    Notes: Summary The immunosuppressive activity of azathioprine (AZA) is unpredictable and depends on the formation of intracellular thiopurine ribonucleotides. However, the quantification of these active thiopurines presents difficult analytical problems. It has recently been postulated that plasma concentrations of 6-thiouric acid (6-TU) and 6-mercaptopurine (6-MP), metabolites of AZA, may provide more readily measurable indices of the pharmacologic activity of AZA. In order to evaluate the utility of 6-TU and 6-MP plasma concentrations in monitoring AZA therapy, we studied their pharmacokinetics in 6 renal transplant patients, and their in vitro immunosuppressive potency in a mixed lymphocyte proliferation assay. A peak plasma 6-TU concentration of 710.7 ng/ml was observed at 3.8 h after oral dosing. Good correlation was observed between the elimination t1/2 of 6-TU and serum creatinine, and between AUC over 24 h and serum creatinine. However, we did not observe a second peak in plasma 6-TU concentration that could be attributed to the degradation of active AZA metabolites. 6-MP plasma concentrations in the patients were low (mean peak concentration 36.0 ng/ml) and rapidly disappeared within 8 h. In vitro immunosuppressive activity could not be demonstrated for 6-TU over a concentration range of 1.25 ng/ml to 0.25 mg/ml. We conclude that 6-TU is pharmacologically inert and is primarily eliminated by the kidneys. Our findings currently do not support the use of plasma concentrations of 6-TU or 6-MP to monitor AZA therapy. In order to optimize AZA therapy, analytical techniques that are technically feasible and that can directly quantify the active intracellular thiopurines are being explored.

    Notes: Extensive light scattering measurements, including the intensity, turbidity, and linewidth, on a three-component microemulsion system consisting of mixtures of water, decane, and a surfactant sodium di-2 ethylhexylsulfosuccinate (AOT) (WDA), have been made. The critical and several off-critical mixtures have been studied along constant microemulsion droplet volume fraction lines in the one-phase region over a very large temperature range. In the vicinity of the lower phase separation temperature Tp the intensity data are very well accounted for by the standard theory of critical binary fluids using a single value for the short range correlation length ξ0=(13.5±1.5) A(ring). By combining a mode-coupling theory including the background effects and a linear model equation of state applicable in the critical region, we have been able to fit the dynamic light scattering data using a Debye cutoff length q−1D which is equal to the constant average diameter of microemulsion droplets. Furthermore, we find clear evidence for a crossover from critical to single particle behavior in both static and dynamic light scattering data. A crossover temperature Tx has been identified at which qDξ(Tx)=1. Analyses of the dynamic light scattering data show that qD, which can only be measured far away from Tp, in fact plays a decisive role in controlling the critical dynamics in the whole temperature range.

    Notes: Poly(γ-benzyl L-glutamate), partially transesterified with 1-dodecanol, and polysiloxane were employed to fabricate a right- and left-handed optical element, respectively. The wavelength of selective reflection was tuned via physical blending or annealing at a proper temperature. An optical notch filter comprising elements with each handedness showed an optical density of 2.0 within the rejection band and 73 to 83% transmission outside the band, suggesting liquid crystalline polymers as promising materials for the construction of environmentally robust optical notch filters.

    Notes: Adhesion and interfacial failure of Cr, Cu, and Cu/Cr films on a thin pyromellitic dianhydride-oxydianiline polyimide substrate have been investigated using a recently developed stretch deformation technique. This method directly measures the energy required to separate the interface from the difference in the load versus elongation curves between film/substrate and substrate structures. The failure mode was observed in combination with morphological studies by in situ optical microscopy and scanning and transmission electron microscopies. The stress distribution in the sample upon stretching has been computed by a finite element analysis. The difference in the stress relief modes, film fracture, and delamination behavior for Cu versus Cr films is discussed. The important role of crack formation in the metal overlayer for initiating failure is demonstrated. The effects due to the addition of a brittle Cr interface layer between polyimide and Cu on the buildup of stress and the onset of failure have also been investigated.

    Notes: Low carrier concentrations of undoped p-type and Te-doped n-type GaSb epilayers have been grown on (100)-oriented Te-doped GaSb substrates by liquid-phase epitaxy with supersaturation and undersaturation techniques, respectively. The net carrier concentrations of undoped and Te-doped layers obtained by C-V measurement can be as low as NA −ND =8×1015 cm−3 and ND −NA =1016 cm−3 , respectively. These GaSb-grown layers were characterized by photoluminescence (PL) measurement. The native neutral acceptor transitions appear at 778.2 meV for undoped layers and 777.2 meV for Te-doped layers. The ionization energy for the Te donor in GaSb was determined to be 3.6 meV below the bottom of conduction band at 18 K. The relation between the energy gap of GaSb and temperature was obtained by PL measurement at various temperatures. The spin-orbit splitting of the valence band was measured to be 0.798 eV at room temperature.

    Notes: Summary Acute disseminated encephalomyelitis (ADEM) is an uncommon inflammatory disorder of the nervous system. The appearance of ADEM on cranial CT scans has rarely been reported. The author reports seven cases in two instititions during a period of seven years. Only four of the seven patients had hypodense lesions in the white matter and six patients showed spotty, nodular, or gyral enhancement after contrast injections. The enhancement resolved with steroid therapy, leaving some persistent hypoattenuated areas even after 10 months follow-up, these findings support the dual components of the pathogenesis of ADEM. The vasculitic component may be responsive to steroids, nevertheless the demyelinating or necrotic areas may not, and the latter may be responsible for the sequelae of ADEM.
