    Notes: Objective To determine the relative value of the woman's age, basal follicle stimulating hormone (FSH), basal luteinising hormone (LH) and menstrual cycle pattern (all denned prior to treatment) in predicting the ovarian response to gonadotrophin stimulation for in vitro fertilisation.Design Open, descriptive cohort study.Patients One hundred and seventy-one consecutive ovulatory women undergoing their first cycle of in vitro fertilisation with ovarian stimulation after pituitary desensitisation.Interventions Measurement of basal (early follicular phase) and down-regulated concentrations of serum FSH and LH, and calculation of age at treatment and of median length of menstrual cycles and range of variation prior to treatment.Main outcome measures Duration and amount of gonadotrophin stimulation to achieve follicular maturity, number of mature follicles, peak serum oestradiol concentration and number of mature oocytes, were compared between banded values of variables studied.Results Increasing basal FSH concentrations were significantly associated with reducing oestradiol levels, numbers of mature follicles and oocyte yield even when the FSH levels were still within the normal range though above average (6 to 8.9 i.u./l). There were similar but weaker associations with FSH levels after down-regulation. There were also significant differences between women over and under 40 years old in their oestradiol levels, numbers of follicles and of oocytes. Analysis of variance showed significant independent association of basal FSH with both oestradiol and numbers of oocytes, but not of age, menstrual pattern, or serum LH concentrations.Conclusion Serum FSH is more sensitive than the woman's age in determining her ovarian responsiveness to stimulation. LH concentrations and menstrual cycle patterns are unhelpful.

    Notes: 2-K photoluminescence was used to determine the amount of nonuniformity present in undoped semi-insulating GaAs bulk substrate materials grown by the liquid-encapsulated Czochralski method. The relative photoluminescence intensities of the intracenter (5T2−5E) transition of the Fe2+ state, the near-band-edge transitions, and the Zn peaks, measured across the wafer diameter, show almost the same "W'' pattern. The deep-center bands at 0.63 and 0.77 eV were also measured. Possible mechanisms of the photoluminescence intensity variation are discussed. In the materials used, a predominant shallow acceptor was observed to be Zn.

    Notes: A photoluminescence study has been made of electrically reversible, bulk, liquid-encapsulated Czochralski GaAs at temperatures 2–300 K. The reversibility from the semiconducting to the semi-insulating state is made by slow or fast cooling, respectively, following a 5-h, 950 °C heat treatment in an evacuated quartz ampoule. A donor level at Ec−0.13 eV and two acceptor levels at Ev+0.069 eV and Ev+0.174 eV are produced after the heat treatment. Only the acceptor levels were detected by photoluminescence. A tentative model assigning the acceptor to the intrinsic defect pair VGa-GaAs is discussed.

    Notes: We have developed an easy to use and inexpensive low-energy electron diffractometer. The system is based on an Apple Macintosh II microcomputer and uses a high-resolution CCD video camera. The video interface is a single plug-in imaging board which digitizes the video signal in real time. No expensive auxiliary video processing devices are required. The system is of moderately high speed. A typical set of 16 IV curves with 125 data points in each curve can be generated from images summed over 16 frames in less than 4 min. Spot profile measurements are also routinely made. A description of the apparatus and the capabilities of the system are presented, illustrated using measurements of the epitaxial growth of bismuth films on III-V semiconductor surfaces.

    Notes: The recirculation of lymphocytes through the tissues and their return to the blood were compared in nude and euthymic rats. Three approaches were used: the organ distribution of 15Cr-labelled lymphocytes from nude or euthymic donors at 24 h after injection: the compartmental distribution of B and T lymphocytes as assessed by autoradiography of the spleen, lymph nodes, and Peyer's patches; and the tempo of recirculation from blood to thoracic duct lymph as estimated by counting timed fractions of lymph from a recipient of labelled lymphocytes. The following conclusions were drawn: (1) The distribution of lymphocytes between organs and wilhin organs is very similar in nude and euthymic recipients. In particular, B lymphocytes proceed normally to the follicular areas in the absence of T cells. (2) The recircufation from blood to lymph is delayed in nude rats. (3) For equal numbers of B and T cells injected intravenously about half as many B cells as T cells enter mesenteric and cervical lymph nodes, but approximately equal numbers of B and T cells enter the spleen and Peyer's patches.

