    Notes: Summary A region of 50 kb around the human PTH gene was cloned and mapped by restriction analysis. Sequence analysis was performed and 3270bp determined, completing the sequence of the gene. The nucleotide sequence was analysed with regard to homology between human, bovine and rat PTH genes, and various potential cis-acting regulatory elements were identified. The gene region lacks an obvious CpG island. The PTH gene region in patients suffering from (pseudo)-hypoparathyroidism was investigated by Southern blotting. No detectable alteration in the fragment patterns was observed. Results of segregation analysis in families with affected individuals was inconclusive.

    Notes: Abstract Lipopolysaccharides were isolated from the moderate halophilic Ectothiorhodospira shaposhnikovii slight to and Ectothiorhodospira mobilis and from the extremely halophilic Ectothiorhodospira halophila by the hot phenol-water and purified by the phenol-chloroform-petroleum ether methods. The isolated lipopolysaccharides of all three species contained 3-deoxy-d-manno-octulosonic acid and d-glycero-d-mannoheptose indicating the existence of a core. They contained additionally glucose and uronic acids (E. shaposhnikovii and E. mobilis) or glucose, uronic acids and threonine (E. halophila). Sodium deoxycholate gel-electrophoresis of the three lipopolysaccharides, each showing only one major band, indicated R-type character of the lipopolysaccharides of the three Ectothiorhodospira species. The lipid A fractions of the lipopolysaccharides from E. shaposhnikovii and E. mobilis represented phosphorylated “mixed” lipid A types with both 2,3-diamino-2,3-dideoxy-d-glucose and d-glucosamine. The lipid A from E. halophila contained also phosphate and 2,3-diamino-2,3-dideoxy-d-glucose but only traces of d-glucosamine, which would indicated lipid ADAG. The fatty acid spectra were characterized by amide-bound 3-OH-10:0 and 3-OH-12:0 (E. shaposhnikovii), 3-OH-10:0 (E. mobilis), or 3-OH-10:0,3-OH-14:0, and 3-oxo-14-0 (E. halophila). The predominant ester-bound fatty acids were 14:0 and 16:0 (E. shaposhnikovii and E. mobilis), or 12:0 and 14:1 (E. halophila).

    Notes: Summary A group of 100 patients submitted to microsurgical treatment for herniated lumbar disc following unsuccessful chemonucleolysis with chymopapain were retrospectively compared to a statistically comparable group of patients primarily submitted to microsurgery. This comparison demonstrated that previous unsuccessful chemonucleolysis has no influence on either the short-term or the long-term results of subsequent microsurgery.

    Notes: Abstract Exposure of dry oxic paddy sol to submerged anoxic conditions resulted in a decrease of the soil redox potential and initiation of CH4 production. However, the most probable numbers (MPN) of hydrogenotrophic and acetotrophic methanogens did not change upon flooding the soil and upon initiation of CH4 production, and stayed constant at 105−106 and 104−105 g−1 d.w. soil, respectively. Cells of Methanobacterium strain Bab1 added to anoxic soil suspensions mostly survived drying and storage under an air atmosphere. Initiation of CH4 production was not affected by storage of dry soil under N2, air, or O2 atmosphere. However, it was negatively affected by storage under the air at high negatively affected by storage under air at high relative humidities, under moist conditions, or by using only the small (〈 100 μ m) soil particle fraction. All these treatments also resulted in decreased contents of dissolved organic carbon (DOC). The soil's capacity for initiation of CH4 production could be regenerated either by addition of glucose or by exposure to H2 and addition of reducing agents. The establishment of decreased redox potentials and the provision of methanogenic substrates by heterotrophic bacteria, but not the number of methanogenic bacteria, seemed to be the essential factors for initiation of CH4 production after submergence of oxic paddy soil. CH4 production could also be initiated in forest and arable soils. Here, however, the population of methanogenic bacteria (MPN) increased from about 103 to 105−106 g−1 d.w. when methanogenesis started.

