Notizen: Abstract The characteristics of the propagation of a detonation in chemically sensitized nitromethane in a dense porous medium are investigated. By introducing liquid NM+15% (by weight) DETA into densely packed beds of solid spherical glass beads 66μm to 2.4 mm in diameter, a highly heterogeneous explosive mixture is obtained. The critical (i.e., failure) charge diameter of this mixture is systematically measured in unconfined charges over a wide range of bead sizes. Velocity measurements are also made for the various charges. It is found that there exists a critical bead size above which the critical diameter decreases with increasing bead size and below which it decreases with decreasing bead size. This result indicates an abrupt change in the mechanism of propagation at the critical bead size. Velocity measurements further support this by emphasizing the different behavior above and below the critical point.

Notizen: Abstract Digital image processing techniques have been applied to the analysis of cellular smoked foil patterns from gaseous detonations. In particular, the two-dimensional autocorrelation function is applied to digital cell pattern images and an orientational correlation parameter is calculated. Taking line profiles along the directions of highest correlation provides an unbiased method of determining the mean cell size in each of the two principal directions. By analyzing the width, amplitude and angular position of the orientational correlation plots, information can be extracted concerning the cellular pattern regularity, the relative angular correlation between two sets of transverse waves in two directions, and the mean shape or elongation of the cells within the pattern. The technique is applied to smoked foils from oxyacetylene mixtures with argon dilutions ranging from 0 to 75% to quantify the increase in regularity with argon dilution. This method provides a simple and useful way of analyzing cellular patterns and constitutes a promising technique for improving smoked foil diagnostics.

Notizen: Abstract Near-limit detonations are highly unstable and characterized by very large longitudinal velocity fluctuations that can range from 0.4 to 1.8 times the normal Chapman-Jouguet value. The period of the fluctuations also varies over a wide range from a few to a hundred tube diameters. In an attempt to establish a criterion for detonation limits, the velocity fluctuations of near-limit detonations are studied. A novel microwave Doppler technique based on a single coaxial mode has been developed for this purpose to give an unambiguous quasi-continuous velocity measurement of the detonation wave over the entire length of its travel. The near-limit unstable behavior in the detonable stoichiometric mixtures of hydrocarbons (C2H2, C2H4, C2H6, C3H8) with O2, air or N2O, tested in this work, have been characterized by four distinct modes of unstable behavior. This classification allows a qualitative description of the wide range of velocity fluctuations occurring near the detonation limit, including galloping waves.

Notizen: Abstract The effect of the non-ionic surfactants on the ethanol fermentation was greatly dependent on the surfactant added. While Tween 20 and Tween 80 slightly enhanced ethanol fermentation, Triton X-100 which exhibited the inghest increase in the enzymatic saccharification had a negative effect on the ethanol fermentation. The negative effect of Triton X-100 on ethanol production was the most pronounced when the cellulosic hydrolyzates were used. Tween 80 showed the best performance for the ethanol production from steam exploded wood hydrolyzate.

Notizen: Abstract: The effects of CNS axotomy on glutamate transporter and glutamate receptor expression were evaluated in adult rats following unilateral fimbria-fornix transections. The septum and hippocampus were collected at 3, 7, 14, and 30 days postlesion. Homogenates were immunoblotted by using antibodies directed against glutamate transporters (GLT-1, GLAST, and EAAC1) and glutamate receptors (GluR1, GluR2/3, GluR6/7, and NMDAR1), and they were assayed for glutamate transport by d-[3H]aspartate binding. GLT-1 was decreased at 7 and 14 days postlesion within the ipsilateral septum and at 7 days postlesion in the hippocampus. GLAST was decreased within the ipsilateral septum and hippocampus at 7 and 14 days postlesion. No postlesion alterations in EAAC1 immunoreactivity were observed. d-[3H]Aspartate binding was decreased at 7, 14, and 30 days postlesion within the ipsilateral septum and 14 days postlesion in the hippocampus. GluR2/3 expression was down-regulated at 30 days postlesion within the ipsilateral septum, whereas GluR1, GluR6/7, and NMDAR1 immunoreactivity was unchanged. In addition, no alterations in glutamate receptor expression were detected within hippocampal homogenates. This study demonstrates a selective down-regulation of primarily glial, and not neuronal, glutamate transporters and a delayed, subtype-specific down-regulation of septal GluR2/3 receptor expression after regional deafferentation within the CNS.

Notizen: Abstract: Low extracellular glutamate content is maintained primarily by high-affinity sodium-dependent glutamate transport. Three glutamate transporter proteins have been cloned: GLT-1 and GLAST are astroglial, whereas EAAC1 is neuronal. The effects of axotomy on glutamate transporter expression was evaluated in adult rats following unilateral fimbria-fornix and corticostriatal lesions. The hippocampus and striatum were collected at 3, 7, 14, and 30 days postlesion. Homogenates were immunoblotted using antibodies directed against GLT-1, GLAST, EAAC1, and glial fibrillary acidic protein and assayed for glutamate transport by d-[3H]aspartate binding. GLT-1 immunoreactivity was decreased within the ipsilateral hippocampus and striatum at 14 days postlesion. GLAST immunoreactivity was decreased within the ipsilateral hippocampus and striatum at 7 and 14 days postlesion. No alterations in EAAC1 immunoreactivity were observed. d-[3H]Aspartate binding was decreased at 14 days postlesion within the ipsilateral hippocampus and at 7 and 14 days postlesion within the ipsilateral striatum. By 30 days postlesion, glutamate transporters and d-[3H]aspartate binding returned to control levels. This study demonstrates the down-regulation of primarily glial, and not neuronal, glutamate transporters following regional disconnection.

Notizen: Abstract: We have identified the regional distributions and developmental expression of NMDA-receptor proteins NR2A and NR2B in rat CNS, using two subunit-specific affinity-purified polyclonal antibodies that recognize NR2A and NR2B. In western blots of cells transfected with NR2A or NR2B cDNAs, and of brain homogenates, each antibody detects a single predominant 172-kDa protein corresponding to its homologous subunit. Both subunits are glycoproteins that are enriched in synaptic membranes. In adult rat CNS, NR2A and NR2B are enriched in cortex and hippocampus but are present in other forebrain regions. In hindbrain, NR2A is present at low levels but NR2B is barely detectable. These subunits are differentially expressed in postnatal CNS development. In cortex and striatum, NR2A is absent at birth but expression increases thereafter, whereas NR2B is expressed at nearly adult levels during forebrain development. In hindbrain, low levels of NR2A are present throughout development, whereas NR2B is expressed only transiently in the first postnatal weeks. These results suggest that native NMDA receptors are modulated by NR2A and NR2B in adult forebrain but not appreciably in hindbrain. In contrast, during early postnatal development, NR2B may have a more dominant role than NR2A in modulating NMDA receptors throughout the CNS. Thus, transient changes in NMDA-receptor function may occur during maturation of certain neuronal and/or glial populations via differential expression of NR2A and NR2B subunits.
