    Notes: Abstract Acid secretion in isolated rabbit gastric glands was measured by means of the14C-aminopyrine accumulation technique. Hoe 760 (TZU-0460) and Hoe 062, the desacetylated compound of Hoe 760, caused a concentration-dependent reduction of histamine (100 μM) induced aminopyrine-accumulation. The IC50-values were 3.16±0.84 μM (n=5) and 1.58±0.6 μM (n=6) for Hoe 760 and Hoe 062, respectively. In comparison an IC50 of 9.0±0.72 μM (n=6) was obtained for cimetidine and 3.3±1.4 μM (n=5) for ranitidine. The IC50-values of ranitidine, Hoe 760 and Hoe 062 were significantly different (p〈0.05) from cimetidine. The addition of increasing concentrations of Hoe 760 to the histamine concentration-response curve caused a parallel rightward shift. The transformation of these concentration-response curves according to Arunlakshana and Schild indicated that this inhibition was caused by a competitive antagonism of the histamine receptor on the parietal cell. In agreement with these findings the dbc-AMP stimulated aminopyrine accumulation remained unaffected by the H2-receptor antagonists.

    Notes: Summary Hexosaminidase isoenzymes P from serum of pregnant women and I2 from serum of leukemic children were compared using various biochemical methods, namely, ion exchange chromatography, isoelectric focusing, and molecular weight determination by gel filtration. Isoenzyme I2 is activated by calcium, magnesium, and manganese. The molecular weight of isoenzyme I2 was 110 000 compared to 130 000 of isoenzyme P. The widely differing molecular weights exclude the so far suggested identity of both enzymes. Both isoenzyme P and isoenzyme I2 possessβ-chains, however, they differ in chain structure and glycosylation. This different structure of isoenzyme P may be one of the reasons for its increase during pregnancy due to the resulting impaired clearance by liver sinusoidal cells.

    Notes: Abstract Screening of neonates for inborn errors of metabolism has been carried out on a national level since 1969 in the Federal Republic of Germany. To raise the reliability of these routine investigations, we introduced an external quality control in March 1982. Every 2 months ten filter paper samples were sent to the screening centres in West Germany. Some of these samples have a normal and others a slightly raised content of phenylalanine, leucine, methionine and galactose. The success of this external quality control is appraised on the basis of the number of false negative results. In the course of time, screening centres in France, Israel, Italy, Japan, Switzerland, Taiwan, Turkey and Yugoslavia have also practicipated in these inter-laboratory quality controls.

    Notes: Abstract Capillary blood samples from almost one million neonates from Baden-Württemberg were investigated for inborn errors of metabolism between 1969 and 1984 in our screening centre. Besides 7 patients with maple syrup urine disease (MSUD), 3 with homocystinuria and 18 with galactosaemia, a follow-up of the positive screening results confirmed 94 patients with phenylketonuria (PKU) and 76 with non-PKU hyperphenylalaninaemia (non-PKU HPA). The incidence of PKU is 1: 10 000, and that of HPA in the wider sense (PKU and non-PKU HPA) as obtained by newborn screening before further classification at 6 months 1: 5532. For West Germany as a whole, the number of newly discovered cases with persistent hyperphenylalaninaemia was 1480 in the same period. The subdivision into PKU and non-PKU HPA is not yet possible from this figure. It is strongly suggested that the abnormal results of newborn screening for phenylalanine be designated as hyperphenylalaninaemia (in the wider sense) and that the terms “PKU” or “non-PKU HPA” be used only after further differentiation as carried out by us at the age of 6 months.

