    Notes: Abstract: Chromaffin cells both recently isolated or in culture present a high-affinity adenosine transporter with a Km value of 1 μM. When cells were exposed to nerve growth factor (NGF; 10 ng/ml), the adenosine transporter affinity decreased to 3μM. This value was maintained from 3 days after plating to the end of the culture period. A change in the transport capacity was observed, with a significant increase (˜200–260%) in NGF-cultured cells throughout the period studied.

    Notes: Abstract: The presence of calmodulin-binding sites on chromaffin granule membranes has been investigated. Saturable, high-affinity 125I-calmodulin-binding sites (KD= 9.8 nM; Bmax= 25 pmol/mg protein) were observed in the presence of 10−4M free calcium. A second, nonsaturable, calmodulin-binding activity could also be detected at 10−7M free calcium. No binding occurred at lower calcium levels. When chromaffin granule membranes were delipidated by solvent extraction, calmodulin binding was observed at 10−4M free calcium. However no binding was detected at lower calcium concentrations. Thus it appears that a calcium concentration of 10−7M promotes the binding of calmodulin to some solvent-soluble components of the chromaffin granule membrane. Calmodulin-binding proteins associated with the granule membrane were identified by photoaffinity cross-linking. A calmodulin-binding protein complex, of molecular weight 82K, was formed in the presence of 10−4M free calcium. This cross-linked product was specific because it was not detected either in the absence of calcium, in the presence of nonlabeled calmodulin, or in the absence of cross-linker activation. When solvent-treated membranes were used, a second, specific, calmodulin-binding protein complex (70K) was formed. Since the apparent molecular weight of calmodulin in our electrophoresis system was 17K, these experiments suggested the presence of two calmodulin-binding proteins, of molecular weights 65K and 53K, in the chromaffin granule membrane. This result was confirmed by the use of calmodulin-affinity chromatography. When detergent-solubilized membranes were applied on the column in the presence of calcium, two polypeptides of apparent molecular weights of 65K and 53K were specifically eluted by EGTA buffers. Since detergent treatments or solvent extractions are necessary to detect the 53K calmodulin-binding protein, it is concluded that only the 65K calmodulin-binding polypeptide may play a role in the interaction between calmodulin and secretory granules in chromaffin cells.

    Notes: Summary. Plasma levels of CA 125 were determined in 113 patients with ovarian cancer of epithelial origin. Of these, 69 patients had CA 125 measured before the first laparotomy and 84.6% of them had a CA 125 level 〉35 U/ml. In 87 of the 113 patients whose tumour was producing CA 125, a good correlation was observed between the CA 125 levels and the clinical follow-up: 95.7% of the patients in remission had levels 〈 35 U/ml, whereas all the patients with no change or with a progressive disease had levels 〉35 U/ml. Furthermore in recurrent disease the levels of CA 125 were also increased (〉35 U/ml) in 92.3% of the patients. Thus, CA 125 measurements at regular intervals are of great clinical value in following the evolution of a tumour or the success of a therapy, but unfortunately do not allow detection of an ovarian tumour at an early stage.

    Notes: Abstract Composites formed by infiltration of an array of fine alumina fibres with aluminium alloy melts have been investigated in terms of fabrication characteristics, microstructural features and mechanical properties. A production method has been developed in which the application of pressure ensures very low porosity levels and strong fibre-matrix bonding. Details of the transport phenomena occurring during fabrication have been explored with a view to optimizing selection of applied pressure, thermal fields, alloy composition and the structure of the fibrous preform. Microstructural examinations revealed an intimate fibre-matrix bond, but the virtual absence of any chemical reaction at the interface. Comparison of property measurements with data from unreinforced alloys revealed increased elastic moduli and marked improvement in tensile strength at elevated temperature, accompanied by reductions in elongation.

    Notes: Summary Chromogranin A (CgA) and related acidic proteins are widely distributed in the organism. They are also present in entero-endocrine cells and in other members of the paraneuron family. Therefore, CgA has been claimed as an universal marker of this cellular community. To yield precise data about the distribution of CgA in entero-endocrine cells, all segments of the gastro-intestinal tract of five mammalian species (man, cattle, pig, cat, guinea-pig) were investigated immunohistochemically for CgA. In serial semithin plastic sections, all CgA-immunoreactive endocrine cells were identified for resident amines or peptides. CgA could be found in ten hormonally identified endocrine cell types and in two or three other endocrine cell types. Entero-endocrine cells containing amines (histamine, serotonin) regularly exhibited CgA-immunoreactivities. In contrast, peptide-containing endocrine cells were largely heterogeneous: Their CgA-immunoreactivities varied among the species, among the gastro-intestinal segments, and even among the members of the same cell population. Hence, seen histochemically, CgA is no universal marker for entero-endocrine cells. Seen biochemically, the observed heterogeneities of CgA-immunoreactivities theoretically can be attributed to various factors (species-specificities of CgA, subclasses of chromogranins, processing of CgA or its proprotein). Most probably, these heterogeneities are caused by species- or cell-specific differences in the extent of processing of CgA. In addition, some findings point to certain interrelations between the processing or storage of CgA and resisdent peptides in the secretion granules of entero-endocrine cells.

    Description / Table of Contents: Zusammenfassung Beim inoperablen Oesophaguscarcinom wurde als neues Alternativverfahren gegenüber der Palliativresektion die intraluminale hochdosierte Afterloadingtherapie mit Iridium 192 angewandt. Voraussetzung für dieses Verfahren, das im letzten Jahr bei 19 Patienten eingesetzt wurde, ist die vorhergehende laserendoskopische Passagewiederherstellung der Tumorstenose. In 2wöchigen Abständen wird bis zu 3mal endocavitär mit einer Dosis von 7 Gy bestrahlt. Bei kontinuierlicher endoskopischer Patientenüberwachung ist die Tubusimplantation bei besserer Lebensqualität und vergleichbaren Überlebenszeiten vermeidbar.
    Notes: Summary In inoperable esophageal carcinoma the intraluminal high dose afterloading therapy with Iridium 192 was employed as an alternative to the palliative resection. Prerequisite for this treatment, which was used in 19 patients during the last year, is the preceding passage restoration of the tumor stenosis by endoscopic laser therapy Every two weeks the patients are endocavitarily irradiated up to 3 times with a dose of 7 Gy. Observing the patients by endoscopic means the implantation of a tubus is avoidable achieving better quality of life and comparable survival times.

    Notes: The geometrical structure of native silicon oxide and ultra thin thermal silicon oxides grown in situ on a Si(100) wafer have been investigated by surface extended x-ray absorption fine structure (SEXAFS) above the oxygen K absorption edge (∼530 eV). The Si—O bond length of the native oxide is identical with that of α-quartz and amorphous silica and the Si—O bond length of the thermal oxides corresponds closely to that of β-quartz which is the medium temperature modification of α-quartz.
