    Notes: Following direct exposure to sunlight while pursuing leisure activities, many have noticed a strong sense of fatigue in the evening. In this regard, our results of a survey of awareness showed that the development of fatigue from solar exposure of the body was generally recognized. On the other hand, a tool for objective and quantitative determination of mental fatigue has recently been reported. Known as the Advanced Trail Making Test (ATMT), it is a method of evaluating brain function. In the present study, we attempted to determine fatigue development caused by exposure of the human body to solar radiation using ATMT results. For 3 days in the summer season, 15 male subjects (26–41 years old) received exposure to the sun equivalent to 100 kJ cm−2 of ultraviolet radiation three to four times each day. During the periods of exposure, the subjects wore short-sleeved shirts and short pants, and covered their heads with a towel. Following the 3-day period, they were divided into two groups based on their subjective evaluation of a sense of tiredness, fatigued (n = 10) and non-fatigued (n = 5). In the fatigued group, a significant increase in the subjective score for fatigue sense was observed in the evening of all 3 days following sun exposure, as well as in the morning of the third day, as compared with those in the non-fatigued group. Further, a significant increase in average ATMT value was also observed in the fatigued group in the evening of the first and second days following sun exposure, as well as in the morning of the third day. These results indicate that ATMT may be a useful evaluation tool for quantitative and objective measurement of mental fatigue caused by exposure to sunlight.

    Notes: In this study the photolightening behavior of blond hair was investigated. The results demonstrated that visible (VIS) and ultraviolet (UV) light lighten blond hair through different mechanisms. VIS light was found to contribute much more to the lightening of blond hair than UV light, and acted directly, while UV light only lightened blond hair that had been washed following irradiation. VIS and UV light both, however, lightened to a similar degree isolated melanin granules and decomposed melanin granules that were exposed on a cross section of blond hair. These results indicate that melanin granules are equally sensitive to both forms of light while blond hair is most sensitive to VIS light. The results also indicate that hair tissues, excluding melanin granules, are damaged by UV light but not by VIS light. Based on these facts, the hypothetical lightening mechanism of UV light is assumed to be that UV light preferentially attacks and damages hair tissues rather than melanin granules. This occurs only after the hair is washed, as the washing process removes the melanin granules that effuse from loose hair fibers. In contrast, VIS light preferentially attacks and decomposes the melanin granules rather than other tissues, and also results in the lightening of blond hair but without the need for subsequent washing. We also found that while VIS light destroys the structure of isolated melanin granules, UV light does not act in a similar manner. Consequently, it is proven that VIS and UV light attack different sites of the melanin granule, even though the lightening rates from both light sources are similar.

    Notes: Sun exposure during leisure activity evokes fatigue. We employed the Advanced Trail Making Test (ATMT), a recently developed objective method of evaluating brain function performance used to measure mental fatigue, for objective determination of fatigue development caused by solar exposure to the human body. First, a survey of consumer awareness was performed, and fatigue development from solar exposure was generally recognized in both summer and spring. In the field test, 15 males (26–41 years old) received sun exposure equivalent to 100 kJ m−2 of ultraviolet radiation three to four times each day for 3 days, during which the subjects wore a short sleeve shirt and a short pant, and covered their head with a towel. A significant increase in scores for subjective sense of fatigue was observed in the evening of all 3 days following sun exposure and on the fourth day, which had no exposure, as well as in the morning of the third and fourth days, as compared with those periods during the control week, which did not have experimental solar exposure. ATMT showed a significant increase in average value in the evening of the first and second days following sun exposure, as well as in the morning of the third and fourth days. In addition, increases in body temperature and heart rate were observed during the exposure periods. The results of multiple regression analysis of subjective feelings showed that fatigue caused by solar exposure was qualitatively different from that in the control week. These results suggest that brain function performance declined following solar exposure as did fatigue development. ATMT results may be useful for quantitative and objective evaluation of mental fatigue caused by sun exposure, along with development of sun care products for the prevention of solar-caused fatigue.

    Notes: Two populations from Wonzye and Nkumbula Island, Lake Tanganyika, containing both rock and shell-bed dwelling individuals of the genus Telmatochromis were analysed by means of comparative morphological and DNA haplotype data from the mitochondrial control region. The data on individuals from rock v. shell-bed habitats allowed a clear discrimination to be made between habitats, while there was no distinction among individuals of the same habitat from Wonzye and Nkumbula. The shell-bed dwellers had small body size, including smaller size at sexual maturity and fewer eggs than rock dwellers. The existence of two distinct species that diversified according to habitat choice is suggested.

