    Notes: Accumulating evidence suggests a link between immediate hypersensitivity and cellular immunity. In this study, we examined the effect of interleukin 2 (IL-2) on basophil histamine release. Histamine-releasing activity of IL-2 was very weak with % histamine release of 2.9 ± 1.3 (mean ± SEM, n= 9) at 1:12 dilution. IL-2 at 1:1200 dilution slightly inhibited anti-IgE-induced histamine release by 22.4 ± 18.6% (P 〉 0.05). There was a significant potentiation of release at 1:12 dilution of IL-2 with % enhancement of 78.7 ± 42.2 (P 〈 0.05). IL-2 enhanced the calcium ionophore A23187-induced histamine release in a dose-dependent fashion. IL-2 at 1:12 dilution significantly potentiated release by 28.8 ± 6.3% (P 〈 0.05). There was a slight suppression of formyl-methionyl-leucyl-phenylalanine (FMLP)-induced histamine release at 1:1200 dilution with % inhibition of 23.4 ± 7 4 (P 〉 0.05). At 1:12 dilution, IL-2 significantly potentiated FMLP-induced release by 73.7 ± 41.6% (P 〈 0.05). Recombinant IL-2 (RIL-2) augmented anti-IgE-induced histamine release with a significant enhancement at 200 units/ml. Conventional IL-2 was more potent than RIL-2 in enhancing release. These results indicate that IL-2 enhances basophil histamine release and some part of the effect of IL-2 on basophils is derived from other factors contained in conventional IL-2.

    Notes: Background  Birt–Hogg–Dubé syndrome (BHD) is a rare autosomal dominant genodermatosis characterized by skin tumours, including multiple fibrofolliculomas, trichodiscomas and acrochordons. BHD patients also may suffer from associated renal and colonic carcinomas. The defective gene in BHD has been recently identified and is suspected of being a tumour suppressor gene. Several mutations of the BHD gene have been reported only in Caucasian patients.Objectives  This study reports the first Asian family that has been demonstrated to carry a BHD mutation.Patients/methods  The proband was a 26-year-old Japanese man with multiple asymptomatic, soft skin-coloured papules on his face, neck and trunk, which were clinically thought to be acrochordon. His father was also affected. Histopathologically, the papules revealed a fibrofolliculoma that had a circumscribed proliferation of fibroblasts and collagen fibres surrounding an abnormal hair follicle.Results  Mutational analysis of the BHD gene of the proband and the father detected 1733insC, a cytosine insertion mutation in an eight-cytosine tract (nucleotides 1733–1740) in exon 11. Analysis of fibrofolliculoma in the proband showed heterozygous 1733insC mutation, suggesting the absence of loss of heterozygosity. Interestingly, previous mutational analysis in Caucasian patients revealed that both1733insC and 1733delC mutations were hot spots.Conclusions  This study is the first to find the same hot-spot 1733insC mutation in Asian kindred. The mutations in this polycytosine tract may have a wide, global distribution despite their arising from a different ethnic background.

    Notes: Nitric oxide (NO) produced in the endothelial cells and other tissues has been known toplay a range of physiological roles including vasodilation. Thus, direct in vivo measurement of NO isof great importance for clarification of physiological roles of NO and development of therapeuticsusing NO. It has long been assumed that NO is readily oxidized in blood and cannot be detected;however, some recent reports demonstrated much longer half-life of NO in the blood stream in vivo.Recently, we reported the basic performance of an NO sensor which is applicable to in vivomeasurement. This sensor can detect NO in the blood stream in an anesthetized rat and somebiological samples including a dialysis solution. In these studies we found that NO itself showeddynamic changes different from these of oxidative products of NO (nitrite and nitrate) that areconventionally used as indices of NO level. We have also developed a catheter-type NO sensor anddemonstrated validity of the sensor for in vivo measurement of NO in the aorta and coronary sinus ofanesthetized dogs and rabbits. Both endothelium-dependent and -independent NO productions couldbe detected. In conclusion, the NO sensor and the catheter-type NO sensor are applicable to directmeasurement of NO in biological samples even in vivo and will contribute to clarification ofphysiological roles of NO

    Notes: Abstract Effects of E-5110, a novel non-steroidal antiinflammatory drug, on interleukin-1 (IL-1) generation from human monocytes were studiedin vitro. E-5110 reduced the amounts of extra- and intracellular IL-1 activity induced by lipopolysaccharide (LPS, 1 μg/ml) in a dose-dependent manner (1–10μM). E-5110 also inhibited the IL-1 generation induced by antigen-antibody complexes, opsonized zymosan and silica particles. It was suggested that the inhibition of IL-1 generation by E-5110 was independent of the inhibitory effects on arachidonate cyclooxygenase and/or lipoxygenase because indomethacin, piroxicam, BW755C and AA861 had no effects on IL-1 generation. Hydrocortisone (IC50:0.084 μM), aurothioglucose (11.5 μM) and lobenzarit (75.0 μM), which are clinically effective antirheumatic drugs, also inhibited IL-1 generation, like E-5110 (1.21 μM). It is expected that E-5110 will be superior to classical non-steroidal antiinflammatory drugs in medical treatment of rheumatoid arthritis.

    Notes: Summary The introduction of acrylate resins (Lowicryl K 4M, LR White) into electronmicroscopic immunocytochemistry applied to embedded tissue (post-embedding method) has improved the localization of antigens because of a satisfactory preservation of both ultrastructure and antigenicity of tissues. Here we describe a method that allows double staining of intracellular and membranous determinants in ultrathin sections of nervous tissue and cultures of peripheral nervous system cells. Ultrathin sections of the rat central nervous system fixed on uncoated grids were stained first for MBP selectively on the one face, then the opposite face was stained for GFAP using monoclonal antibodies and indirect immunogold staining method (IGS). Cultured Schwann cells induced to express major histocompatibility complex (MHC) class II antigens were stained for class. H antigens by pre-embedding method then followed by post-embedding IGS for the other intracytopasmic antigens.

    Notes: Summary The binding of disopyramide (DSP) to plasma (or serum) proteins was determined using an ultrafiltration technique in three patients undergoing haemodialysis. An increase in the free fraction (FF) of DSP during dialysis occurred together with elevation of the free fatty acid (FFA) level in plasma. The effect of FFA on protein binding in vitro was examined using DSP- and FFA-spiked solutions containing human α1-acid glycoprotein and serum albumin. The FF of DSP rose in proportion to increasing FFA levels, supporting the in vivo observations. The findings suggest that the free concentration of DSP should be routinely monitored, especially in haemodialysis patients.
