Notes: Abstract Both calreticulin (CRT) and immunoglobulin binding protein (Bip) have a role in the folding and assembly of oligomeric membrane proteins in the endoplasmic reticulum (ER). Recent studies have demonstrated the generation of β-amyloid protein (Aβ) 1–42, a key peptide for amyloid deposits, in the ER. We, therefore, examined the localization and expression of CRT, Bip and their mRNA by immunohistochemistry, Western blot, in situ hybridization and semiquantitative reverse transcription polymerase chain reaction (RT-PCR) in both neurologically normal and Alzheimer’s disease (AD) brains. Two polyclonal anti-CRT antibodies gave similar positive staining of CRT in neurons and glia. In neuronal cells, the cytoplasm, nucleoli and their processes were positive for CRT. In glial cells, perinuclear staining was frequently seen and the processes of some glial cells were also stained. In AD, these antibodies stained clearly damaged neurons but the number and the intensity of positive cells were decreased compared to controls. Processes of microglial cells were markedly positive in the AD white matter. Western blots using an anti-CRT antibody showed significantly lower immunoreactive bands in AD than control brains. By in situ hybridization, the number of neurons which express the CRT mRNA was less in AD than in controls. Using RT-PCR, the relative levels of the CRT mRNA in AD brains were also found to be significantly lower than those in controls. On the other hand, the number of Bip-positive cell, the production of Bip and the expression of mRNA for Bip did not differ between control and AD brains. These results suggest that CRT may be a multifunctional protein in human brain, and that the weak expression of CRT and the positive staining of microglial processes in AD brain may be part of the pathological processes in AD.

Notes: Blood flow stagnation is said to be an important factor for the incidence of dark eye circles, but there are few reports, which have studied dark eye circles and the relationship to blood flow. We have examined dark eye circles dermatologically using a non-invasive method and made clear the factor related to it. We have also studied the effect of newly developed cosmetics on improvement of dark eye circles. Melanin and haemoglobin content, haemoglobin oxygen saturation and blood flow rate were measured to compare dark eye circles with a normal control. An increased amount of haemoglobin and decreased haemoglobin oxygen saturation were observed at the site of dark eye circles. We also observed that the blood flow rate was slow at dark eye circles in comparison with cheek. From these results, blood flow stagnation seems to be one of the factors that could induce dark eye circles. In addition, the increase of melanin at dark eye circles was also observed in older subjects. It was considered that not only congestion but also an increase of melanin content in the skin could induce dark eye circles. On the basis of these results, we developed anti-dark eye circle cosmetics that can improve blood circulation, reduce melanin content and maintain high moisture retention in the skin. After 3 weeks’ usage of this product, it was confirmed that dark eye circles were improved, judging from the result of non-invasive instrumental measurements.

Notes: Background Increased vascularity in airway mucosa is a distinctive feature of airway remodelling in asthma. While corticosteroids have proved most effective in modifying airway inflammation, the effect of inhaled corticosteroids on increased airway mucosal vascularity in asthmatics has been little studied. Objective We examined the effect of inhaled corticosteroid on airway vascularity in bronchial biopsy specimens taken from asthmatic patients.Subjects and methods We studied bronchial biopsies from 28 asthmatic patients before and after treatment with inhaled beclomethasone dipropionate (BDP) 800 µg/daily, or placebo, for 6 months in a double-blind manner. Biopsy specimens were evaluated for number of vessels and percentage of area occupied by vessels, using computerized image analysis after staining for type IV collagen in vessel walls. Specimens were also examined for extent of collagen III in the subepithelial basement membrane. In addition, we compared asthmatic specimens with biopsy specimens taken from non-asthmatic control subjects. Results There was a significant increase in number of vessels (P 〈 0.01) and percent vascularity (P 〈 0.001) in the submucosa of asthmatic patients compared with control subjects. After 6 months of treatment, we observed significant improvements in forced expiratory volume in 1 s (FEV1), FEV1% and airway responsiveness (P 〈 0.05, each) in the BDP treatment group compared with the placebo group. This was accompanied by significant decreases in both vessel number and percent vascularity in the airways of BDP-treated patients (P 〈 0.05, each). We also observed a significant correlation between change in percent vascularity and change in collagen III thickness in the BDP-treated patients (rs = 0.90, P 〈 0.001). Furthermore, the change in percent vascularity was inversely correlated with both FEV1 (rs = −0.49, P 〈 0.05) and airway responsiveness (rs = −0.36, P 〈 0.05).Conclusion These findings suggest that inhaled corticosteroid treatment of asthma reduced airway wall vascularity during airway remodelling.

