    Notizen: Summary Much more tannic acid or pin oak tannin is required to precipitate the abundant leaf protein, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPC), from Manduca sexta gut fluid adjusted to pH 6.5 than is required to precipitate this protein from an aqueous buffer at the same pH. This finding demonstrates that some characteristic of M. sexta gut fluid, in addition to its basicity, counteracts the potential of tannins to precipitate ingested proteins. Gut fluid of M. sexta has a surface tension of 36–39 dynes/cm, indicating the presence of surfactants. Lysolecithin and linoleoylglycine, surfactants known to be present in insect gut fluids, also interfere with the precipitation of RuBPC by tannins at pH 6.5. It is concluded that detergency is a widespread property of insect gut fluids that counteracts the potential of tannins to precipitate die ary proteins, and it is argued that there is no longer any justification for continuing to refer to tannins as digestibility-reducing-substances. Finding that there has been no formidable barrier to the evolution of mechanisms that counter a generalized antidigestive action by tannins is difficult to reconcile with the idea that reduced digestibility is an evolved anti-herbivore adaptation of apparent plants.

    Notizen: Summary There is no correlation between protein-precipitating capacity and either total phenolic or proanthocyanidin content of extracts of mature foliage from six species of oaks: Quercus alba (white oak), Q. bicolor (swamp white oak), Q. macrocarpa (bur oak), Q. palustris (pin oak), Q. rubra (red oak), and Q. velutina (black oak). It is argued that studies which probe the role of tannins in the selection and utilization of food by herbivores should include a protein-precipitation assay, since such an assay provides a measure of the property of tannins which is presumed to contribute to their utility as defensive compounds. A convenient modification of the bovine serum albumin (BSA) precipitation assay, which measures the amount of protein precipitated when a plant extract is added to a BSA solution, is described. Advantages of this procedure recommend its routine adoption in studies of the role of tannins in plant-herbivore interactions.

    Notizen: Abstract We examined several of the mechanisms that have been reported to enable polyphagous grasshoppers (Orthoptera: Acrididae) to tolerate ingested hydrolyzable tannins: hydrolysis, adsorption on the peritrophic envelope, and peritrophic envelope impermeability. None of these mechanisms explain the tolerance ofMelanoplus sanguinipes to ingested tannic acid. In this species, tannin hydrolysis was 12–47% complete, adsorption accounted for less than 1% of the tannic acid contained in the midgut, and the peritrophic envelope was permeated by several gallotannins. The foregut is the main site for the chemical transformation of tannic acid in this species. InPhoetaliotes nebrascensis, hydrolysis was more extensive (82% complete), but the peritrophic envelope was readily permeated by two gallotannins. Oxidizing redox conditions were found in the guts of both species, and ingested tannins were oxidized inM. sanguinipes. We hypothesize that the tolerance of some polyphagous grasshoppers to ingested hydrolyzable tannins may be the consequence of their ability to tolerate the reactive oxygen species generated by polyphenol oxidation, whereas others may rely on rapid and extensive hydrolysis.

    Notizen: Abstract Tannic acid and quebracho precipitate many times their weight of the abundant leaf protein, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPC). The use of this protein in protein precipitation assays for tannin content is described. Extracts of mature foliage of pin, bur, and black oak precipitate 2.01, 0.69, and 0.09 mg RuBPC/ mg (dry wt) of leaf powder extracted, respectively, at pH 6.1. From these measurements it can be calculated that all three of these oak species have sufficient tannins to precipitate all of the RuBPC present in their foliage. At mildly alkaline pHs, however (pH ⩾ 7.5), RuBPC is not precipitated by tannins. Since RuBPCis the most abundant protein present in photosynthetic tissues, often constituting as much as 50% of the soluble proteins and 25% of the total proteins in leaf tissue, the interactions of this protein with tannins are highly relevant to an evaluation of the role of tannins as antiherbivore, digestibility-reducing substances. Our measurements provide no basis for arguing that differences in tannin levels in different species reflect differences in the digestibility of leaf proteins or that tannins have any effect whatsoever upon the digestibility of leaf protein under conditions which normally prevail in most insects' guts. These findings emphasize the need to test more of the assumptions underlying contemporary interpretations of the importance of tannins in plant herbivore interactions.
