Notizen: Fusarium head blight (FHB), primarily caused by Fusarium graminearum in North America can result in significant losses in the yield and quality of wheat (Triticum aestivum L). Resistance sources have been largely limited to Chinese germplasm and, in particular, Sumai 3 or its derivatives. In recent years, resistance has been identified in Europe. Previous studies using the wheat line ‘Bizel’, developed in France, have shown that it has resistance to Fusarium head blight. Pedigree information shows that one of its progenitors is rye. This experiment was conducted to determine if ‘Bizel’ has rye chromatin, with the goal of developing a strategy for mapping FHB resistance genes. Two methods based on repetitive DNA sequences specific to rye were implemented. With both approaches, it was demonstrated that ‘Bizel’ does not contain rye chromatin. Consequently, wheat SSRs can be used to map ‘Bizel’ resistance genes for FHB.

Notizen: Summary A DNA marker detection strategy that allows the rapid, efficient resolution of high levels of polymorphism among closely related lines of common wheat (Triticum aestivum) has been developed to circumvent the apparent lack of restriction fragment length polymorphism in many important self-pollinated crop species. The technique of randomly amplified polymorphic DNA (RAPD) was combined with a denaturing gradient gel electrophoresis system (DGGE) to explore DNA sequence polymorphisms among different genotypes of wheat. Of the 65 primer combinations used for the polymerase chain reaction (PCR) amplifications, over 38% of them produced readily detectable and reproducible DNA polymorphisms between a spring wheat line, SO852, and a winter wheat variety, ‘Clark’. A high level of polymorphism was observed among a number of commercial varieties and breeding lines of wheat. This procedure was also used to detect polymorphisms in a recombinant inbred population to test the feasibility of its application in genome mapping. This DNA polymorphism detection system provides an opportunity for pedigree analysis and fingerprinting of developed wheat lines as well as construction of a high density genetic map of wheat. Without the need for 32P and sophisticated DNA extraction procedures, this approach should make it feasible to utilize marker-based selection in a plant breeding program.

Notizen: Abstract The Hessian fly [Mayetiola destructor (Say)] is a major pest of wheat (Triticum aestivum L.) and genetic resistance has been used effectively over the past 30 years to protect wheat against serious damage by the fly. To-date, 25 Hessian fly resistance genes, designated H1 to H25, have been identified in wheat. With near-isogenic wheat lines differing for the presence of an individual Hessian fly resistance gene, in conjunction with random amplified polymorphic DNA (RAPD) analysis and denaturing gradient-gel electrophoresis (DGGE), we have identified a DNA marker associated with the H9 resistance gene. The H9 gene confers resistance against biotype L of the Hessian fly, the most virulent biotype. The RAPD marker cosegregates with resistance in a segregating F2 population, remains associated with H9 resistance in a number of different T. aestivum and T. durum L. genetic backgrounds, and is readily detected by either DGGE or DNA gel-blot hybridization.

Notizen: Abstract  To study the inheritance of resistance in wheat to Fusarium graminearum, six resistant cultivars from China were crossed to two susceptible cultivars. The parents and their progenies were evaluated in the greenhouse for resistance to the spread of scab within a spike. A central floret was inoculated by injecting a droplet of inoculum at the time of anthesis. Inoculated plants were kept in a moist chamber for three subsequent nights. The proportion of scabbed spikelets was recorded six-times from 3-days to 21-days after inoculation, and the area under the disease progress curve (AUDPC) was calculated from these proportions. One to three genes, depending on the cultivar, conditioned resistance to scab as reflected by the AUDPC. A simple additive-dominance effect model fitted the segregation data for 8 of the 11 crosses. Dominance and epistatic effects were significant in a few crosses. These effects increased resistance in some crosses but decreased resistance in others. However, relative to additive effects, dominant and epistatic effects accounted for only a small portion of the genetic effects in the populations evaluated. The importance of additive effects means that it should be possible to accumulate different genes to enhance resistance to scab in wheat.

