    Notes: Natural killing (NK) activities were measured in peripheral blood lymphocytes (PBL) from 14 patients with Werner's syndrome (WS), and the results were compared with those of 187 normal individuals at different ages. The NK activities of healthy donors were demonstrated in a characteristic and invariable manner in accordance with the age and the sex, whereas the activities of WS patients were considerably reduced irrespective of the age and the sex. When normal PBL were preincubated with WS sera containing anti-lymphocyte antibodies (ALA) plus rabbit complement, the NK activities of the surviving PBL were markedly reduced when compared with those of PBL treated with other heterologous ALA of rabbit origin in the same manner. PBL from WS patients treated with purified human leucocyte interferon augmented the NK activity even beyond the range of controls. These results suggested that NK activities in WS patients were reduced in a manner similar to that in normal old individuals. The ALA reactive to NK cells in WS patients may participate in the reduction of NK activities. The augmentative effect of interferon on NK activities in WS patients may also suggest the possible blocking of potent NK activities by an unknown mechanism rather than an intrinsic defect of their NK cells.

    Notes: Accumulating evidence suggests a link between immediate hypersensitivity and cellular immunity. In this study, we examined the effect of interleukin 2 (IL-2) on basophil histamine release. Histamine-releasing activity of IL-2 was very weak with % histamine release of 2.9 ± 1.3 (mean ± SEM, n= 9) at 1:12 dilution. IL-2 at 1:1200 dilution slightly inhibited anti-IgE-induced histamine release by 22.4 ± 18.6% (P 〉 0.05). There was a significant potentiation of release at 1:12 dilution of IL-2 with % enhancement of 78.7 ± 42.2 (P 〈 0.05). IL-2 enhanced the calcium ionophore A23187-induced histamine release in a dose-dependent fashion. IL-2 at 1:12 dilution significantly potentiated release by 28.8 ± 6.3% (P 〈 0.05). There was a slight suppression of formyl-methionyl-leucyl-phenylalanine (FMLP)-induced histamine release at 1:1200 dilution with % inhibition of 23.4 ± 7 4 (P 〉 0.05). At 1:12 dilution, IL-2 significantly potentiated FMLP-induced release by 73.7 ± 41.6% (P 〈 0.05). Recombinant IL-2 (RIL-2) augmented anti-IgE-induced histamine release with a significant enhancement at 200 units/ml. Conventional IL-2 was more potent than RIL-2 in enhancing release. These results indicate that IL-2 enhances basophil histamine release and some part of the effect of IL-2 on basophils is derived from other factors contained in conventional IL-2.

    Notes: Abstract Effects of E-5110, a novel non-steroidal antiinflammatory drug, on interleukin-1 (IL-1) generation from human monocytes were studiedin vitro. E-5110 reduced the amounts of extra- and intracellular IL-1 activity induced by lipopolysaccharide (LPS, 1 μg/ml) in a dose-dependent manner (1–10μM). E-5110 also inhibited the IL-1 generation induced by antigen-antibody complexes, opsonized zymosan and silica particles. It was suggested that the inhibition of IL-1 generation by E-5110 was independent of the inhibitory effects on arachidonate cyclooxygenase and/or lipoxygenase because indomethacin, piroxicam, BW755C and AA861 had no effects on IL-1 generation. Hydrocortisone (IC50:0.084 μM), aurothioglucose (11.5 μM) and lobenzarit (75.0 μM), which are clinically effective antirheumatic drugs, also inhibited IL-1 generation, like E-5110 (1.21 μM). It is expected that E-5110 will be superior to classical non-steroidal antiinflammatory drugs in medical treatment of rheumatoid arthritis.

    Notes: Summary The introduction of acrylate resins (Lowicryl K 4M, LR White) into electronmicroscopic immunocytochemistry applied to embedded tissue (post-embedding method) has improved the localization of antigens because of a satisfactory preservation of both ultrastructure and antigenicity of tissues. Here we describe a method that allows double staining of intracellular and membranous determinants in ultrathin sections of nervous tissue and cultures of peripheral nervous system cells. Ultrathin sections of the rat central nervous system fixed on uncoated grids were stained first for MBP selectively on the one face, then the opposite face was stained for GFAP using monoclonal antibodies and indirect immunogold staining method (IGS). Cultured Schwann cells induced to express major histocompatibility complex (MHC) class II antigens were stained for class. H antigens by pre-embedding method then followed by post-embedding IGS for the other intracytopasmic antigens.

    Notes: Summary The direct innervation of the pial blood vessels by serotonin neurons has been demonstrated with a modified peroxidase-antiperoxidase technique in the mammalian central nervous system. The pia mater covering the ventrolateral surface of the medulla oblongata is innervated by numerous varicose serotonin fibers originating from the serotonin neurons of the lower brainstem. Scattered serotonin fibers were observed in the pia mater in every part of the brain and spinal cord.

    Notes: Summary The distribution and morphological aspects of the serotonin-containing neurons in the paraventricular organ of the carp, frog, turtle and chicken were studied by means of an immunoperoxidase technique using serotonin antiserum. In all species the serotonin-containing neurons were seen to have the appearance of the CSF-contacting neurons and to be distributed in the pars ependymalis and the pars hypendymalis of the organ. Particularly, in the frog, the serotonin-containing CSF-contacting neurons, mostly bipolar in shape, were also observed in the pars distalis. Their proximal processes protruded into the ventricular lumen through the ependymal layer with a globular- and triangular-shape. The distal processes projected ependymofugally to the pars distalis and formed a fine plexus in the neuropil of this part. The density of the serotonin fibers in the pars distalis was greater in the carp than in the other species.
