    Notes: X-ray Laue diffraction patterns with an exposure time of 120 ps have been obtained from single crystals of an indole alkaloid and of the enzyme lysozyme using the X-rays emitted as a single bunch of electrons traverses a hard X-ray undulator inserted in CESR, the Cornell Electron–Positron Storage Ring. The patterns were recorded on a sensitive storage-phosphor detector. Despite complexities posed by the sharp variation of the incident X-ray spectrum with wavelength and the weakness of the diffraction patterns, accurate crystallographic structure amplitudes were extracted from the Laue intensities by the generalized scale-factor approach to the determination of the wavelength- and position-dependent correction factors. The results show that crystallography is feasible on the 100 ps time scale and open up the prospect of time-resolved measurements of ultra-rapid changes in molecular structure.

    Notes: Summary Cyclosporin A (CsA) pharmacokinetics was studied in 19 diabetic children (mean age: 10.6 y). They were divided into prepubertal (I) and pubertal (II) groups according to plasma oestradiol or testosterone concentrations. The kinetic study was performed after a 72 h wash out period and a single oral dose of 7.5 mg/kg CsA. CsA in blood was measured by HPLC. The kinetic parameters: Cmax, tmax, t1/2, AUC, CL/f, Vz/f and tss were calculated. No significant difference was found between the two groups. A significant negative correlation was found between Vz and both total cholesterol (r=0.46), VLDL+LDL−cholesterol (r=−0.49) and VLDL+LDL−phospholipids (r=−0.58). CsA kinetics at steady-state were simulated by superimposition of single dose kinetics derived from each single dose. Measured steady-state blood concentrations were correlated (r=0.80) with the values predicted by the simulation. The results suggest that CsA adjustment dosage of the CsA may be performed after a single oral dose using blood levels measured by HPLC. This procedure requires validation in further studies.

    Notes: Summary A comparison of newly synthesized, reversed-phase bonded stationary phases is presented. The materials studied include monomeric n-octyl, n-octadecyl, aryl and n-octyldihydrogeno bonded-phases. Differences in reactivity of the silane functional groups are demonstrated and the high coverage obtained with the hydrogeno-bonded phases is shown. The structures of the bonded phases have been determined by29Si and13C CP/MAS NMR spectroscopy.

    Notes: Proton spin-spin relaxation time measurements have been performed at 300 and 400 MHz on bound rubber-filler mixes resulting from the solvent extraction of silica-filled and silicafilled-modified polybutadiene, on the one hand, and of carbon black-filled and carbon blackfilled-modified styrene butadiene rubber, on the other hand. Two relaxation times are observed for all samples. They are ascribed to the occurrence of tightly and loosely bound rubber components, respectively. The effect of the grafting modification of the fillers on the formation of bound rubber and its two components is discussed. © 1992 John Wiley & Sons, Inc.

    Notes: Summary Cu(II) complexes of poly(1-vinylimidazole) immobilized on silica particles have been used as stationary phases for immobilized metal affinity chromatography of some proteins. Chromatographic data are reported which suggest that protein retention depends on the ionic strength in the column before and during loading with Cu(II). It has been shown by EPR spectroscopy that [Cu(Im)6]2+-type complexes were predominant when Cu(II) binding was performed in pure water. When performed under conditions of greater ionic strength, lower complexes are probably also formed. Protein retention has been shown to depend on other factors, such as the amount of Cu(II) bound to the column and the composition of the mobile phase.