    Notes: Abstract A 3-deoxy-2-heptulosaric acid (DHA), very probably with the lyxo-configuration, was identified in the R-core region of lipopolysaccharides from modulating strains of Rhizobium leguminosarum, Rhizobium meliloti and from all three biovars of the phytopathogenic Agrobacterium tumefaciens. Its structure could be deduced from the fragmentation pattern of the corresponding alditol acetates obtained after reduction of the 2-keto and the 1,7-carboxy groups by sodium borohydride or sodium borodeuteride. DHA in lipopolysaccharide was not destroyed by periodate and is therefore not in a terminal position. Two DHA-containing oligosaccharides, namely glucosyl (1 → 4)-3-deoxy-2-heptulosaric acid and rhamnosyl-rhamnosyl-(1 → 5)-3-deoxy-2-heptulosaric acid could be tentatively identified by mass spectrometric methods amongst the products of mild acidic hydrolysis of lipopolysaccharides of Rhizobium leguminosarum strain 24. The two types of non-nodulating mutants of Rhizobium leguminosarum included in this study did not contain 3-deoxy-2-heptulosaric acid.

    Notes: Kefirs were produced under laboratory conditions using 10 different kefir grain cultures from different households, and reconstituted cow, ewe, goat and mare milk as substrates. The base milks and corresponding kefirs were examined for their content of water-soluble vitamins and orotic acid. On average, enrichment of vitamin concentration of〉20% was observed with thiamine (only ewe milk kefirs), pyridoxine (kefirs from all milk species except cow milk), and folic acid (kefirs from all milk species except mare milk). Orotic acid content was reduced during fermentation throughout.

    Notes: The characteristics of a compact space and wavelength-resolving spectrometer-detector arrangement are described which is used to measure spectral line shapes simultaneously along several viewing chords. The system is based upon a freely programmable slow-scan charge-coupled device (CCD) camera gated by an image intensifier tube. Visible and near-UV light emitted near the plasma boundary is collected from up to 12 viewing chords and transmitted to the spectrograph by quartz fibers. Spectra can be recorded with a temporal resolution of 20 ms by means of multiple pixel binning during CCD readout. Integration of the spectral line profiles yield particle fluxes of various low-Z species for ohmically heated helium discharges. The calibration, sensitivity, and signal-to-noise ratio of the system are reconstructed considering each individual element, allowing an easy prediction of the performance of similar applications.

    Notes: The development and the test results of an electron-beam furnace for the later utilization in a microgravitational environment are reported. By just varying the deflection pattern by means of the electron-optical components two reference profiles, a gradient profile with a maximum slope of 220 K/cm, and a hot zone profile with a zone temperature of 1520 K could be established and maintained. A beam power of 550 W had to be applied to a sample made of massive Ta for the gradient profile, for creating a hot zone profile an input power of only 250 W onto a sample with a ceramic core was sufficient. A continuous pyrometric measurement system with a high local and time resolution has been realized. By temperature sensing of the sample with this system an intrinsic feature of electron-beam heating could be directly observed, the sharply localized energy deposition at the sample surface.

    Notes: Abstract— The significance of interaction effects has been studied in tests with a continuous repetition of blocks with low and high amplitude cycles under conditions characteristic for ultrasound fatigue. Significant retardation effects were observed. However, low-intensity cycles, which according to a previous investigation might be harmless, did contribute to crack growth. Delayed retardation explains this observation. A fractographic analysis indicated the significance of surface roughness. Low-amplitude cycles may induce a smoother fracture surface, and thus increase the crack growth rate at higher amplitudes. Accurate predictions are therefore problematic. For similar reasons, omitting numerous low-amplitude cycles in random load experiments should be done with great care.

    Notes: Rough fracture surfaces usually influence substantially the fatigue growth properties of materials in the regime of low growth rates. Friction, abrasion, interlocking of fracture surface asperites and fretting debris reduce the applied load amplitude to a smaller effective value at the crack tip (“sliding crack closure”, or “crack surface interaction” or “crack surface interference”). The influence of these phenomena on the fatigue crack growth properties of structural steel is discussed and compared for the two kinds of mixed mode loading employed in this work. Mixed mode loading was performed by (A): cyclic mode III + superimposed static mode I and (B): cyclic mode I + superimposed static mode III loading. Such loading cases frequently occur in rotating load-transmission devices.Several differences are typical for these two mixed-mode loading cases. A superimposed static mode I load increases the crack propagation rate under cyclic mode III loading whereas cyclic mode I fatigue crack propagation is retarded when a static mode III load is superimposed. Increase of the R -ratio (of the cyclic mode III load) leads to an insignificant increase of fracture surface interaction and subsequently to a small decrease of the crack growth rate for cyclic mode III loading, whereas higher R -values during cyclic mode I+ superimposed static mode III loading lead to a significant reduction of the crack growth rates.
