    Notizen: Platinum-silicon alloys have been prepared over a wide composition range by reactive magnetron sputtering of platinum in a SiH4 plasma. Studies on the stoichiometry of the films show that it depends sensitively on the silane partial pressure and the cathode potential. An analysis of the intensities of the emission lines of platinum and hydrogen present in the plasma indicates that at lower silicon concentrations the stoichiometry of the film bears a one-to-one correspondence with the stoichiometry of the plasma. X-ray diffraction studies on the films deposited at room temperature show metastable products such as extended solid solutions on either side of the equilibrium phase field and the formation of a bcc phase (a=4.169 A(ring)) for the Pt67Si33 alloy. Stoichiometric intermetallic phases Pt2Si, PtSi, and PtSi2 have been synthesized by depositing the films at 100, 300, and 500 °C, respectively.

    Notizen: Summary We investigated basal and ACTH stimulated levels of cortisol, corticosterone, 17α-hydroxyprogesterone, 11-deoxycortisol and 11-deoxycorticosterone as well as plasma levels of ACTH before and during the oral administration of ketoconazole in five patients with Cushing's syndrome (3 with bilateral adrenal hyperplasia, 1 with adrenal adenoma and 1 with adrenal carcinoma) and in three controls. The influence of ketoconazole on the transformation of3H-17α-hydroxyprogesterone to3H-11-deoxycortisol and3H-cortisol and of3H-11-deoxycortisol to3H-cortisol as well as of3H-11-deoxycorticosterone to3H-corticosterone was also examined in slices or homogenates of normal and hyperplastic adrenal tissue from four patients. Ketoconazole induced a rise of 11-deoxycortisol and 11-deoxycorticosterone, but not of cortisol and inconsistantly of corticosterone which were increased by ACTH. Thus the ratio 11-deoxycortisol/cortisol rose more after ketoconazole than after ACTH and the ratio 11-deoxycorticosterone/corticosterone rose after ketoconazole but fell after ACTH. Plasma ACTH levels were stimulated 2–50 fold by ketoconazole. Incubation studies of adrenal tissue slices with3H-17α-hydroxyprogesterone showed that ketoconazole inhibited the transformation of3H-17α-hydroxyprogesterone to3H-cortisol but not to3H-11-deoxycortisol so that the ratio3H-11-deoxycortisol/3H-cortisol increased 15–80 fold. After incubation of adrenal slices with3H-11-deoxycortisol or3H-11-deoxycorticosterone and ketoconazole, a 2–260 fold increase of the ratios3H-11-deoxycortisol/3H-cortisol and3H-11-deoxycorticosterone/3H-corticosterone were also found. In conclusion, the in vivo data indicate and the in vitro data confirm that ketoconazole inhibits cortisol and corticosterone secretion by blocking adrenal 11β-hydroxylase activity in normal subjects as well as in patients with Cushing's syndrome, an effect which is compensated in vivo by high ACTH levels.