    Notes: Abstract In 34 children with phenylketonuria (PKU) treated early the prognostic value of the age on institution of the diet (within the first 3 months of life) and of the quality of dietary treatment was determined in two different ways: 1) following intelligence closely (IQ) and (2) evaluating the EEG development up to their 12th (n=34) and 15th (n=18) years of life as appropriate. In general, IQ scores were found to be normal from the 4th–15th years of life. In our group of patients there was no effect on the IQ of the timing of diet onset. Children with “strict” dietary control showed a significantly higher IQ than those with “loose” control. One hundred and fifty-four EEGs (10/20 system, awake with eyes closed) were recorded at intervals of 2 years and conventionally evaluated. The development of alpha-activity was found to be normal. Beta-activity was enhanced. Abnormal EEG findings like general slowing and generalized paroxysmal activity (GPA) with or without spikes were more frequent in children with PKU than in controls, with the exception of focal abnormalities. EEG abnormalities increased with advancing age independently of IQ development and showed no relation to either the age at the onset nor the quality of dietary treatment.

    Notes: Summary The pharmacokinetics of fluocortolone and its effect on the circadian rhythm of plasma cortisol and ACTH have been studied during different schedules of oral administration. Groups of 6 healthy male adults were given a single daily dose of fluocortolone 5, 10 and 20 mg, and another group received 20 mg every second day. Administration to all groups was continued for 8 days. Pharmacokinetic parameters of fluocortolone (half lives of absorption- t1/2a and elimination t1/2e, volume of distribution V and oral clearance (CL/f) were independent of the duration of treatment and dose. Areas under the plasma level curves, AUC and Cmax values increased in proportion to the dose, indicating dose linearity of fluocortolone pharmacokinetics. A normal morning cortisol peak occurred during all treatment schedules, which indicates that circadian cortisol secretion was not seriously affected by the glucocorticoid treatment. On the other hand, when the level of circulating fluocortolone was high the 12.00 a.m. and 4.00 p.m. cortisol levels were diminished compared to the pre and posttreatment values. There was no definite correlation between the ACTH and cortisol levels, either on treatment or on control days. ACTH levels were suppressed by daily treatment with the 20 mg dose. Thus, even during administration of a high dose of a glucocorticoid, the circadian secretion of cortisol can be preserved if the treatment is adjusted according to the half-life of the drug.

    Notes: Summary A specific radioimmunoassay for the quantitative measurement of ACTH 4-10 and a procedure for its extraction from plasma have been developed. Its pharmacokinetics was studied in eight healthy male volunteers given ACTH 4-10 125 µg/kg body weight as a bolus i.v. injection, by infusion and intranasally. Following the i.v. bolus, plasma levels rapidly declined biexponentially, with half-lives of 0.39±0.05 min for the α-phase and 3.84 ± 1.5 min for the β-phase (mean±SD). The constant rate i.v. infusion yielded steady-state levels between 0.74 and 5.06 ng/ml plasma. Administered as intranasal spray, absorption of intact ACTH 4-10 was low and variable (maximal bioavailability 7.6%). The results are discussed in relation to the dose-dependent effects of ACTH 4-10 on the auditory evoked potential.

    Notes: A vibrational analysis for the excitation and fluorescence spectra of the S1–S0 transition in jet-cooled indole and N-deuterated indole is presented. This analysis yields the frequencies for eight low-lying S1 vibrational modes of both isotopic species. Single vibronic level fluorescence spectra also enable us to refine many of the S0 vibrational frequencies. Quantum interference effects in the energy-resolved fluorescence decays for one vibrational level of S1 indole are observed as a result of picosecond laser excitation. Through analysis of the fluorescence spectra and the modulated fluorescence decays, the coupling responsible for the quantum beats was found to result from a zero-order in-plane mode coupled to a level which is described by several zero-order out-of-plane modes. The effect of the deuterium substitution on the quantum beats is also investigated.

    Notes: We have analyzed the jet-cooled torsional spectra of the S1–S0 transitions for the seven monomethyl indoles. The potential curves which describe the hindered internal rotation in both the ground and excited states are modeled with the form: V(φ)= 1/2 ∑n Vn(1−cos nφ). The changes in the torsional potential upon electronic excitation can be interpreted as arising from changes in the electron density adjacent to the methyl rotor. We present evidence that the methyl rotor is then a unique probe with which to obtain information about the detailed electronic structure of the parent indole molecule. This approach provides new evidence for the assignment of the first excited electronic state of indole observed in the vapor phase to 1Lb.
