune response. In the present study, anti-idiotype antibodies were used to neutralize the pathogenic effects induced by the injection of pemphigus immunoglobulin(Ig)G into BALB/c mice. To achieve our goal, antidesmoglein 1 IgG was obtained from a patient with pemphigus foliaceus with high titer of antiepithelial antibodies. The IgG was isolated by ion exchange chromatography, then digested by pepsin. F(ab′)2 fragments were purified in Sephacryl S-300 and injected in rabbits to produce anti-idiotype antibodies. The rabbit sera reacted with the pemphigus F(ab′)2 fragments. Eleven pemphigus foliaceus sera were recognized by the anti-idiotype serum at the light or heavy chains whereas bullous pemphigoid and normal IgG were negative. Neonatal BALB/c mice injected with pemphigus IgG developed intraepidermal blisters, mimicking the clinical and immunopathological features of the pemphigus. In contrast, the animals treated with anti-idiotype antibodies and pemphigus IgG did not develop blisters. Thus, anti-idiotype antibodies neutralize in vivo the pathogenic effects of pemphigus IgG.

    Notizen: The structures of the choline-dependent pneumococcal murein hydrolases, LYTA amtdase and CPL1 Iysozyme, and the choline-independent CPL7 Iysozyme were analysed by controlled proteolytic digestions. The trypsin cleavage of the CPL1 and CPL7 lysozymes produced two resistant polypeptides, F1 and F7 respectively, corresponding to the N-terminal domain of the enzymes, whereas the amidase LYTA was completely hydrolysed by the protease. Interestingly, the F1 and F7 fragments showed a low, but significant, choline-independent lysozyme activity. Choline reduced the rate of proteolytic hydrolysis of choline-dependent enzymes, suggesting that the C-terminal choline-binding domain adopts a more resistant conformation in the presence of the ligand. On the other hand, the regions encoding the N-terminal domains of the three enzymes have been cloned and expressed in Escherichia coli, showing that these domains adopt an active conformation even in the absence of their C-terminal domains. The lower activity shown by the catalytic domains when compared with that of the complete enzymes suggests that the acquisition of a substrate-binding domain represents a noticeable evolutionary advantage for enzymes that interact with polymeric substrates, allowing them to achieve a higher catalytic efficiency. These results strongly reinforce the hypothesis that the pneumococcal murein hydrolases have been originated by fusion of two structural and functional independent domains, and provide new experimental support to the theory of modular evolution of proteins.

    Notizen: In order to enhance growth, survival and quality during early juvenile stages of marine fish it is important to avoid lipid oxidation problems that are known to cause pathologies and disease. The aim of the present study was to characterize and compare the antioxidant systems in juvenile marine fish of commercial importance in European aquaculture, namely turbot (Scophthalmus maximus), halibut (Hippoglossus hippoglossus) and gilthead sea bream (Sparus aurata). The experiment investigated the interaction of the dietary antioxidant micronutrient, vitamin E, with antioxidant defence systems. Fish were fed with diets of identical unsaturation index supplemented with graded amounts of vitamin E. The relationships between dietary and subsequent tissue vitamin E levels were determined as well as the effects of vitamin E supplementation on lipid and fatty acid compositions of both liver and whole fish, on the activities of the liver antioxidant defence enzymes, and on the levels of liver and whole body lipid peroxidation products, malondialdehyde (thiobarbituric acid reactive substances, TBARS) and isoprostanes. Growth and survival was only significantly affected in sea bream where feeding the diet with the lowest vitamin E resulted in decreased survival and growth. A gradation was observed in tissue vitamin E and polyunsaturated fatty acid (PUFA)/vitamin E levels in response to dietary vitamin E levels in all species. The activities of the main radical scavenging enzymes in the liver, catalase, superoxide dismutase and glutathione peroxidase generally reflected dietary and tissue vitamin E levels being highest in fish fed with the lowest level of vitamin E. The indicators of lipid peroxidation gave consistent results in all three species, generally being highest in fish fed with the unsupplemented diet and generally lowest in fish fed with the diet with highest vitamin E. In this respect, isoprostane levels generally paralleled TBARS levels supporting their value as indicators of oxidative stress in fish. Overall the relationships observed were logical in that decreased dietary vitamin E led to decreased levels of tissue vitamin E, and generally higher activities of the liver antioxidant enzymes and higher levels of lipid peroxides.

    Notizen: Scleroderma is an autoimmune disease characterized by early inflammatory infiltrates followed by fibrosis in the skin and internal organs. CREST is a relatively benign cutaneous variant of scleroderma that features calcinosis, Raynaud's phenomenon, oesophageal dysfunction, sclerodactyly and telangiectases. Glomerulonephritis is a rare association of CREST. We are reporting a patient with CREST who developed glomerulonephritis and had anticentromere and antineutrophil cytoplasmic autoantibodies (ANCA) in her serum.