    Notizen: Summary Cytomegalic inclusion disease (CID) is caused by a horizontally or vertically transmitted human herpes virus infection and may persist for life without obvious clinical symptoms. A serious course of horizontal primary and recurrent infections, however, is often observed in immunocompromised persons such as recipients of organ transplants and patients receiving fresh blood transfusions. Vertical infection may cause fetopathies. The human cytomegalovirus (HCMV) is thought to inherit an oncogenic potential as lately discussed for AIDS and M. Kaposi. Laboratory diagnosis of HCMV infection is performed by light microscopy (inclusion bodies), electron microscopy, virus isolation in cell culture, demonstration of viral DNA and antigen in clinical specimens, by histochemical methods (e.g. immunoperoxidase technique) and by DNA and peptide analysis for identification of different isolates and viral finger prints. Evaluation of cell-mediated immunity in HCMV infection is performed quantitatively (assessment of Thelper/Tsuppressor ratios) or qualitatively (specific lymphocyte stimulation by the antigen). In most cases laboratory diagnosis is achieved by serological methods, i.e. demonstration and quantitation of HCMV-specific antibodies. In this context, a number of liquid- and solid-phase immunoassays have been developed, of which immunofluorescence and ELISA are most commonly used, besides complement fixation and passive haemaglutination. These procedures on the one hand allow the use of different antigen preparations as early and late viral proteins, and on the other hand permit a specific determination of different Ig classes and subclasses. A variety of assays has been established especially for determination of virus-specific IgM antibodies, which are predominantly found in active infection. These, however, at least in part may show non-specific results caused by interference of rheumatoid factor or IgG competition. Such problems have now been dealt with and are avoided by IgG precipitation or IgM immunosorption (“μ-capture” technique). These recent methods allow an exact epidemiological identification of risk groups for CMV infection. Results from our laboratory revealed 13% HCMV-IgM positive patients among pregnant women, 16% IgM positive patients among renal transplant recipients, 4% igM positive cases in patients after cardiosurgery and 1.7% IgM positives among prostitutes. The prevalence of HCMV infection as indicated by specific IgG antibodies was 56%, 90%, 83%, and 90%, respectively. No IgM antibodies were found in haemophiliacs and healthy blood donours, which showed a prevalence of HCMV infection in 69% and 47% of tested serum samples.

    Notizen: Abstract The results of RNA analysis for the detection of rotavirus were compared with those of a standard enzyme-linked immunsorbent assay (ELISA) and electron microscopy using 212 faecal specimens obtained from 200 children with gastroenteritis. Rotavirus was extracted directly from faecal specimens and RNA segments were made visible by polyacrylamide gel electrophoresis using a silver staining technique. Of the 212 faecal specimens 137 were found to be positive in ELISA, 125 in RNA analysis and 121 in both methods. Forty-nine of the 212 specimens were also investigated by electron microscopy. Thirty-five were positive when examined by electron microscopy, 37 were positive in ELISA and 33 in RNA analysis. RNA analysis of 119 faecal samples in outbreaks and sporadic cases of rotavirus infection yielded 42 different rotavirus electrophoretypes. The results indicated that no one method was sufficient to detect all positive specimens and that RNA analysis is useful in epidemiological studies.

    Notizen: Abstract Angular distributions of fission-like fragments have been measured for50Ti+208Pb and56Fe+208Pb collisions. Z-dependent asymmetries around Θincm= 90° preclude their interpretation in terms of compound nucleus fission with the transition state theory. Fits of the data with a simple ansatz for statistical angular momentum fluctuations (tilting) give evidence for an incomplete relaxation of the tilting mode in quasi fission reactions. Recently, there has been much focus on “fusion-fission” angular distributions in heavy systems with unusually large “anisotropies” /1–5/ if compared to expectations based on the transition state theory (TST) and on rotating liquid drop model (RLDM) saddle point shapes. The validity of these expectations for very heavy systems is being questioned /6/ and modifications of the TST /7,8/ are being vividly discussed /9,10/. Deviations from the RLDM expectations are being associated with the onset of quasi-fission reactions /1,3,5,11,12/ occuring whenever complete fusion is suppressed by dynamical entrance channel limitations /13/. We have measured differential cross sections d2 σ/dωdZ as a function of scattering angle Θcm for quasi-fission fragments from50Ti +208Pb collisions at 5.0 and 5.5 MeV/A and56Fe +208Pb collisions at 5.7, 6.1, 6.8, 8.3 MeV/A. The measurements involved an off-line X-ray activation technique /14/ where up to 15 catcher foils representing slices of lab. angles of 5° were measured simultaneously by 15 absolutely calibrated Ge low-energy photon spectrometers. Transformation of the cross sections into the centre of mass system was performed by assuming full momentum transfer and by using the experimental total kinetic energies of Ref./11/. Examples for distributions of d2σ/dΩdZ are given in Fig 1. Qualitatively similar results are obtained at the other bombarding energies and with the56Fe projectile.