    Notes: Background Mast cells (MCs) arise from haematopoietic stem cells. We have recently reported that CD34+ progenitors derived from human bone marrow (BM) develop into tryptase+, chymase+ MCs when cultured in the presence of recombinant human stem cell factor (rhSCF) and recombinant human IL-6 (rhIL-6). In an experiment for the expression of chymase during differentiation, chymase+ cells were detected in human BM, but tryptase+ cells were not found.Objective The purpose of this study was to show the appearance of chymase+ cells in CD34+ cells with an origin different from MC differentiation.Methods CD34+ cells from human BM were sorted with anti-CD117 monoclonal antibody (mAb), and cytospins of CD34+, CD34+CD117+, or CD34+CD117− were prepared. These cells were cultured with rhSCF+rhIL-6 for 12 weeks. Some of the cells were subjected to either histological stain with Wright–Giemsa or immunocytochemistry with anti-chymase mAb. Real-time RT-PCR was also performed to compare the transcriptional level of chymase from each cell preparation.Results Chymase was expressed in CD34+ cells as well as human MCs by immunocytochemistry. Substantial CD34+CD117− cells, but not CD34+CD117+ cells, were stained immunocytochemically with anti-chymase mAb. For 1 week culture with rhSCF+rhIL-6, no cells expressed chymase in any preparation. Real-time RT-PCR revealed positivity for chymase mRNA in CD34+ cells, but it reduced at 1 week of culture, and increased as cells developed into MCs. Chymase mRNA in CD34+CD117+ cells was negligible compared with that in CD34+CD117−. Tryptase mRNA was below the detectable level in CD34+ cells, and increased along with MC differentiation. After 12 weeks of culture, CD34+CD117+ developed predominantly into MCs, whereas CD34+CD117− developed into monocytes/macrophages.Conclusion Our findings suggested that chymase is present not only in MCs but also in CD34+CD117− BM progenitors, but that its origin is different from the MC lineage.

    Notes: Background CD34+ progenitor cells develop into tryptase+, CD117+ mast cells when cultured in the presence of recombinant human stem cell factor (rhSCF). However, spontaneous IgE receptor (FcɛRI) expression during human mast cell development is not well examined.Objective Here, the expression and function of FcɛRI in and on human bone marrow-derived mast cells (HBMMCs) during development were investigated.Methods and Results At 4 weeks of culture, predominant cells expressed high-affinity IgE receptor α chain (FcɛRIα) on the cell surface determined by flow cytometry, but CD117 was less expressed. Immunocytochemistry with antitryptase mAb and anti-FcɛRIα mAb revealed intracellular and surface expression of FcɛRIα at 2 weeks of culture, but tryptase was less expressed. FcɛRIα mRNA transcript preceded that of tryptase mRNA at 2 weeks of culture determined by real-time RT-PCR, and FcɛRIα, FcɛRIβ, FcɛRIγ, and tryptase mRNA increased along with differentiation. FcɛRIα cross-link on HBMMC and 4-week-old mast cells/mast cell precursors induced the release of IL-5 and granulocyte macrophage-colony stimulating factor, which was enhanced by rhSCF.Conclusion These data indicated that HBMMC constitutively and spontaneously expressed functional FcɛRI subunits at the early stage of differentiation, probably because of the differences in the ability and functional property of progenitors.

    Notes: Background : Our recent histochemical studies have revealed an increase in myofibroblasts and in leptin and its receptor in endothelial cells, and myofibroblasts in Helicobacter pylori-infected human and Mongolian gerbil fundic mucosa.Aim : The present study was undertaken to clarify the H. pylori–induced interaction between leptin in cultured gastric surface mucous cells and fibroblasts.Methods : GSM06 cells were incubated with an air− liquid interface on a collagen gel layer containing mouse fibroblast cell line L929. Medium containing H. pylori bacilli (ATCC43504) at 10–100 times higher concentration than the GSM06 cells was added from the luminal side and the localization of leptin was observed by immunohistochemistry. The transformation of L929 cells to myofibroblasts was detected by electron microscopy and PR 2D3 immunoreactivity.Results : L929 cells in the control group showed a spindle shape with scarce cytoplasm. In the H. pylori-treated group, L929 cells showed features characteristic of myofibroblasts, and most GSM06 and L929 cells showed leptin immunoreactivity. In contrast, L929 cells incubated with H. pylori alone did not undergo this differentiation.Conclusions : Attachment of H. pylori to surface epithelial cells caused conversion of fibroblasts to myofibroblasts. We suggest that leptin plays a role in this transformation.
