    Notizen: Abstract. We have studied inclusive muon events using all the data collected by the TOPAZ detector at $\sqrt{s}=58$ GeV with an integrated luminosity of 273pb $^{-1}$ . From 1328 inclusive muon events, we measured the ratio $R_{q\bar{q}}$ of the cross section for $q\bar{q}$ production to the total hadronic cross section and forward-backward asymmetry $A_{FB}^{q}$ for b and c quarks. The obtained results are $R_{b\bar{b}}=0.13\pm0.02(stat)\pm0.01(syst)$ , $R_{c\bar c}=0.36\pm0.05(stat)\pm0.05(syst)$ , $A_{FB}^{b}=-0.20\pm0.16(stat)\pm0.01(syst)$ and $A_{FB}^ c=-0.17\pm0.14(stat)\pm0.02(syst)$ , in fair agreement with a prediction of the standard model.

    Notizen: Electron emission spectra obtained by thermal collisions of He*(2 3S) metastable atoms with CO on Ni(100) in the c(2×2) structure and on Ni(111) in the c(4×2) structure were measured to probe directly the spatial electron distribution. For a systematic comparison, the metastable spectra of free CO, condensed CO on Ni(111), and gaseous Cr(CO)6 were also measured under the same beam conditions. Our data showed that the relative ionization cross sections for the CO 4σ-, 1π-, and 5σ-derived states depend drastically on the molecular orientation of CO with respect to the metastable beam, reflecting the local electron density of CO in the impact region. Moreover, it was found that the 4σ- and 5σ- derived states of CO at hollow sites on Ni(111) are strongly modified in space by mixing with each other, where considerable charge transfer occurs from the C site to the O site in the 5σ-derived state and in the opposite way in the 4σ-derived state. In contrast, such a strong charge redistribution was not seen in the cases of terminal CO on Ni(100) and Cr(CO)6. These findings were in good accordance with the crystal orbital overlap population obtained by density functional theory through a generalized gradient approximation. © 2001 American Institute of Physics.

    Notizen: CD30+ large anaplastic lymphoid cells are seen in anaplastic large cell lymphoma (ALCL), and also in lymphomatoid papulosis (LyP) and other lymphoproliferative disorders. It can be difficult precisely to categorize these disorders with CD30+ cells. We report a case of primary cutaneous CD30+ ALCL with systemic metastases in whom the clinical disease subsequently evolved into LyP. The patient was initially administered cisplatin and etoposide and made a good response. Eighteen months later, recurrent, self-healing cutaneous small nodules appeared around the original tumour site without any systemic involvement. Histopathological examination of the recurrent lesions revealed infiltration with a mixture of cells that included neutrophils, eosinophils and CD30+ large anaplastic cells cytologically identical with those in the primary lesion. The anaplastic cells in both the primary and recurrent lesions were positive for monoclonal antibodies CD30, CD25 and a monoclonal antibody directed against the chimeric protein p80NPM-ALK. These observations suggest the possibility that the ALCL and the subsequent LyP represent different clinical manifestations of proliferation of the same clone.

    Notizen: Background Kimura's disease (KD) is a chronic inflammatory disorder characterized by tumours in the head and neck region, enlarged lymph nodes, increased eosinophil counts and high serum IgE. Mast cells are known to play a central role in IgE-mediated allergic diseases through the release of inflammatory mediators like IL-4, IL-5 and chemokines. We hypothesized that mast cells may play a role in the pathogenesis of KD by regulating eosinophilic infiltration and IgE synthesis.Objective In order to investigate the role of mast cells in the pathogenesis of KD, we examined the expression of cytokines/chemokines in the lesions of KD.Methods We examined the number of tryptase+ cells, EG2+ cells, CD3+ cells, IL-4+ cells, IL-5+ cells, eotaxin+ cells, RANTES+ cells and CCR3+ cells in five specimens of KD versus normal tissues by immunohistochemistry. The sources of IL-4, IL-5, eotaxin and RANTES and the expression of CCR3 were examined by immunostaining of serial sections with antibodies to IL-4, IL-5, eotaxin, RANTES and CCR3, and antibodies to tryptase, ECP (EG2) and CD3.Results Mast cells, activated eosinophils, T cells, IL-4+ cells, IL-5+ cells, eotaxin+ cells, RANTES+ cells and CCR3+ cells were all increased in the lesions of KD as compared with those in normal tissue. Mast cells and T cells were the major source of IL-4, whereas mast cells, T cells and activated eosinophils were the main source of IL-5. Mast cells, T cells and activated eosinophils were the main source of eotaxin and RANTES.Conclusions The number of IL-4, IL-5, eotaxin and RANTES-expressing mast cells and T cells were increased in the lesions of KD. As mast cells are lesional resident cells, these cells may play an important role in the pathogenesis of KD by regulating IgE synthesis and orchestrating eosinophilic infiltration.