Notizen: Abstract A band with a rotational pattern based on a state at 585.1 keV has been identified in theN=59 neutron-rich nucleus97Sr. Its properties lead to the [422] 3/2 Nilsson-orbital assignment for the band head. There is evidence for a second band with the head at 644.7 keV and the configuration [541] 3/2. Since the ground state and the lowest excited levels are spherical, shape coexistence is established for97Sr. A deformed nature of several levels at 500–600 keV results also from QRPA-model calculations. The structure of the low-lying spherical levels has been studied in the frame of the IBF model. The results of the present investigations lead to a better understanding of theN=59 isotones which constitute the link between the spherical and deformed nuclei atA∼100 as a species with shape coexistence but without any indications of particular softness.

Notizen: Abstract The lifetimes of the three lowest excited members of the rotational ground-state band of103Mo at 102.6, 241.1 and 433.2 keV as well as of the levels at 353.8 and 456.1 keV have been determined through a measurement of the delayed coincidences betweenβ particles from the decay of the parent103Nb and theγ rays which depopulate the levels. A plastic and a BaF2 detector have been used for theβ andγ rays, respectively, in these experiments at the fission product separator JOSEF. The results for all three band members evince that this nucleus has a sizeable deformation, with a value ofβ q=0.34(1). This value is larger than that of the neighbouring even-even Mo isotopes which indicates an odd-even effect. On the other hand it confirms the observation that the Mo nuclei are in general less strongly deformed than their Sr and Zr isotones. Particle-rotor model calculations with the Nilsson parametersκ=0.084,μ=0.28 provide a good description of the ground-state band and of a side band based on the 346.5 keV level, including the new lifetimes. This substantiates the Nilsson orbital assignments of [411]3/2+ and [532]5/2− for these two bands, respectively, where the sideband is strongly affected by a [541]3/2− admixture. There is evidence for an additional band based on the 641.1 keV level for which the assignment [422]3/2+ is proposed.

Notizen: Abstract The γ radiation from the ß− decay of the high-spin isomer in98Y has been studied at the separator JOSEF. A main goal was to get insight into the spins of levels in98Zr which have been interpreted as members of an intruder band based on the 02 + state at 854 keV. The measuredγ-γ angular-correlations lead to the assignment of spins of 4 and 3 ℏ to the 2491 and 1844 keV levels, respectively, and contradict their interpretation as the 6+ and 4+ states of the proposed band. For the 2.0s isomer of98Y, the ß{−}-decay properties suggest I=5 with a major configuration [πg9/2,νs1/2(g2 7/2)0].

Notizen: Abstract Tie half-lives of 8 low lying levels of103Nb have been determined at the fission-product separator JOSEF. A B-γ-γ triple-coincidence method was used which consists in a measurement of the time delay between the feeding of the levels through the B− decay of103Zr and their γ decay, in coincidence with a tagging γ ray, with plastic, BaF2 and Ge detectors, respectively. Most of the investigated levels are members of the three known rotational bands based on the ground state and the 164 and 248 keV levels. The deformation Bq=0.31(3) of103Nb could be deduced from the half-life data. The half-lives are well reproduced through calculations in the frame of the Nilsson model.

Notizen: Abstract The half-lives of low-lying levels of the neutron-rich isotopes102Mo and104Mo have been measured at the fission product separator Josef with the ß-γ-γ triple coincidence technique. Values of t1/2= 126(4)ps and 0.72(4)ns have been obtained for the 21 + levels at 296 keV and 192 keV in102Mo and104Mo, respectively. Deformation parameters of ßq=0.28(1) and 0.31(1), respectively, are deduced, which are smaller than those of the isotones of Sr and Zr. The interacting boson model accounts well for the trend of 2+ energies in the Mo isotopes but slightly overpredicts the B(E2) values at saturation. A smooth dependence of τ 21 + vs. E 21 + is found for the A ∼ 100 region in agreement with the hydrodynamical model.

Notizen: Abstract: The lifetime of the Λ-hyperon for the nonmesonic decay ΛN → NN has been determined by a measurement at COSY Jülich of the delayed fission of heavy hypernuclei produced in proton-Au collisions at T p = 1.9 GeV. It is found that heavy hypernuclei with mass numbers A≈ (180±5) and atomic numbers Z≈ (74±2) fission with a lifetime τΛ = 130±13 (stat.) ±15 (syst.) ps. This value together with the results obtained for other heavy hypernuclei in previous investigations indicates --on the confidence level of 0.9-- a violation of the phenomenological ΔI = 1/2 rule for the ΛN → NN transitions as known from the weak mesonic decays of kaons and hyperons.
