Notes: Summary We report two patients who developed benign plasmacytosis with multiple skin lesions. The cases were characterized by hyperplasia of mature plasma cells, and polyclonal hypergammaglobulinaemia. One patient had hyperplasia of mature plasma cells not only in the skin, but also extensively in lymph nodes and the retroperitoneal areas around the ureters. The other had plasma cell hyperplasia limited to the skin. Extensive investigations failed to reveal any clinical or laboratory evidence suggesting the presence of any underlying disease accompanying the hypergammaglobulinaemia and/or plasma cell proliferation, such as chronic infectious disease, collagen disease or other chronic inflammatory disorder. Clinically and histologically, the first patient showed features compatible with a diagnosis of systemic plasmacytosis and the second with a diagnosis of cutaneous plasmacytosis. Significant serum interleukin-6 (IL-6) levels were detected in both patients, suggesting that IL-6 may be involved in the pathogenesis of these conditions.

Description / Table of Contents: Summary The incorporation of H-3-thymidine and H-3-uridine into the cells of a guinea pig's Reissner's membrane by intralabyrinthine and by intraperitoneal administration was studied with autoradiography. 1. Even after 24 h exposure to isotopes, the incorporation of H-3-thymidine could not be detected in any part of Reissner's membrane. 2. The epithelial cells on the side of the scala media displayed a higher up-take of H-3-uridine than the mesothelial cells on the side of the scala vestibuli. 3. The epithelial cells on the inner side of Reissner's membrane showed a higher up-take of H-3-uridine in comparison with those on the outer side of it. 4. The area of the epithelial cells on the inner side of Reissner's membrane showing a higher up-take of H-3-uridine, was narrower in the base becoming wider towards the apex. This may lead to the assumption that the epithelial cells of Reissner's membrane of the lower coils will reach functional maturity sooner than those of the upper ones. 5. Reissner's membrane is a very good material for autoradiography.

Notes: Abstract Some properties of salivary amylases of the adults of Adelphocoris suturalis Jakovlev and Dolycoris baccarum L. were studied in vitro and compared with those of several other heteropteran species. The reducing sugar produced by the action of the salivary amylase of D. baccarum increased in proportion to substrate concentration while the concentration was relatively low (below 0.67% in its final concentration). Its increase stopped at the concentrations from 0.67 to 2.0%, and then it increased again constantly but slowly. The optimum temperature for the action of the enzyme was found to be about 50° (in A. suturalis) and about 40° (in D. baccarum), and the optimal pH was 3.5–4.0 in A. suturalis and 6.0 in D. baccarum. The salivary amylase activity of D. baccarum was scarcely affected by NaCl, KNO3, and other compounds, while the salivary amylase of A. suturalis was strongly activated by NaCl and moderately by KNO3. It seems highly probable that taxonomically related heteropterous insects might well be grouped with respect to the degree of activation of the salivary amylase by Cl- or NO3-.

Notes: Summary 3Y1 rat fibroblasts transformed by adenovirus type 12 or by its E1A gene formed syncytia when cocultivated with the Friend as well as other ecotropic murine leukemia-virus-producing cells. In contrast, the parental 3Y1 cells and those transformed by other tumor viruses or by a chemical carcinogen exhibited no syncytium formation.

Notes: Summary The distribution of125I-labelled recombinant mouse interferon-β (rMuIFN-β) in normal and glioma (203 glioma) bearing mice was studied by radioassay and macro-autoradiography at 15 and 30 min after a single intravenous injection. The level of rMuIFN-β in the spleen was about 20-fold higher than in serum. Concentrations higher than the serum level was detected in the lung, liver and kidney. The concentration of rMuIFN-β in the brain was 8% of the serum level and the concentration in the glioma 30 min after administration was about 10-fold higher than in normal mouse brain. Macro-autoradiographic study demonstrated a wide distribution range and selective uptake in glioma tissue. Furthermore, we found that mouse gliomas were sensitive to mouse IFN-β. Our findings demonstrate that in the mouse glioma model, intravenously administered interferon reaches the tumour.

Notes: Summary In order to investigate the developmental mechanism of cerebral aneurysms, the in vivo flow pattern around human cervical carotid bifurcations was studied by flow visualization using digital subtraction angiography with an isotonic contrast medium. The blood stream containing the medium impinged on the apex, then proceeded along the walls of the branches. After opacification of the whole lumen around the apex, most of the medium was carried away, while some remained for a few seconds at the carotid sinus. In the internal carotid artery, the blood struck the wall at an oblique angle near the tops of the arterial curvatures. In cases with atheromatous plaque or kinking of the branch, the blood passed through the stenosed segment and moved upstream, indicating turbulence. The study suggests that haemodynamic forces around the apex consist mainly of impingement on the apex and shear stress to the wall at and around the apex. In branches, high shear stress seems to exist. It might be possible that high shear stress causes degenerative changes in the endothelial layer, initiating the formation of saccular and fusiform cerebral aneurysms.

Notes: Summary The effects of local and systemic administration of tumour necrosis factor (TNF) on the mouse-tail model of psoriasis were examined histologically at 4 days after administration. Subcutaneous injection of TNF at 1×101 to 1×104 units/mouse resulted in dose-dependent formation of a granular layer in the tail scales, which was quantified as the percentage of tail scales having granular layer formations extending over half or more of their surface and was larger than that occurring in untreated mice and in mice receiving injections of vehicle solution alone. Daily intraperitoneal injection of TNF at 1×103 or 1×104 units/mouse for 28 days resulted in similar granular layer formation and in a significantly reduced mitotic index of the tail keratinocytes, indicating an antiproliferative effect of TNF. The lowered mitotic index and the induction of granular layers, apparently attributable to a modulation of keratinocyte proliferation and differentiation by TNF, suggest that TNF may be effective as a drug for treatment of psoriasis.