    Notizen: Background The production of IgE in B lymphocytes is down-regulated by IFN-γ. IL-12 induces IFN-γ production by T lymphocytes and natural killer cells by binding to its specific receptor. RNA editing is a post-transcriptional modification.Objective Here we show that the RNA editing of IL-12 receptor (R) β2 is associated with atopy.Methods Atopic patients and non-atopic healthy controls were studied. Fragments of IL-12R β2 cDNA and genomic DNA were amplified and sequenced. Furthermore, the function of the IL-12R β2 chain was investigated.Results Sequence analysis of the cDNA clones representing IL-12R β2 mRNA transcripts revealed a C-to-U conversion at nucleotide 2451 (Ala 604 Val) on exon 13 in some atopic patients. Surprisingly, sequence analysis of their genomic DNA showed no 2451 C-to-T (Ala 604 Val) mutation. We concluded that the observed C-to-U mismatch in the cDNA clone is due to a post-transcriptional modification, RNA editing. The C-to-U conversion was observed in 21 (20.6%) of 102 atopic patients, whereas this conversion was observed in only 4 (3.8%) of 104 non-atopic subjects (P〈0.001). IFN-γ production by peripheral blood mononuclear cells (PBMCs) stimulated with IL-12 in the subjects with the C-to-U conversion was significantly lower than that in the subjects without the C-to-U conversion. In atopic patients with the C-to-U conversion, PBMCs faintly showed the tyrosine phosphorylation of Stat4, and the IgE production by PBMCs was not suppressed by IL-12 whereas it was suppressed by IFN-γ.Conclusions The RNA editing of IL-12R β2, 2451 C-to-U (Ala 604 Val) conversion causes impairment of the IL-12 signal cascade and the subsequent reduction in IFN-γ production, resulting in the impaired down-regulation of IgE production. This is the first report indicating that atopy is associated with RNA editing.

    Notizen: Background Food allergies are more prevalent in children, due to the immature gastrointestinal epithelial membrane barrier allowing more proteins through the barrier and into circulation. Ovomucoid (OM) is one of the major allergens that is found in egg white.Objective The aim of this study was to determine T cell epitopes, antigen-presenting human leucocyte antigen (HLA) class II molecules of the T cell lines (TCLs) and T cell clones (TCCs), and complementarity determining region (CDR) 3 loops of the T cell receptor (TCR) α and β chains of the TCCs specific to OM.Methods We established TCLs and TCCs specific to OM from peripheral blood mononuclear cells (PBMCs) of four atopic patients with egg-white allergy using a mixture of a panel of overlapping synthetic peptides corresponding to the amino acid sequence of the entire OM. We identified the T cell epitopes by antigen-induced proliferative responses, antigen-presenting molecules using allogeneic PBMCs and CDR3 loops of the TCR α and β chains by cloning and sequence analysis.Results The TCLs and TCCs responded to seven different peptides, and their antigen-presenting molecules were different from each other. Sequence analysis of the TCR α and β gene usage of the TCCs showed marked heterogeneity, and the usage of the CDR3 loop of the TCCs involved heterogenous amino acid residues. Interestingly, TCCs ‘IH3.3’ and ‘YT6.1’ recognized the same OM peptides, and had the same TCR Vβ-Jβ gene usage. Considering that peptide motifs bind to HLA class II molecules, the electrically charged residue (positive or negative) on the CDR3α and the CDR3β loops of TCR of TCC may form ionic bonds with a charged residue on the HLA class II molecules-peptide complex.Conclusions TCCs that have the same TCR gene usage were established from patients who had shown similar hypersensitivity-type, indicating that antigen recognition by a specific TCR is closely associated with the characteristics of each patient's symptoms.
