    Notes: Mid-infrared lead-chalcogenide diode lasers can be operated at convenient operation temperatures using short current pulses and thus allow a wide range of potential applications. Our goal was to optimize the spectral characteristics of pulsed lasers by well-defined current pulse shaping. The high-frequency current tuning coefficient has been evaluated experimentally for this reason. Different tuning mechanisms are discussed by use of a model calculation. The qualitative features can be explained by assuming a separate temperature for the carriers that form a thermodynamic system of their own and are heated by the current. The tuning characteristics can be explained due to carrier concentration variations induced by free-carrier absorption. © 1996 American Institute of Physics.

    Notes: Optical components for the X-ray radiometry beamline of the Physikalisch-Technische Bundesanstalt at BESSY II were investigated at BESSY I. Reflectometry measurements on different mirror coatings were used to calculate their capability to suppress higher-order radiation. An MgF2 coating proved to be best suited for photon energies below 4 keV. The reflectance of silicon (111) monochromator crystals at low photon energies around 2 keV was homogeneous within ±1%. In a test at a superconducting wavelength shifter the cooling system for the first crystal was shown to cope with the absorbed power of 14 W.

    Notes: Abstract Experiments with inside-out patches excised from pancreatic B-cells have yielded evidence that mitochondria are often contained in the cytoplasmic plug protruding into the tip of patch pipette. When intact B-cells were loaded with the fluorescent mitochondrial stain, rhodamine 123, and membrane patches excised from these cells, a green fluorescence could be observed in the lumen at the tip of the patch pipette. The same result was obtained with the mitochondrial stain, MitoTracker Green FM, which is only fluorescent in a membrane-bound state. Furthermore, the open probability of ATP-dependent potassium (KATP) channels in inside-out patches was influenced by mitochondrial fuels and inhibitors. Respiratory substrates like tetramethyl phenylene diamine (2 mM) plus ascorbate (5 mM) or α-ketoisocaproic acid (10 mM) reduced the open probability of KATP channels in inside-out patches significantly (down to 57% or 65% of control, respectively). This effect was antagonized by the inhibitor of cytochrome oxidase, sodium azide (5 mM). Likewise, the inhibitor of succinate dehydrogenase, malonate (5 mM), increased the open probability of KATP channels in the presence of succinate (1 mM). However, oligomycin in combination with antimycin and rotenone did not increase open probability. Although it cannot be excluded that these effects result from a direct interaction with the KATP channels, the presence of mitochondria in the close vicinity permits the hypothesis that changes in mitochondrial metabolism are involved, mitochondria and KATP channels thus forming functional microcompartments.

    Notes: In situ hybridization with fluorescently monolabelled rRNA-targeted oligonucleotide probes (17 to 18 nucleotides) was used to discriminate between Alcaligenes eutrophus JMP 134 and Acinetobacter calcoaceticus 69-V by flow cytometry. The strains were grown in batch experiments in a mixed population. The forward light scatter and fluorescence of each bacterial cell were measured with a single laser cytometer. The intensity of fluorescence after rRNA staining depended on the content of ribosomes, which correlated with the growth rate of bacteria. Therefore exponentially growing cells could be clearly detected. For other growth phases, signal amplification was necessary using multiple probes. The two bacterial strains were identified with differently labelled probes under an epifluorescent microscope. Using a single laser cytometer, rRNA based identification was possible nut not ideal. Better discrimination between the two strains of the mixed population was achieved by DNA staining, combined with the different forward light scatter signals. Due to the significantly different cellular DNA and GC content of both strains, the fluorescent dye DAPI (4′, 6-diamidino-2-phenylindole), preferring AT-rich regions of DNA, was found to be a supplementary tool for population analysis. The abundance ratios of the two strains in mixed culture determined by DNA or rRNA staining were similar.
