    Notes: Stimulation of [3H]inositol monophosphate 〈[3H]InsP) formation by ibotenate or trans-1-aminocyclopentyl-l, 3-dicarboxylic acid (t-ACPD) in rat hippocampal slices was enhanced after tetanic stimulation of die Schaffer collaterals projecting to the CA1 region (in vitro) or the perforant pathway projecting to the dentate gyms (in freely moving animals). This effect was observed 5 h (but not 2 h) after long-term potentiation (LTP) induction and was abolished if tetanic stimulation was performed in the presence of specific antagonists of N-methyl-D-aspartate receptors. The delayed increase in excitatory amino acid-induced polyphosphoinositide (PPI) hydrolysis was accompanied by an enhanced responsiveness to norepinephrine, whereas the basal and carbamylcholine-stimulated [3H]InsP formation were unchanged. These results suggest that an increased activity of „metabotropic” glutamate receptors may contribute to the synaptic mechanisms enabling the late expression and or maintenance of LTP. Accordingly, LTP decayed more rapidly (within 5 h) in rats repeatedly injected with LiCl (60–120 mg/kg, i.p., for 10 days), a treatment that led to a reduced efficacy of ibotenate and norepinephrine in stimulating PPI hydrolysis in hippocampal slices.

    Notes: The spontaneous muscular activity of the masseter and two non-masticatory muscles, and their activity under conditions of noise and flickering light, were recorded in 30 patients with mandibular dysfunction, and in 25 control subjects. The mood of the subjects during the investigation, and the possible presence of emotional disturbances was evaluated by means of questionnaires. The enhanced muscular activity of the masseter muscle differentiated patients from the control group, although it does not constitute a pathognomic sign. Unilaterality of complaints, and dysfunctions of differing intensity were not reflected in differential EMG findings. One-fifty of the patients exhibited neck muscle activity occurring simultaneously with the masseter activity. The effect of the applied stimuli on the muscular activity was not homogeneous, leading to activation in some cases and inhibition in others. Symptoms of anxiety could be observed in seven TMJ patients. Anxious patients showed higher levels of muscular activity and emotional irritability during the experiment than non-anxious patients.

    Notes: Factor B of human complement is encoded within the Major Histocompatibility Complex (MHC) and is polymorphic, with up to 30 alleles defined by electrophoretic mobility. One of the most common alleles, BF*F, is subdivided into the FA and FB subtypes, which differ at the gene level by non-synonymous base substitutions in the seventh codon. We have found at this position a new restriction site polymorphism, as a Bsl I site absent from the FB allele. Using this restriction polymorphism, we have developed a method for BF F subtype determination, based on amplification by polymerase chain reaction of the 5’ end of the BF gene, and digestion with Bsl I. This new method has been applied to a panel of 29 selected BF F individuals. A single strand DNA conformation analysis of the same region of the gene allowed us to confirm the above DNA-based BF F subtyping. During this study, two BF*F1 alleles showed discrepancies between protein and DNA typing, which were confirmed by our sequencing data. These were identical, in the 5’ region, to BF*S and BF*FB genes, respectively. In a comparison with two protein subtyping methods, identical results were found for only one third of the selected samples. The conflicting results may arise, in part, from previously undescribed molecular heterogeneity within BF F subtypes, or from the presence of a null allele. Our new method allows RF*F subtyping to be used with confidence in the definition of disease-associated MHC haplotypes.

    Notes: Twenty-seven cases of the hyaline vascular variant and 10 cases of the plasma cell variant of Castleman's disease were studied with the paraffin resistant monoclonal antibodies Ki-FDC1p and/or Ki-M4p against follicular dendritic cells. Studies with the monoclonal antibody Ki-M9, for the detection of sinus lining cells, were also performed on the available frozen tissue in four cases of the hyaline vascular variant. In nine of the 10 plasma cell variant cases, the predominant type of follicular dendritic cell network was similar to that seen in normal or reactive germinal centres. In contrast, the hyaline vascular variant demonstrated either an expanded, disrupted, follicular dendritic cell network (10 cases) or multiple tight collections of follicular dendritic cells (16 cases). Sinus lining cells were not detected in the four cases studied. The difference in the predominant type of dendritic meshwork is an additional distinguishing feature to separate the plasma cell and hyaline vascular variants of Castleman's disease. The patterns of dendritic network seen in the hyaline vascular type, together with the absence of sinus lining cells, appear to favour the hamartoma theory proposed for this variant.

