Notes: Plasma production and heating in the central cell of the Tara tandem mirror [Nucl. Fusion 22, 549 (1982); Plasma Physics and Controlled Nuclear Fusion Research, 1986, Proceedings of the 11th International Conference, Kyoto, Japan (IAEA, Vienna, 1987), Vol. 2, p. 251] have been studied. Using radio-frequency excitation by a slot antenna in the ion cyclotron frequency range (ICRF), plasmas with a peak β⊥ of 3%, density of 4×1012 cm−3, ion temperature of 800 eV, and electron temperature of 75–100 eV were routinely produced. The plasma radius decreased with increasing ICRF power, causing reduced ICRF coupling and saturation of the plasma beta. About 70% of the applied ICRF power can be accounted for in direct heating of both ions and electrons. Wave field measurements have identified the applied ICRF to be the slow, ion cyclotron wave. In operation without end plugging, the plasma parameters were limited by poor axial confinement and the requirements for maintenance of magnetohydrodynamic stability and microstability.

Notes: Cytolylic T lymphocytes (CTL) cause cytolysis of foreign or virus-infected syngeneic cells when recognition of the target plus major histocompatibility complex (MHC) occurs via the T-cell receptor (TCR). The recognition event leads to intimate contact between the two cells and activation of the cytolytic effector. Activation and target cell lysis can also occur in the presence of antibodies to the TCR. This is accomplished by bridging the effector cell TCR to the target cell FcR by an anti-TCR monoclonal antibody (MoAb). Recent findings have placed the role of the FcR in this event in a questionable light. We confirm the importance of FcγR by demonstrating that: (a) melanoma cells are killed by CTL clones in the presence of anti-TCR CD3 antibodies only when the melanoma cells express the FcγR on their surface; (b) native Ig, heat-aggregated Ig, or an Fc fragment from an antibody expressing the same isotype as the anti-TCR antibody can block the killing of high avidity FcγRI-bearing cells mediated by anti-TCR antibody (F23.1); and (c) anti-FcγR MoAb (2.4G2)and a truncated soluble FcγRII molecule inhibit the killing of low-avidity FcγRII-bearing cells mediated by anti-CD3 MoAb (145–2C11). Thus, we show that both high-avidity FcγRI and low-avidity FcγRII can mediate sideways killing depending upon the isotype of the anti-TCR antibody and the type of FcR present on the target cell surface.

Notes: The effects of intraperitoneal injection of monoclonal anti-rat T-lymphocyte antibodies were evaluated immunohistochemically and functionally in normal rats and in rats with experimental allergic neuritis. In the normal animals a single injection of OX8 antibodies, reactive with suppressor/cytotoxic T cells, completely eliminated OX8-reactive cells from peripheral lymphoid organs and from circulation, whereas the ‘pan’ T-cell-reactive W3/13 antibodies and the helper T-cell-reactive W3/25 antibodies only caused a partial elimination of their respective target cells. Injection of the W3/13 and W3/25 antibodies but not of OX8 antibodies led to a diminished responsiveness to allogeneic stimulation in vitro for spleen cells obtained from the treated rats, whereas the OX8 injection caused a complete elimination of the in vitro cytotoxic response to allogeneic cells in the mixed lymphocyte reaction-activated spleen cell population. When Lewis rats were injected with peripheral nerve myelin and Freund's adjuvant for the induction of EAN, treatment with W3/13 antibodies completely prevented the onset of disease, whereas treatment with the OX8 antibodies exaggerated the disease symptoms.

Notes: Abstract A method for growing millimeter-sized crystals of YBa2Cu3O7−δ is described. Processing conditions were developed to improve the superconducting transition temperature and sharpness, as observed by measurements of the dc magnetic susceptibility. Some of our crystals have been used in observations of the Raman Effect, specific heat, nuclear magnetic resonance, and nuclear quadrupole resonance, with results indicating that the samples are excellent.

Notes: Summary Anti-B cell auto-immunity may play a role in the pathogenesis of diabetes in mice resulting from multiple subdiabetogenic doses of the pancreatic B cell toxin, streptozotocin. In the present study we have investigated the cytotoxic anti-B cell response in these mice. A major role for B lymphocytes, macrophages, or their products in the cytotoxic response originally detected in vitro was eliminated by passing splenocytes from the mice treated with multiple subdiabetogenic doses of streptozotocin over a nylon wool column. The removal of the adherent cells enhanced the cytotoxicity against a rat insulinoma cell line in vitro by that expected due to enrichment of T-lymphocytes by approximately twofold. The induction of diabetes after multiple subdiabetogenic doses of streptozotocin is strain dependent. Mice of five strains were immunized with rat insulinoma cells, but only splenocytes from the two strains susceptible to multiple subdiabetogenic doses of streptozotocin demonstrated a significant cytotoxic response against the rat insulinoma cells in vitro. Mice pre-immunized with either the rat insulinoma cells or with syngeneic islets labelled in vitro with the hapten trinitrophenol developed hyperglycaemia more rapidly than control mice after multiple subdiabetogenic doses of streptozotocin. In the latter experiment the control mice immunized with complete Freund's adjuvant alone also became hyperglycaemic after a modified multiple subdiabetogenic dose of streptozotocin that did not cause diabetes in non-immunized mice. In mice pre-treated with either adjuvant or cyclophosphamide and then given a modified multiple subdiabetogenic dose of streptozotocin (35 mg/kg × 5 rather than 40 mg/ kg) the degree of hyperglycaemia was reduced and there was no protective effect of cyclophosphamide. However, the mice pre-treated with adjuvant again developed hyperglycaemia more rapidly and to a much higher level than did the mice given multiple subdiabetogenic doses of streptozotocin only. These additional data further support the hypothesis that B-cell destruction after multiple subdiabetogenic doses of streptozotocin results from triggering of an immune response against these insulin-producing cells.

Notes: Abstract Unique features of doubly-charged stable organic ions are examined and the results correlated with experimental observations. Self-consistent field molecular orbital methods are used to compute structures and stabilities of CnH 2 2+ (n=2–9) ions which are prominent in electron impact ionization of hydrocarbon molecules. A simple curve crossing model is employed to rationalize charge transfer reactions of these ions.

Notes: Doubly charged ion mass spectra of alkyl-substituted furans and pyrroles were obtained using a double-focusing magnetic mass spectrometer operated at 3.2 kV accelerating voltage. Molecular ions were the dominant species found in doubly charged spectra of lower molecular weight heterocydic compounds, whereas the spectra of the higher weight homologues were typified by abundant fragment ions from extensive decomposition. Measured doubly charged ionization and appearance energies ranged from 22.8 to 47.9 eV. Ionization energies were correlated with values calculated using self-consistent field-molecular orbital techniques. A multichannel diabatic curve-crossing model was developed to investigate the fundamental organic ion reactions responsible for development of doubly charged ion mass spectra. Probabilities for Landau-Zener type transitions between reactant and product curves were determined and used in the collision model to predict charge-transfer cross-sections, which compared favorably with experimental cross-sections obtained using time-of-flight techniques.
