    Notes: Summary Lactate production by immobilized Lactobacillus casei has been studied. The cells were immobilized in alginate and the effect of variations in different parameters on product formation and productivity was investigated. The performance of the reaction was evaluated in stirred batch as well as in packed-bed conditions. pH control was a problem in the packed-bed reactor. In stirred batch experiments, nearly total glucose utilization was observed with a lactate yield of 90–99% and a total productivity of 1.6 g·l−1·h−1. Under standard conditions only a low percentage (3–4%) of the total lactate formed was the abetd-isomer. When immobilized cells were reused, increased formation of abetd-lactate took place, especially when the cell conditions were sub-optimal. After revitalization by exposure to growth nutrients the balance was restored.

    Notes: Summary The cells of Lactobacillus casei were adsorbed to Poraver, foam glass particles pretreated with polyethylenimine (PEI). Exposure of cells for a relatively short period to Poraver beads coated with a high concentration of PEI resulted in maximal adsorption with good retention of metabolic activity. The immobilized cells were tested in packed-bed and stirred-tank reactors for lactic acid production. Stirred-tank operations were more effective in terms of productivity but the support was sensitive to attrition. The beads exhibited good mechanical stability to withstand pressure in the packed-bed reactor.

    Notes: Summary The potential of aqueous two-phase systems for the estimation of molecular weights of proteins and enzyme-inhibitor complexes has been demonstrated. The method is simple and rapid, and employs extremely small amounts of the test sample. It is based on the measurement of the phase transition behaviour on dilution of the system as a function of the molecular weight of the molecule or complex under study.

    Notes: Summary A property characteristic of a large number of enzymes is their low shelf stability at ambient temperatures. The denaturation of these biomolecules is promoted by processes facilitated by water. A study has been carried out on the storage of a few enzymes stored in three different dry states: lyophilized in air in a sealed vessel, in dry heptane (predried over molecular sieves) or in dry heptane in the presence of molecular sieves. Pronounced improvements in storage stability at elevated temperatures were found when the enzymes were stored under strictly anhydrous conditions (i.e. in the presence of molecular sieves) as compared to when the enzyme was stored in what is regarded as a dry state.

    Notes: Summary Endo-xylanase from T. viride has been purified 4.2 fold by precipitation with a commercially available enteric polymer Eudragit S-100. Electrophoretic analysis also indicated removal of contaminant proteins. The enzyme could be recovered in more than 89% yield. The binding of the enzyme to the polymer was predominantly by electrostatic interaction.

    Notes: Aqueous two-phase extraction incorporated affinity precipitation was examined as a technique for protein purification. An enteric coating polymer, Eudragit S100, was employed as a ligand carrier. Eudragit was specifically partitioned to the top phase in the aqueous two-phase systems. For application of this method to purification of recombinant protein A using human IgG coupled to Eudragit in an aqueous two-phase system, 80% of protein A added was recovered with 81% purity. The purity was enhanced 26-fold by thid method. The IgG-Eudragit could be used repeatedly for the purification process. This seperation method should be applicable to industrial-scale purification as a new purification procedure combining the advantages and compensating for the disadvantages of the aqueous two-phase method and affinity precipitation method. © 1992 John Wiley & Sons, Inc.
