    Notes: Helminthosporium leaf blight (HLB) is the most important disease constraint to wheat (Triticum aestivum L.) cultivation in the eastern Gangetic Plains of South Asia. A Helminthosporium Monitoring Nursery (HMN) including potential adapted and exotic sources of HLB resistance was developed in Bangladesh, India and Nepal to assess the stability of genetic resistance across locations. The 8th, 9th and 10th HMN assessed the HLB resistance and agronomic traits of 17 wheat genotypes across 20 environments of Bangladesh, India and Nepal in the 1999-2000, 2000-2001 and 2001-2002 cropping seasons, respectively. The area under the disease progress curve (AUDPC) for HLB, grain yield, thousand-kernel weight (TKW), days to heading, days to maturity, and plant height were examined. The 17 genotypes showed a range of variability for disease and agronomic characters. Disease severity (AUDPC) differed in the 3 years and showed the highest values in 2002. The increase in AUDPC in 2002 caused the lowest grain yield, with an average 18% reduction due to HLB. A few genotypes (SW 89-5422, Yangmai-6 and Ning 8201) appeared to have stable HLB resistance across environments. However, most of the higher-yielding genotypes, except BL 1883, were unstable. The results suggest that careful selection of HLB resistance with acceptable grain yield, TKW and plant height may be possible using the wheat genotypes included in the HMN.

    Notes: In ulcerative colitis, hydrogen sulphide is postulated to impair colonocyte butyrate metabolism, leading to cellular energy deficiency and dysfunction.〈section xml:id="abs1-2"〉〈title type="main"〉Aims:To determine the effects of sulphide exposure on butyrate metabolism and adenosine triphosphate levels of HT29 colonic epithelial cancer cells, and to establish whether energy deficiency can be prevented by increased butyrate concentrations or the presence of glucose.〈section xml:id="abs1-3"〉〈title type="main"〉Methods:HT29 cells were maintained in medium containing 3 mM butyrate, 5 mM glucose, or both substrates. Oxidation rates were measured by 14CO2 release from 14C-labelled substrates. Cellular adenosine triphosphate was assayed using the luciferin/luciferase chemiluminescent method. The effects of sulphide (0–5 mM) on substrate oxidation and adenosine triphosphate levels and of increasing butyrate concentration (0–30 mM) with sulphide were observed.〈section xml:id="abs1-4"〉〈title type="main"〉Results:HT29 cells showed similar energy substrate usage to primary colonocyte cultures. Sulphide exposure inhibited butyrate oxidation and led to a reduction in cellular adenosine triphosphate. This fall was prevented by co-incubation with glucose, but not by increasing concentrations of butyrate.〈section xml:id="abs1-5"〉〈title type="main"〉Conclusions:HT29 cells utilize butyrate as an energy substrate and represent a useful in vitro model of the effects of sulphide on colonocytes. Sulphide inhibits butyrate oxidation and leads to demonstrable energy deficiency, prevented by the presence of glucose but not by increased butyrate concentrations.

    Notes: CD40 is a member of the tumour necrosis factor (TNF) cell surface receptor family. It plays an important role in T-cell dependent B-cell functions and in dendritic cell development. It has also been identified as a cell surface receptor on endothelial cells. In this manuscript, we report that human dermal microvascular endothelial cells (HDMEC) express cell surface CD40 and that the expression of CD40 is increased by the proinflammatory cytokines TNF-α and interferon (IFN)-γ. Additionally, we demonstrate that engagement of HDMEC CD40 with its recombinant CD40 ligand augments the induction of E-selectin, but not of intercellular or vascular cell adhesion molecules on the surface of HDMEC. Furthermore, we show that IFNγ stimulation of HDMEC results in increased binding of Jurkat leucocytes to HDMEC by a CD40–CD154 dependent pathway. This study thus provides evidence that CD40 expression in HDMEC is regulated by proinflammatory cytokines, and that CD40 functions as an important mediator of cutaneous inflammation.

    Notes: Pre-operative optimisation of high-risk patients undergoing major elective surgery has been shown to decrease peri-operative morbidity and mortality. It is also cost effective because of the resulting decrease in postoperative complications. A questionnaire was sent to 170 intensive care and high dependency units in Britain in order to quantify the number of units practising pre-operative optimisation. There was a 91% response rate. Of the respondents familiar with the evidence advocating pre-operative optimisation, 91% believe pre-operative optimisation improves outcome but only 62% admit patients for such preparation. Moreover, only eight units (6%) admit more than 25% of eligible patients. The reasons given for not admitting such patients pre-operatively are a lack of manpower, beds or both. This survey demonstrates the need for greater investment of resources into intensive care and high dependency units, so that clinicians can deliver high-quality evidence-based healthcare in accordance with the principles of clinical governance.

    Notes: Purified mouse serum amyloid P-component (SAP; 0.5-50 µg/kg), injected intravenously into Swiss mice, induced the production of serum colony-stimulating factors (CSFs); the maximum induction was observed at 10.0 µg/kg. Further, in vitro purified mouse SAP (0.1-50 µg/ml) stimulated the mouse elicited peritoneal macrophages to elaborate CSFs in the conditioned medium (CM); 5.0 µg/ml SAP appeared to be the optimum. Both in vivo and in vitro the maximum production of CSFs occurred 6 h after initiation of stimulation, and returned to the background levels by 48 h. Mannose 6-P, mannose 1-P and mannose, and not other sugars inhibited the SAP-induced production of CSFs by macrophages which suggests that SAP interaction with macrophages was mediated by specific glycoprotein-receptors. A neutralizing (100%) concentration of rabbit antimouse interleukin (IL)-1 polyclonal antibody had no effect on the SAP-induced CSF production, indicating that it would be IL-1-independent. SAP-induced CSFs, both in serum and CM, were functionally similar as they supported the formation of granulocyte (G), macrophage (M) and GM colonies in similar proportions. The production of CSFs appeared to be lipopolysaccharide (LPS)-independent as it was not inhibited by polymyxin B sulfate (25.0 µg/ml), and heat-inactivated (80 °C, 1 h, pH 7.0) SAP did not induce the production of CSFs. The CSFs were produced de novo because cylcoheximide (50.0 µg/ml) completely inhibited their production. These results demonstrate that purified mouse SAP, in a dose-dependent manner, can induce the production of serum CSFs in mice, and can induce LPS-independent de novo production of CSFs by elicited macrophages in vitro.
