    Notes: It is found that a reversed-field pinch (RFP) dynamo uniformly generates most of the toroidal flux in a RFP plasma with a high magnetic Reynolds (S) number. Hereafter, this is referred to as the global dynamo. The toroidal flux generated by the global dynamo is proportional to the plasma current. The effective inductance in a RFP plasma increases with S number and remains constant against the changing plasma current. This means that the pinch parameter aitch-theta is held constant, that is, the global dynamo has a self-organizing effect sustaining a particular RFP configuration. By comparing simulation results and analysis of magnetic fluctuations, it is confirmed that a global dynamo is generated by the nonlinear evolution of the single-helicity of the m=1 mode alone.

    Notes: Abstract A nerve-sheath myxoma beneath the dermis in the arm of a 5-year-old boy is reported. The tumor had developed on the site of chicken-pox vaccination the patient had received 2 years previously, and a foreign-body granuloma involving a nerve-fiber bundle was seen adjavent to the tumor. On the basis of previous reports, it is doubtful whether the varicella-zoster virus is tumorigenic. The present case suggests a possible relationship between the development of this tumor and factors associated with the vaccination.

    Notes: Abstract We have isolated cDNA clones encoding cysteine synthase (CSase, EC 4.2.99.8), which catalyzes the terminal step in cysteine biosynthesis, by direct genetic complementation of a Cys− mutation in Escherichia coli with an expression library of Citrullus vulgaris (watermelon) cDNA. The library was constructed from 8-day-old etiolated seedlings of C. vulgaris in the λZAPII vector, converted to a plasmid library by in vivo excision, and then used for transformation of cysteine auxotroph E. coli NK3, which lacks the cysK and cysM loci. The complementing cDNA containing a 560 by 5′-untranslated region encodes a polypeptide of 325 amino acids of Mr 34342. The translational product reacted with an antibody raised against CSase A of Spinacia oleracea. CSase and β-pyrazolealanine synthase activities were demonstrated in vitro in extracts from E. coli cells expressing the cDNA. Genomic DNA blot analysis indicated the presence of a single copy of the gene, designated cysA, in the C. vulgaris genome. RNA blot hybridization indicated constitutive expression of cysA in cotyledons, hypocotyls and radicles of green and etiolated seedlings. These data suggested that this cDNA clone encodes CSase A the homolog of which in spinach is localized in the cytoplasm. The molecular phylogenetic tree of the amino acid sequences of CSaes from plants and bacteria suggested that there are three families in the CSase superfamily; the plant CSase A family, the plant CSase B family and the bacterial CSase family. The proteins in the plant CSase A family are the most conserved relative to the ancestral CSase protein.

    Notes: Abstract Malignant fibrous histiocytoma of the upper respiratory tract is rare. We report a case of a glottic malignant fibrous histiocytoma in a 46-year-old man. Ultrastructural findings demonstrated that tumor cells were composed of fibroblast-like cells with abundant rough endoplasmic reticulum, histiocyte-like cells with numerous lysosomes and transitional cells with characteristics of both the fibroblast-like and histiocyte-like cells. Laryngeal microsurgery was performed as surgery and the patient has done well without recurrence to date (8 months after surgery).

    Notes: In a number of infectious and inflammatory diseases, stimulation of the immune system can lead to increased accumulation of tryptophan metabolites via induction of kynurenine pathway enzymes in extrahepatic tissues. We developed a liquid chromatographic/mass spectrometric (LC/MS) method suitable for tracing the disposition of 13C isotopomers of L-tryptophan and L-kynurenine in various cultured cell, tissue slice, and whole animal model systems used to investigate tryptophan flux through the kynurenine pathway. The method employs extractive derivatization of the analytes and their 2H internal standards with pentafluorobenzyl bromide in order to enhance the negative ion chemical ionization (NICI) mass spectrometric response. Normal-phase liquid chromatographic separation of derivatized analytes was optimized using a silica column with organic solvents, followed by particle beam transfer and NICI-MS. Standard curves were linear over the range 1-250 ng per sample. Particle beam and mass spectrometric operating parameters were optimized with direct flow injections of 1-(methylamino) anthra-quinone, which is an ideal test compound for the evaluation of LC/NICI-MS. The developed method was used to quantify the conversion of (13C6)L-tryptophan to (13C6)L-kynurenine by human monocytes (THP-1) stimulated with interferon-γ, lung and brain tissue slices obtained from gerbils immune-stimulated with pokeweed mitogen. The effect of whole body immune stimulation on the plasma levels of engogenous L-kynurenine in mice stimulated with interferon-γ was also quantified.

    Notes: The secondary ion yields of six element in silicon were measured as a function of the primary oxygen ion energy in a Cameca IMS-4f SIMS instrument. The ion yields of elements with a high ionization potential decrease steeply with primary ion energy owing to the decreasing surface oxygen concentration. The decrease of the ion yield depends on the ionization potential. This relation and the known relative sensitivity factors enable a semiquantitative analysis of impurities in silicon to be carried out without standard samples.A similar study combined with oxygen flooding shows that the yields are now very high and independent of the primary ion energy. Profile distortions due to segregation effects, however, can occur.A comparison of the ion yields obtained with and without energy offset under low and high oxygen surface coverage shows that the shape of the arsenic secondary ion energy distribution is drastically changed leading to pronounced differences in ion yields.