    Notes: The lymph nodes (LN) of the congenitally athymic nude rat (rnu) were compared with the LN of non-nude littermates. The mesenteric and coeliac LN were smaller in the rnu rats, but the axillary and the cervical LN were larger. Germinal centres were found in the LN of nude rats. They were scarce and often very small, but some were of normal structure, especially in the cervical LN. The endothelium of post-capillary venules in the LN paracortex was usually flat, but occasionally plump endothelium, typical of post-capillary venules in the LN of normal rats, was seen. The paracortex was extremely depleted of lymphocytes but remained a distinct area occupied predominantly by pale interdigitating cells. In some LN the medullary sinuses were distended and the cords engorged with plasma cells. The predominant cells in thoracic duct lymph were immunoglobulin-positive B lymphocytes. The hourly output of these cells was the same as that in non-nude littermates. However, small numbers of thoracic duct cells were positive for the markers identified by the monoclonal antibodies W3/13 and W3/25, although in normal rats these are not expressed on small B lymphocytes.

    Notes: We have examined the cellular changes taking place in rat popliteal lymph nodes undergoing a graft-versus-host (GvH) reaction. Lamination of immunoperoxidase-stain lymph node sections, using a panel of mouse monoclonal antibodies directed against different rat lymphoid cell subsets, revealed a disorganization of the lymph node architecture with disappearance of the follicles, and an intermingling of T and B cells, so that no distinct T - and B-cell areas were visible any more. Since the GvH nodes showed a preferential accumulation of host B cells over host T cells (particularly over the W 3/25’T helper cell subset), we also investigated the requirements for host B cell activation. The popliteal lymph node GvH reaction was induced in (PVG x DA)F1 rats by the injection of PVG cells into one foot and by DA cells into the other foot, and then immunoglobulin kappa allotype marked PVG B cells from athymic donors were injected intravenously. The allotype marked B cells proliferated vigorously in response to the DAT cells, but much less in response to the PVG T cells. These results indicate that the massive B-cell activation taking place in GvH reactions may require an alloantigen incompatibility between donor T cells and host B cells, and argue against non-specific mitogenic induction of the B cells.

    Notes: The primary migration of lymphocytes from the blood was compared in nude rats and in euthymic rats. The flatter endothelium in the post-capillary venules (PCV) in the lymph nodes of nude rats was as efficient as the high endotheluim of PCV in euthymic rats at capturing both T and B lymphocytes from the blood, although lymphocytes took a longer time to cross the PCV wall in nude recipients. The organ distribution of both lymphocytes and lymphoblasts ([125I]UdR-labelled cells) was broadly similar in nude and euthymic recipients. A second aim was to compare B and T lymphocytes with respect to the rate and sites at which they leave the blood after intravenous injection. As judged by sampling venous blood, B lymphocytes left the blood faster, but this was partly attributable to a larger intravascular pool of B lymphocytes in small blood vessels, especially in the lung. Thoracic duct lymphocytes from nude rats collected under standard conditions (16 h, 0°C) entered the cervical lymph nodes very poorly, but when lymphocyte transfer was performed under more physiological conditions entry of B lymphocytes into lymph nodes was about half that of T lymphocytes. B lymphocytes did show a slight preference for entry into Peyer's patches compared with lymph nodes.

    Notes: Two experiments are described concerned with the mechanism of host cell activation in the rat popliteal lymph node (LN) undergoing a graft-versus-host (GVH) reaction. (1) Irradiated, F1 hybrid hosts (750 rad) mounted an impaired response to parental strain T cells. This was augmented by an intravenous injection of F1 hybrid lymphocytes but not by parental strain B lymphocytes syngeneic with the initiating T cells. When the donor T cells (footpad) und B lymphocytes (intravenous) were completely allogeneic the residual response of the irradiated F1 was completely inhibited. (2) The popliteal LN response in the semi-allogeneic situation of the type (A × C)F1→(B × C)F1 was, if anything, weaker than in the allogeneic situation AA→BB. These results and other data are discussed in terms of a possible major histocompatibility complex (MHC) requirement for host cell activation. The sharing of an MHC haplotype between donor and host cells is unlikely to be a necessary or sufficient condition for host cell activation.
