    Notes: The dark decay of holograms stored in iron-doped photorefractive lithium-niobate crystals is studied for samples containing up to 0.25 wt% Fe2O3 (iron concentration 71×1018 cm−3). The oxidation/reduction state of the crystals, i.e., the concentration ratio of Fe2+ and Fe3+ ions, is changed in a wide range by thermal annealing. The dark decay is attributed to two effects: An ionic dark conductivity arising from mobile protons and an electronic dark conductivity caused by tunneling of electrons between iron sites. The latter is proportional to the effective trap density, i.e., to the density of charge carriers which can be moved between the iron sites. The proportionality factor is the specific dark conductivity which increases exponentially with the third root of the entire iron concentration. © 2000 American Institute of Physics.

    Notes: We have investigated tunable distributed feedback (DFB) lasers based on InGaAsP quantum wells grown by molecular-beam epitaxy. Two-section tunable DFB lasers were fabricated by patterning laterally gain coupling binary superimposed gratings perpendicular to the ridge waveguide. Side mode suppression ratios of up to 42 dB have been achieved. The tuning range covers 25 nm. © 2001 American Institute of Physics.

    Notes: Summary Surgical patients develop a fluid deficit during pre-operative starvation. This study examines the effects of pre-operative fluid administration on haemodynamic variables, oxygenation and splanchnic perfusion in patients undergoing elective coronary artery bypass grafting. Forty-eight patients were randomised to receive either a pre-operative crystalloid infusion (crystalloid group, n = 24) or no infusion (control group, n = 24). Patients in the crystalloid group received a continuous infusion of Ringer's solution at 1.5 ml.kg−1.h−1 from 22:00 h until induction of anaesthesia the next morning. Immediately before induction of anaesthesia, all patients were given a colloid infusion to increase pulmonary capillary wedge pressure and central venous pressure to similar levels inboth groups. Haemodynamic and oxygenation parameters were measured using invasive cardiovascular monitoring, and splanchnic perfusion was assessed by indocyanine green clearance. Patients in the crystalloid group received a mean (SD) of 1008 (140) ml of Ringer's solution overnight. Patients in the crystalloid group had a higher splanchnic blood flow than the control group before induction of anaesthesia [mean (SD) = 1782 (573) ml.min−1 vs. 1391 (333) ml.min−1, p 〈 0.05]. There were no significant differences in systemic haemodynamic data and global oxygenation parameters between the two groups. Pre-operative infusion of crystalloid appears to result in an improvement in pre-operative splanchnic perfusion.

    Notes: Summary Volatile anaesthetics differ in the effects they have on splanchnic haemodynamics and oxygenation. The aim of this study was to evaluate the effects of desflurane and isoflurane as part of a balanced anaesthetic technique on intestinal tissue oxygenation during colorectal surgery. Data were analysed from 44 patients randomly assigned to receive either desflurane (desflurane group, n= 20), or isoflurane (isoflurane group, n= 24) for inhalational anaesthesia. Tissue oxygen pressure (Ptisso2)␣was measured on the serosal side of the large intestine prior to colonic resection (T1) and␣following the completion of the bowel anastomosis (T2). In addition, haemodynamic and oxygenation parameters were assessed. No difference in mean Ptisso2 was observed between the groups at T1 [desflurane group: 8.1 (2.9) kPa vs. isoflurane group: 7.7 (2.7) kPa]. Following completion of the anastomosis (T2) mean Ptisso2 was higher in the isoflurane group [9.6 (2.9) kPa] than the desflurane group [7.7 (2.4) kPa, p = 0.025]. During surgery no difference between the␣groups could be observed with regard to haemodynamics and global oxygenation parameters. The lack of a difference between the groups in Ptisso2 before resection of the colon suggests that,␣under normal conditions, desflurane and isoflurane have comparable effects on intestinal blood␣flow and oxygenation. However, following local ischaemia, the reactive hyperaemia seems␣to␣be better preserved during isoflurane anaesthesia indicated by a local increase in Ptisso2␣(p = 0.013) following completion of the bowel anastomosis.

    Notes: Slow relaxation due to dense packing encumbers the simulation of a variety of many particle systems like, for instance, collapsed polymers (folded proteins) or structural glasses. We propose to overcome this problem by a new algorithm, assigning each particle a fourth space coordinate and treating an expanded ensemble of systems of which the one with all fourth coordinates equal to zero is the physical system. An application of this simulation strategy to the coil–globule transition of homopolymers using the bond-fluctuation model reveals a hitherto unfound first-order liquid to solid transition of the collapsed globule. © 2001 American Institute of Physics.

