    Notes: Abstract In a randomized, prospective, multicenter trial, 227 children ranging in age from 3 to 17 years who had tonsillopharyngitis and a throat culture positive for group A beta-hemolytic streptococci (GABHS) were treated with erythromycin estolate (40 mg/kg/d in two divided doses for five days) or penicillin V (30 mg/kg/d in three divided doses for ten days). Clinical signs and symptoms of tonsillopharyngitis were recorded, and throat cultures were obtained before treatment as well as one to three days and six weeks after treatment. Clinical success (cure and improvement) was observed on days 6 to 8 in 100 of 102 (98%) assessable children treated with erythromycin estolate and on days 11 to 13 in 97 of 99 (98%) assessable children treated with penicillin V. Of all patients showing clinical success, 11 were rated as improved, all of whom were treated with erythromycin estolate. There was a trend towards increased use of analgesic treatment in the erythromycin estolate group (41 % vs 33%). On completion of treatment, the rate of eradication of GABHS was 83.3% in the erythromycin estolate group compared with 87.9% in the penicillin V group. The difference is not significant but does not take into account patients excluded because of erythromycin resistance (3.7%). Clinical recurrence was observed in 11 (10.8%) patients treated with erythromycin estolate and in 6 (6.1%) patients treated with penicillin V (non-significant difference). Compliance in the erythromycin estolate group was statistically superior to that in the penicillin V group. The incidence and nature of adverse events were similar in both treatment groups.

    Notes: Abstract The quantity and quality of seeds were studied in 886 shortleaf pine cones collected from 48 trees in each of two 15-ha natural stands in western Arkansas, USA. The number of seeds per cone ranged from 0 to 102, and averaged 34.6 and 38.0 in the two stands. Between 30 and 50 percent of the seeds were sound. Overall mean values for total number and total sound seeds compare with previous work. However, the wide variation between individual trees in a stand needs to be considered when evaluating cone abundance and estimating potential seed yields. For the stands observed in this study, a sample of 65 to 85 cones would be needed to estimate the number of sound seeds per cone (within ±3 seeds) with 95 percent probability. Sampling 1 or 2 cones from each of about 3 trees per ha would provide a reliable estimate of sound seed contents.

    Notes: Abstract Analysis of RNA maturation in ribosome-deficient plastids of four non-allelic barley mutants revealed an increased accumulation and altered processing of transcripts of the ribosomal protein gene CS12 (rps12) compared to normal chloroplasts. The three exons of rps12 are part of two different polycistronic transcription units. Generation of mature rps12-mRNA involves both cis-and trans-splicing. In ribosome-deficient plastids, the cis-intron separating exons 2 and 3 remains entirely unspliced whereas the splicing of the bipartite rps12 trans-intron between exon 1 and exon 2 occurs, but at a reduced level. A comparison of the 3′ and 5′ ends of the two RNAs that are generally assumed to interact during trans-splicing showed a difference in the processing pathways of 3′ rps12 transcripts between mutated and normal chloroplasts. Nonetheless, the final products were identical.

    Notes: Abstract Transcription of plastid genes in higher plants is driven by two RNA polymerases. One is encoded in the chloroplast, the other is encoded in the nucleus. RNA synthesis in ribosome-deficient plastids is performed exclusively by the nuclear-encoded enzyme. In vitro capping was used to identify the transcriptional start sites of the genes clpP and rpl23 in ribosome-free plastids of the barley mutant albostrians. No transcript initiation was found at sequences similar to eubacterial promoters. Instead, transcription started near the motif 5′-YRTA-3′, which is also conserved in mitochondrial promoters of higher plants. Our data suggest that the nuclear encoded RNA polymerase is active in mature chloroplasts and is the sole polymerase involved in transcription of rpl123.
