    Notes: This study was carried out to investigate whether readily available paprika can replace astaxanthin as a source of carotenoids in soft-dry pellets for broodstock yellowtail Seriola quinqueradiata (Temminck & Schlegel). Five months prior to spawning, 120 fish of average body weight (BW) 6.2 ± 1.5 kg were divided into three groups and fed basal soft-dry pellets (SDP), SDP supplemented with 30 mg/kg astaxanthin (a-SDP), or SDP supplemented with 2% paprika (p-SDP). Feeding was carried out in floating net cages and two spawning trials were performed. Fish were selected (five male and five female per group per trial), injected with human chorionic gonadotropin (600 IU kg−1 fish) and transferred to 100 m3 indoor concrete tanks for spawning. In trial 1, egg production during the first day of spawning was highest for the p-SDP group, followed by the a-SDP and SDP groups. For the p-SDP, a-SDP and SDP groups, fertilization rates were 76.0%, 69.7% and 61.9% and hatching rates were 75.2%, 63.9% and 59.4%, respectively. In trial 2, fertilization rates were 80.9%, 78.0% and 86.8% and hatching rates followed the same trend as in trial 1, at 61.8%, 52.9% and 37.2%, respectively. Moreover, larvae of the p-SDP groups showed better survival at day 3 after hatching. Apart from the darker yellow colour of fertilized eggs of the astaxanthin or paprika supplemented diet groups, analysis showed that carotenoids from paprika were successfully incorporated into the eggs, mainly as zeaxanthin and lutein. Paprika supplementation did not affect the fatty acid and lipid class profiles of eggs and unfed larvae. These results indicate that paprika supplementation in soft-dry pellets is effective for good quality egg production in yellowtail.

    Notes: Investigations concerning surface crack growth are necessary for understanding the mechanism of rolling contact fatigue (RCF) of bearings because the surface defects cause flaking failures. In the present work, micro holes were artificially made prior to the RCF tests and the initiation of the surface cracks from the micro holes was observed in order to find the key factors for understanding their features. Crack initiation directions were compared to the stress intensity factors calculated by a simple method based on the 〈inlineGraphic alt="inline image" href="urn:x-wiley:8756758X:FFE771:FFE_771_mu1" location="equation/FFE_771_mu1.gif"/〉 theory. The extent to which ‘contact pressure (wedge effect)’ and ‘contact stresses’ are applicable for understanding the correlations between the crack initiation directions and stress intensity factors is discussed. The crack initiation directions are strongly correlated to the stress intensity factors caused by the contact stresses alone. We concluded that the crack growth and initiation are dominated by stress intensity factors caused by contact stresses rather than the wedge effect.

    Notes: Surface crack growth of silicone nitride ceramic bearings under rolling contact fatigue has been investigated from the viewpoints of contact stresses (ring crack model) and fluid pressure (wedge effect model). The mechanisms of these two models have been investigated independently; however, it was impossible to separate the effects of contact stresses and fluid pressure on surface crack growth. In this paper the effects of contact stresses (ring crack model) on surface crack growth are investigated. In the ring crack model the crack growth is caused by contact stresses around the circumference of the contact circle. The growth of surface cracks located inside and outside the contact track was observed in order to obtain data from which we could reexamine the ring crack model. The outside cracks under rolling contact fatigue were propagated by contact stresses alone and also the inside cracks grew as slowly as the outside cracks. We concluded that the cracks are propagated by the single effect of contact stresses. Preliminary observations of surface crack growth showed that the cracks were unaffected by wear and residual stresses.

    Notes: Summary Cardiac hypertrophy is an adaptive process to an increased hemodynamic overload. When cardiomyocytes cultured on silicone dishes were stretched, second messengers such as protein kinase C (PKC), Raf-1 kinase, and mitogen-activated protein (MAP) kinases were activated, which were followed by increased protein synthesis. Moreover, pretreatment with an angiotensin II (AngII) type 1 receptor antagonist dimished an increase in protein synthesis, MAP kinase activity, and c-fos gene expression induced by the stretching of cardiomyocytes. These suggest the linkage of the cardiac renin-angiotensin system to the formation of pressure-overload hypertrophy. Indeed, in the stretch-conditioned medium the levels of AngII concentration were increased. Also, mechanical stretch enhanced endothelin (ET)-1 release from the cardiomyocytes and activated the Na+/H+ exchanger independently of these vasoactive peptides. In the second part, we examined AngII-induced signaling pathways both in cardiac myocytes and in cardiac fibroblasts. AngII-evoked signal transduction pathways differed between cell types. In cardiac fibroblasts AngII activated MAP kinases through a pathway including the Gβγ subunit of Gi protein, Src, Shc, Grb2, and Ras, while Gq and PKC activation was necessary in cardiac myocytes. We further explored norepinephrine (NE)-induced signaling pathways in cardiac myocytes. NE activated Raf-1 kinase and MAP kinases and increased amino acid uptake in cardiomyocytes of neonatal rats. β-adrenoceptor (AR) stimulation as well and α1-AR stimulation was involved in NE-induced MAP kinase activation. It is noteworthy that unlike in other cell types not only PKC activation but also protein kinase A (PKA) activation increased the activaties of Raf-1 kinase and MAP kinases in cardiac myocytes and induced cell growth. Finally, we observed that β-AR-induced activation of MAP kinases is dependent on both Gs/cAMP/PKA and Gi/Src/Ras signaling pathways and that phosphorylation of β-AR is critical to the cross talk between these signaling pathways.

    Notes: Abstract The priA gene of the basidiomycete Lentinus edodes possesses a pyrimidine (CT)-rich stretch (26 bp) that includes a short (6-bp) repeat, the elements of which form a mirror repeat at and near the transcriptional initiation sites. A DNA fragment that included this sequence was inserted into pBR322, and the resulting plasmids were introduced into Escherichia coli. Analysis of the susceptibility of these pBR322 derivatives to cleavage by S1 nuclease, following isolation from E. coli, indicated the formation of an open, S1-sensitive structure within and just downstream of the CT/AG-biased sequence. Replacement of two dTMP residues in one of the repeat elements by dGMP resulted in the elimination of the S1-cleavable open structure from the plasmids. To analyze the effect of the CT/AG-biased sequence from priA in the basidiomycete Coprinus cinereus, the integrating vectors pLC2 and pLC2mutCT were used; these contained the wild-type priA promoter and the mutant priA promoter with the aforementioned mutation in the mirror repeat, respectively. The Streptomyces-derived bialaphos resistance gene (bar) was fused downstream of the promoters, and the resulting plasmids, pLC2-bar and pLC2mutCT-bar, were introduced into C. cinereus. Transformants carrying pLC2mutCT-bar grew significantly more slowly on bialaphos-containing agar plates and contained a noticeably lower level of the bar transcript when compared with the transformants obtained with pLC2-bar. These results suggest that an unusual structure induced by the CT/AG-biased sequence is required for efficient gene expression from the priA promoter.

    Notes: Abstract. The fifth harmonic pulses of an intense femtosecond Ti:sapphire laser were experimentally shown to be negatively chirped by using an LiF plate as a positive dispersive medium. The chirp of the harmonic pulse originates from the intensity-dependent atomic dipole phase, which is estimated to be proportional to 25 U p, where U p is the ponderomotive energy. Consequently, we have succeeded in compressing the chirped pulses to 13 fs by compensating the intrinsic negative chirp. Chirp effects of the fundamental laser on the pulse width of the fifth harmonic were consistent with the negative chirp of the fifth harmonic.