    Notes: We have carried out Monte Carlo simulation of the fusion of bilayers of single chain amphiphiles which show phase behavior similar to that of biological lipids. The fusion mechanism we observe is very different from the "stalk" hypothesis. Stalks do form on the first stage of fusion, but they do not grow radially to form a single bilayer diaphragm. Instead, stalk formation destabilizes the membranes and results in hole formation in the vicinity of the stalks. When holes in each bilayer nucleate spontaneously next to the same stalk, an incomplete fusion pore is formed. The fusion process is completed by propagation of the initial connection, the stalk, along the edges of the aligned holes. © 2002 American Institute of Physics.

    Notes: The wetting behavior of a polymer liquid on a coated substrate is investigated via numerical self-consistent field calculations and experiments. The polymer does not wet the coated substrate, but the substrate might stabilize a mesoscopic film. Experiments observe a stable mesoscopic film of nanometer thickness in coexistence with macroscopic drops at high temperatures. Upon cooling, this mesoscopic polymer film breaks up into droplets (nano-dewetting). Our self-consistent field calculations suggest that the stability of the mesoscopic film is determined by a subtle interplay between van der Waals forces and short-ranged forces due to the distortion of the profile in the vicinity of the substrate. © 2001 American Institute of Physics.

    Notes: A new algorithm for generating starting polymer structures for molecular simulations (e.g., MD) in dense phase is presented. The algorithm yields structures that fulfill to a large extent rotational isomeric state (RIS) probabilities and avoid atomic overlap. The heuristic search bases on the new parallel-rotation (ParRot) technique. We tested the performance of the algorithm on two polymeric systems: Atomistic polyethylene and polystyrene. The algorithm permits to tackle the problem of packing chains into large boxes of size up to 50 Å in a couple of hours on common workstations. Moreover, our packing algorithm is applicable for general polymer systems. The algorithm requires CPU effort scaling with a power 2.8 in the chain length, and with a power 1.5 in the number of chains. © 2001 American Institute of Physics.

    Notes: Growing experimental evidence supports a broadening role for the caspases; not only do they participate in the process of apoptosis but also in the control of the cell cycle and cellular proliferation. The biological role of the caspases in the process of T-cell activation and proliferation is still not defined. In the present study, we propose a potential role, by demonstrating an association of T-cell receptor-mediated caspase activity with the development of an apoptosis-resistant memory CD45RO+ T-cell population. As previously shown by us, a time-dependent induction of caspase activity, in the absence of apoptosis, can be observed in CD3-stimulated human peripheral blood lymphocytes. We here show that a population of CD45RO+ cells, with activated caspase-3 and with resistance to tributyltin-induced apoptosis, develops after 3 days of stimulation. A concomitant expression of the anti-apoptotic protein Bcl-xL accompanied the caspase activity and the development of the apoptosis-resistant phenotype. Finally, upon co-culturing with dexamethasone (DEX), the CD3-induced caspase-3 activity was blocked. During this condition, the expression of the activation marker HLA-DR as well as the cellular proliferative response was strongly suppressed. The development of memory cells with a CD45RO+ phenotype was also blocked. Our data support the hypothesis that caspase-3 activity, observed in CD3-stimulated cells, may be an important component in the proliferation process and, furthermore, might play a role for the development of memory T cells, and DEX inhibits this process.

    Notes: Numerous investigations have confirmed an important role for multidrug-resistance gene 1-type P-glycoproteins (MDR1-type P-gps) in the blood–brain barrier, protecting the brain against the accumulation of a wide range of toxic xenobiotics and drugs. Several studies have provided evidence in vitro that certain steroid hormones are transported by MDR1-type P-gps; however, the question of whether this might also apply to the situation in vivo still remained to be determined. We used mice deficient for both murine mdr1a and mdr1b P-gps [mdr1a/1b(−/−)] to determine the uptake of [3H]-cortisol, [3H]-corticosterone, [3H]-aldosterone and [3H]-progesterone into the plasma, brain, testes, liver, spleen, pituitary and adrenal glands. We provide evidence that the access of the endogenous steroid hormones corticosterone, cortisol and aldosterone is regulated by MDR1-type P-gps in vivo. As peripherally administered steroid hormones accumulate in the brain of mice deficient for MDR1-type P-gps, mdr1a/1b proteins are likely to transport these hormones out of the brain, providing a kinetic barrier to their entry. Intracerebral progesterone concentrations are influenced by MDR1-type P-gp function as well; however, the effects are only small. In addition, all four endogenous glucocorticoid hormones accumulated in the testes of mdr1a/1b(−/−) mice. Our findings underline the importance of MDR1-type P-gps as an endogenous barrier system controlling the access of endogenous steroid hormones at the blood–brain barrier to maintain homeostatic control and to protect central nervous system neurones.
