Notes: Summary  A new tool for the packing of bone dust has been developed. The instrument compresses a mixture of bone dust and saline into suitable cylindrical plugs, fitting neatly into craniotomy burrholes. The skullbone at the craniotomy area will heal neatly also assuring the best healing possibilities for the overlying skin. The cosmetic results are excellent as no skin dents or cavities will be formed over the burrholes.

Notes: Human neutrophils were isolated from cord blood drawn after Caesarean section deliveries without labour and from peripheral blood from healthy adults. The alkaline phosphatase (ALP) activity in the cell populations was compared with the release of respiratory burst products after activation with the chemoattractant n-formyl-methionyl-leucyl-phenylalanine (fMLP). In contrast to cord blood neutrophils obtained from babies after normal vaginal deliveries, the Caesarean section neutrophils were not metabolically primed. However, like the neutrophils from vaginally delivered babies, the Caesarean section neutrophils showed an increased ALP activity compared to adult control neutrophils. These findings indicate that priming of cord blood neutrophils appears to be induced by labour and can be dissociated from increased ALP activity.

Notes: Abstract A model has been developed for calculating the electrical resistivity of commercial aluminium alloys from composition and heat treatments using the Matthiessen rule. The model is based on the approximation that the solubility of the alloying elements in heat treated alloys is equivalent to the equilibrium solubility at a higher temperature. These temperatures were determined from heat treatment data. The resistivity of a wide range of commercial alloys was calculated using the model, showing an agreement with most observed resistivity values of within 3 nΩm, except for alloys with special composition characteristics. According to the model, magnesium and manganese are important contributors to the resistivity for all main groups of alloys. In heat treated alloys the contribution of precipitates is 6–17% of the total resistivity. Thermal conductivity was calculated for alloys given in the literature using the Wiedeman–Franz law and the calculated resistivity. The calculated thermal conductivity agreed with the experimental values for the AlMg-alloys, but it was lower than the experimental values for pure aluminium and the AlCu-alloys in the annealed condition.

Notes: Summary Peak compression of a dihydropyridine drug, clevidipine, is obtained in both analytical and semi-preparative scale supercritical fluid chromatography, resulting in extremely high apparent efficiencies. The observed effect, when utilising a carbon dioxide/2-propanol mobile phase with a bare silica stationary phase, is achieved when the retention of the clevidipine peak is controlled to coalesce with a system peak, generated as a result of having water in the sample. Apparent efficiencies of 350,000 and 170,000 plates meter−1 were obtained when 0.25 and 0.5 mg, respectively, are directly injected to a 200×4.6 mm ID 5 μm Hypersil silica packed column. The effect was extended to a semi-preparative system where apparent efficiencies in the region of 2,000,000 plates meter−1 were observed when 0.3 mg of a clevidipine sample containing 80% water was injected to a 250×10 mm I.D. column containing 5-μm Hypersil silica particles.

Notes: Summary A strategy for the optimisation of direct chiral separation of omeprazole and a metabolite, hydroxi-omeprazole, in reversed phase liquid chromatography is described. A factorial design was used, where mobile phase pH, concentration of a mobile phase modifier, ionic strength and column temperature were tested as the variables and enantioselective retention, column efficiency and asymmetry factor as the responses. The experimental results were evaluated with multivariate analyses, which demonstrated that the column temperature and content of mobile phase acetonitrile were by far the most important variables. The enantiomers of omeprazole and one of its metabolites were baseline resolved within 15 minutes. The optimised chromatographic system was used for a separation of the enantiomers of omeprazole and its main metabolite in a patient plasma sample.

Notes: Summary In this paper chemometrics have been used to study and characterize peak compression phenomena in packed column SFC. A carbon dioxide/2-methyl-1-propanol mobile phase was used in the experimental design (modifier concentration, temperature and pressure) and modelling part of the investigation. A cubic interaction term was needed in the model to obtain a reasonable fit, suggesting that all three parameters are of significance in terms of controlling peak compression. At the optimum conditions derived from the model a narrow peak was obtained as predicted.

Notes: Summary Enantiomeric separation of mosapride and a structurally related compound was performed using chiral chromatography and experimental design. Unique effects of mobile phase pH and column temperature made it possible to control the elution order of the enantiomers when using Chiral-AGP as the solid phase. At a low mobile phase pH (〈6) the (R)-enantiomer of mosapride elutes before the (S)-form whereas the (S)-enantiomer elutes first at a high mobile phase pH (〉6). By using a mobile phase pH around 6, the column temperature could also be used to control the elution order of the enantiomers of mosapride. Similar effects of mobile phase pH and column temperature were obtained for the enantiomers of a structurally related compound, a metabolite (M1). Isocratic chromatographic systems made it possible to determine enantiomeric impurities less than 0.1% in the respective enantiomer of mosapride. The enantiomers of mosapride as well as the enantiomers of M1 could easily be separated simultaneously using Chiral-AGP and a simple gradient elution.

Notes: Abstract The presence of estramustine-binding protein has been suggested to positively correlate with the effect of the cytotoxic drug estramustine, a combination of estradiol and nornitrogen mustard used in the treatment of prostatic carcinoma. This study demonstrates expression of estramustine-binding protein in a series of meningioma using different ligand-based and immunological techniques. Scatchard plot analysis showed specific binding sites for [3H]estramustine in meningioma tissue with a dissociation constant of 22–26 nM. Immunohistochemistry revealed an immunoreactivity in meningioma comparable to that demonstrated in prostatic carcinoma. The mean concentration (n = 6) of estramustine-binding protein in meningioma, as determined by radioimmunoassay was 159 ng/g tissue (range 18–274 ng/g). Moreover, partial characterization using size exclusion chromatography of [3H]estramustine-labeled tumor extracts and Western blot analysis of immunoprecipitated samples indicated that the structure of the estramustine-binding protein in meningioma is similar to that in rat prostate, with three polypeptide components of 10, 14, and 16 kDa, as compared to 8, 10–11, and 12 kDa in rat prostate. In conclusion, the novel observation of estramustine-binding protein and specific binding of estramustine in meningioma justify further evaluation regarding the role of estramustine-binding protein in the growth behavior of meningioma and the potential for estramustine and similar hormone-related drugs in the treatment of relapsing meningioma.

Notes: Abstract The potential for biological transformation of 23 xenobiotic compounds by microorganisms in municipal solid waste (MSW) samples from a laboratory scale landfill reactor was studied. In addition the influence of these xenobiotic compounds on methanogenesis was investigated. All R11, 1,1 dichloroethylene, 2,4,6 trichlorophenol, dimethyl phthalate, phenol, benzoate and phthalic acid added were completely transformed during the period of incubation (〉 100 days). Parts of the initially added perchloroethylene, trichloroethylene, R12, R114, diethyl phthalate, dibutyl phthalate and benzylbutyl phthalate were transformed. Methanogenesis from acetate was completely inhibited in the presence of 2,5 dichlorophenol, whereas 2,4,6 trichlorophenol and R11 showed an initial inhibition, whenafter methane formation recovered. No transformation or effect on the anaerobic microflora occurred for R13, R22, R114, 3 chlorobenzoate, 2,4,6 trichlorobenzoate, bis(2 ethyl)hexyl phthalate, diisodecyl phthalate and dinonyl phthalate. The results indicate a limited potential for degradation, of the compounds tested, by microorganisms developing in a methanogenic landfill environment as compared with other anaerobic habitats such as sewage digestor sludge and sediments.