Notes: There are two major subpopulations of peripheral helper T lymphocytes: T helper 1 (Th1) and T helper 2 (Th2) cells. Surgical stress increases the number of Th2 cells, and decreases that of Th1 cells, resulting in a decrease in the Th1/Th2 ratio, and, consequently, in suppressed cell-mediated immunity. Since anaesthesia can suppress the stress response to surgery, it may inhibit the decrease in the Th1/Th2 ratio. Using flow cytometry, we studied whether propofol anaesthesia (n = 9) or isoflurane anaesthesia (n = 9) had more effect on the decrease in the Th1/Th2 ratio after surgery in patients undergoing craniotomy. The Th1/Th2 ratio decreased significantly after isoflurane anaesthesia (p = 0.011), while it did not change after propofol anaesthesia. The ratio was significantly lower with isoflurane than propofol (p = 0.009). Propofol anaesthesia attenuated the surgical stress-induced adverse immune response better than isoflurane anaesthesia.

Notes: An electron of a negative hydrogen ion (H−) can be easily detached by collisions with an external laser beam or residual gas molecules in the beam line, because its electron affinity is small (0.754 eV). The detached electrons provide information about the original negative hydrogen ions and should have the same velocity as the original hydrogen ions. By measuring the energy of the detached electrons, the energy of a negative hydrogen-ion beam can be estimated. A preliminary experimental result of an energy analysis of a H− beam using detached electrons is presented. © 2000 American Institute of Physics.

Notes: We present a study of the dynamics of the excitonic magnetic polarons in Cd1−xMnxTe epilayers. The transient photoluminescence measured by a streak camera under resonant excitation is analyzed with a special attention devoted to line shape of the emission signal. A nonradiative spectral region is observed in zero field and is strongly suppressed in a field of 6 T. This observation is understood to be directly related to the alignment of the Mn2+ spins and is therefore closely related to the formation of magnetic polarons. A few Mn spins are found to be aligned already within 10 ps, but the total Mn spin polarization in the sample is found to increase exponentially with a rise time of 37 ps in Cd0.73Mn0.27Te epilayers. Furthermore, the values of 19.2 and 12.4 meV were determined for the energies of the excitonic magnetic polarons in Cd0.73Mn0.27Te and Cd0.79Mn0.21Te epilayers, respectively. © 2000 American Institute of Physics.

Notes: A new apparatus has been developed to detect and measure angular correlations between energy-selected photoelectrons and coincident mass-analyzed fragment ions from photoionization at selected wavelengths. It achieves velocity imaging for electrons and ions simultaneously and has high collection efficiency for both particles, with moderate mass and energy resolution. Angular and energy correlations between the two particles are measured, as are the angular distributions of each particle independently relative to the light polarization direction. Fixed-molecule electron angular distributions are deduced in cases of pure axial recoil. Examples of angular distributions from photoionization of diatomic molecules are reported. © 2000 American Institute of Physics.

Notes: In order to develop a magnetic material for high-density magnetic recording media which overcomes the thermal agitation effect, the magnetic properties, such as saturation magnetization and magnetic anisotropy, were evaluated for Co–M (M=Pt, Pd, Ru, Rh, Os, Cr, Ge, Mo, Mn, Re, and Si) binary alloys. Ge addition was found to be effective in maintaining the value of the magnetic anisotropy field of Co. Furthermore, a ternary magnetic alloy of Co–Ge–Cr (85 at. % Co, 10 at. % Ge, 5 at. % Cr) was found to have the following magnetic properties: Kugrain≥3×106 erg/cm3, Hkgrain(approximate)10 kOe, and 4πMs/Hkgrain〈1.0. This magnetic material is a very promising candidate for use in high-density magnetic recording media. © 2000 American Institute of Physics.

Notes: We present theoretical evidence for the creation of an electron–hole pair at an edge-sharing SiO4 site that is supposed to exist in a-SiO2 as an intrinsic structural defect. The present electron–hole pair consists of a nonbridging oxygen hole center and an E′ center, but these paramagnetic defects do not form a close pair but are separately located by over ∼4 Å. The subsequent decay mechanism along with the related radiolytic process is also discussed. © 2002 American Institute of Physics.