    Notes: New technology coupled with a better understanding of tissue biology has played a key role in restoring the somewhat tarnished image of implant dentistry, Thanks to carefully conducted longitudinal studies, it has now become clear that the replacement of missing teeth by artificial implants integrated into the living tissues of the jaws is a predictable procedure, as long as certain guidelines are followed in the manufacture of the implant, its placement, its eventual functional loading, and its maintenance. Yet, functional success for a natural tooth is dependent on a number of anatomic, physiologic, and environmental considerations which differ in some important respects from those that insure the functional success of an implant. In this paper, we review the anatomic features of the natural dentition with emphasis on the periodontal tissues, and contrast these characteristics with those that exist around well-integrated endosseous implants.

    Notes: Abstract The siliceous spongeGeodia cydonium Jameson was used to study the influence of pollution in marine environments on selected parameters of the intracellular signal transduction pathway. The parameters chosen were: intracellular distribution of protein kinase C (PK-C),ras-gene expression and DNA polymerasealpha (DNA Polalpha) activity. Both PK-C andras-gene product (ras-protein) have previously been established to be key molecules in the intracellular signalling pathway in sponges; increased level ofras-protein mediates events following sponge cell-cell contact. Three unpolluted and three polluted sites in the off-shore seawater around Rovinj (Yugoslavia) were selected for the study in 1989. The state of pollution of these sites has been well-defined in a series of previous studies (1976 to 1989). Transplantation of regenerating sponge cubes ofG. cydonium to the polluted sites resulted in pronounced changes in the parameters chosen, compared to controls exposed to unpolluted environments. Expression ofras gene was increased by three- to five-fold after exposure of regenerating sponge tissue to the impacted sites. At the site with the highest pollutional load,ras mRNA level was about 50% of that at the reference sites. In parallel experiments it was established that, in response to pollution, a translocation of PK-C from the cytosolic to membrane fraction occurred. At the most impacted site, most of the enzyme activity was cytosolic. DNA Polalpha-activity, as a measure of sponge cell proliferation, decreased in a pollutioncorrelated manner. Our results indicate that the intracellular signalling system within sponge cells is activated in response to moderate pollution but is depressed in heavily polluted environments.

    Notes: Summary In an attempt to establish maturational alterations in the morphology of the articular tissue layer, mandibular condyles of four immature and four mature male monkeys (Macaca fascicularis) were studied using light microscopy as well as scanning and transmission electron microscopy. Specimens were fixed in situ by perfusion in the presence of ruthenium red to stabilize proteoglycans. Preparations intended for observation in the scanning electron microscope were first dehydrated and sputtered for the examination of articular surfaces, and afterwards treated with trypsin to expose the spatial arrangement of collagen fibrils. Gross anatomical relations between joint components indicated that the anterior and central, but not the posterior region of the condylar articular surface can be subject to compressional load. Load-bearing and non-load-bearing regions differed with respect to the morphology of the articular layer. Load-bearing surfaces were covered by a prominent articular surface lamina similar to that observed on articular cartilage. This lamina seemed to constitute an integral part of the articular layer, distinct from the lining of synovial fluid, and to be composed largely of proteoglycans. It was unaffected by maturation. The subjecent, load-bearing articular layer differed markedly in structure, both from articular cartilage, and between immature and mature animals. Articular cells of immature animals were classified as fibroblastlike, but unlike typical fibroblasts, were surrounded by a thin, often incomplete halo of fibril-free pericellular matrix, presumably consisting of proteoglycans. In mature animals, articular cells closely resembled chondrocytes, but exhibited prominent nuclear fibrous laminae, which usually are found only in fibroblasts. Thus, the load-bearing part of the articular layer seems to undergo a maturation-dependent metaplastic conversion, from a dense connective tissue with some features of fibrocartilage, to a fibrocartilage-like tissue containing chrondrocytelike cells with some features of fibroblasts. This conversion might reflect an adaptation to a maturation-associated increase in articular stress.
