    Notes: A mock crime experiment was conducted to examine whether enhanced responding to concealed information during a polygraph examination is due to orienting or defensive responding. Thirty-six undergraduate students enacted one of two mock crimes. Pictures related to both crimes were presented while heart rate, magnitude of the skin conductance response, and reaction times to a secondary probe were measured. Compared to control pictures, participants showed heart rate deceleration and enhanced electrodermal responding to pictures of the crime they had committed. Probe reaction times did not differ significantly between crime and control pictures. The present findings support the idea that the orienting reflex accounts for the enhanced responding to concealed information. Theoretical and practical implications of the orienting account are discussed.

    Notes: Treatment of male Sprague–Dawley rats with kainic acid (10 mg/kg, i.p.) triggered limbic seizures in 60% of the animals starting within 30 min and lasting for about 6 h. Cyclooxygenase-2 (COX-2) mRNA was strongly induced in the pyramidal cells of the hippocampus, in the amygdala and the piriform cortex after 8 h, as shown by in situ hybridization, and returned to control levels after 72 h. At this time marked cell loss occurred in the CA1–CA3 areas of the hippocampus. We hypothesize that rofecoxib, a selective COX-2 inhibitor, might abbreviate the late neurotoxicity, possibly associated with COX-2 induction. Animals which developed seizures were treated for 3 days with rofecoxib (10 mg/kg, i.p., n = 12) starting 6 or 8 h after kainic acid injection. Histological staining of viable cells confirmed that rofecoxib treatment selectively diminished cell loss in the hippocampus. The TdT-mediated dUTP nick end labelling (TUNEL) technique was used to estimate delayed cell death. Abundant TUNEL-positive cells were detected in seizure rats 72 h after kainic acid injection in pyramidal cells of the hippocampus (CA1–CA3), in cells of the thalamus, the amygdala and the piriform cortex. Treatment with rofecoxib selectively and significantly (P 〈 0.05) attenuated the number of TUNEL-positive cells in the hippocampus, whereas the cells of the thalamus, amygdala and piriform cortex were not protected. Therefore we conclude that COX-2 might contribute to cell death of pyramidal cells of the hippocampus as a consequence of limbic seizures.

    Notes: A theory is presented together with simulation results that describe three-dimensional position detection of a sphere located in a highly focused beam by back-focal plane interferometry. This technique exploits the interference of scattered and unscattered light, which is projected on a quadrant photodiode placed in the back-focal plane of a condenser lens. Due to the Gouy-phase shift inherent in focused beams, it is not only possible to determine the lateral but also the axial position of a spherical particle with nanometer accuracy. In this paper we describe the calculation of arbitrary focused electromagnetic fields, the Gouy phase shift, Mie scattering by focused beams and the resulting position signals using the angular momentum representation. The accuracy and the sensitivity of the detection system are investigated theoretically for various sphere parameters. Both accuracy and sensitivity depend on the incident light distribution as well as on the particle's properties and position. It is further shown that the maximum capture angle of the detection lens influences the detector's sensitivity in a nonlinear manner. Additionally, for optical trapping applications the influence of the laser power is taken into account and is considered through a noise analysis. For all investigated trapping conditions the reconstructed position deviates on average 〈1 nm laterally and 〈5 nm axially from the actual particle position. © 2002 American Institute of Physics.

    Notes: We present a scanning probe microscope based on optical tweezers for three-dimensional imaging of the topology of transparent material in the nanometer range. A spherical nanoparticle serves as a probe. An optical trap moves it through the sample (e.g., a polymer network), while the position of the particle center is recorded by three-dimensional interferometry. Accessible volumes are reconstructed from the histogram of thermal position fluctuations of the particle. The resolution in determining the position of surfaces in three dimensions is about 20 nm. © 2001 American Institute of Physics.
