    Notes: Summary Oxygen intermediates (OIs) generated by stimulated polymorphonuclear leukocytes (PMNs) are known to induce auto-oxidative tissue damage at the site of inflammation. PMNs from five patients with severe and chronic cement dermatitis generated markedly increased levels of OIs. However, only a slight increase in OI generation by PMNs was observed in cement workers without cement dermatitis. Dapsone, which has recently been shown to decrease OI levels, was found to be clinically effective in the treatment of cement dermatitis in these five patients. After treatment, a significant decrease in OI generation was observed in all patients studied. In skin tissues from the cement workers without cement dermatitis, enhanced superoxide-dismutase (SOD) activities as well as increased OI generation by PMNs were noted. In spite of the greatly increased OI generation by PMNs, the SOD activities in the patients were comparable to those in healthy controls. These findings suggest that the severe skin manifestations in patients with cement dermatitis can partly be explained by a defective capacity for enhancing SOD activity which removes increased PMN-derived OIs and thus prevents subsequent tissue injury by OIs at the site of inflammation.

    Notes: The effects of potassium iodide, colchicine and dapsone on the in vitro generation of polymorphonudear leukocyte (PMN)-derived oxygen intermediates were investigated. These three drugs have beneficial effects on those conditions in which PMNs play an important pathogenetic role. Three oxygen intermediates, superoxide anion (O2−), hydrogen peroxide (H2 O2), hydroxyl radical (OH·) and chemiluminescence were included in assay studies. Dose response studies were performed with therapeutic doses of the drugs (10 μM–1 mM). We found that both potassium iodide and dapsone significantly suppressed the generation of oxygen intermediates, except for O2−. Colchicine decreased OH · production.Our results show that these agents to some extent exert their anti-inflammatory effects by interfering with the PMN-dependent production of oxygen intermediates, thus conferring protection from auto-oxidative tissue injury. This may account for their clinical efficacy in many PMN-mediated dermatological diseases.

    Notes: Serum levels of squamous cell carcinoma-related antigen (SCC-RAG) were measured in five cases of pemphigus, five cases of bullous pemphigoid and 18 cases of benign and malignant dermatoses other than SCC. The SCC-RAG titres were significantly raised in four of five patients with pemphigus, while they remained within the normal range in the other dermatoses except in one case. In three pemphigus cases in whom serial measurements were made, SCC- RAG levels seemed to be related to disease activity. The SCC-RAG levels in blister fluids were much higher than those in serum, suggesting that the skin is a major source of serum SCC- RAG.These results show that SCC-RAG is increased not only in SCC, but also in some cases of pemphigus, and suggest that pemphigus antibodies may cause the production or release of SCC-RAG.

    Notes: Ultrastrutual, immunohistochemical and gene analytical studies were carried out on a 39-year-old patient with lymphomatoid papulosis. Two different cell groups were demonstrated in the papulonodular eruptions: large atypical cells with multiple nuclei that were well stained with anti-Tac, but not with Leu 3a, and other cells that possessed prominent hyperchromatic nuclei and which stained well with Leu 1 and Leu 3a but not with anti-Tac. Gene analytical studies using EcoRI, BamHI and HindIII revealed no rearrangement, indicating a non-clonal T-cell proliferation unlike malignant T-cell lymphoma. These results suggest that the present case was benign.

    Notes: The effects of local hyperthermia treatment on contact sensitivity (CS) and on the number of Langerhans cells (LCs) were studied in mice. CS was significantly suppressed when mice were sensitized in the hyperthermia treated skin 1, 2 or 4 days after treatment (43°C for 45 min). This suppressive effect was not observed 7 or 14 days after the treatment. CS was also suppressed when mice were sensitized in non-treated skin 1 day after the treatment. The density of LCs detected as ATPase-positive cells also decreased significantly 1, 2, 4 and 7 days after the treatment. There appeared to be a positive correlation between the number of LCs and the extent of CS when mice were sensitized at hyperthermia treated skin. It was observed that this suppressive effect on CS was dose- and temperature-dependent. It could be transferred by spleen cells from the hyperthermia treated and DNFB-sensitized donors, and was antigen specific when spleen cells were transferred before sensitization of the recipient mice. This indicated it was, in part, associated with the induction of suppressor cells. These findings suggest that local hyperthermia treatment reduces the number of LCs with subsequent suppression of the induction phase of delayed-type hypersensitivity by the genertion of antigen-specific suppressor cells.

    Notes: To elucidate the mechanism of action of metronidazole in the treatment of rosacea, the effect of metronidazole on the generation of reactive oxygen species was examined both in neutrophil and xanthine-xanthine oxidase systems. Metronidazole had anti-oxidant action which was not exerted by the scavenging of reactive oxygen species, but by having an effect on neutrophil cell functions. Beneficial effects of metronidazole in the treatment of papulopustular rosacea can in part be attributable to this anti-inflammatory effect.

    Notes: We describe a patient in whom a suppressor/cytotoxic T cell lymphoma was suspected on the basis of histology and immunohistochemistry. DNA analysis of specimens from three different lesions, using restriction enzymes Eco RI and Bam HI, showed the same rearrangement of the T cell receptor β-chain genes. We consider this technique to be of value in the early diagnosis of malignant lymphoma when differential diagnosis from other lymphoproliferative disorders is difficult with conventional methods.

    Notes: The anti-oxidant efficacy, in vitro, of the gold compounds auranofin (AF) and gold sodium thiomalate (GST) was examined by studying their effects on the generation of reactive oxygen species (ROS) using zymosan-stimulated polymorphonuclear leukocytes (PMNs) and a cell-free, xanthine-xanthine oxidase system. The oxygen species investigated were the superoxide radical anion (O2−) hydrogen peroxide (H2O2) and the hydroxyl radical (OH·).AF had an inhibitory effect on ROS production by PMNs. In particular, OH· generation was significantly suppressed in a dose-dependent fashion. AF did not inhibit ROS production in the cell-free system. GST produced only a small degree of inhibition at higher concentrations.These findings suggest that AF may play an important role in the inhibition of respiratory bursts and the generation of inflammatory reaction products. Since the products of the respiratory burst, especially potent oxidants such as OH· and H2O2, are thought to be important inflammatory mediators, it is postulated that the blockade of toxic ROS generation by AF affects rheumatoid as well as dermatological inflammation and tissue damage.

    Description / Table of Contents: Zusammenfassung Experimentelle Hautinfektion mit Candida albicans bei Meerschweinchen wird histologisch durch intensive epidermische Akkumulation der polymorphonuclearen Leukocyten (PMN) charakterisiert. Um die Rollen der PMN in vivo zu studieren, injizierten wir i.p. 250 μg Colchicin/kg, einen potenten Inhibitor der PMN-Chemotaxis, und beobachteten Einflüsse der reduzierten PMN-Migration bei der experimentellen Candidiasis, mit spezialem Interesse am Eliminationsverlauf des Organismus. Es gab bei der Colchicin-behandelten Gruppe eine signifikante Verzögerung mit der Abnahme der epidermischen PMN-Infiltration und der Verlängerung des sichtbaren Schuppenprozesses. Unsere Daten zeigen, daß die Verzögerung der epidermischen PMN-Migration dem Verschwinden des Organismus aus der infizierten Haut entspricht und daß PMN in der Abwehr gegen die Candida-Infektion, besonders in dem Eliminationsverlauf, eine wichtige Rolle spielen.
    Notes: Summary Experimental cutaneous Candida albicans infections in guinea pigs are histologically characterized by intense epidermal polymorphonuclear leukocyte (PMN) accumulation. To study the role of PMNs in vivo, we injected 250 μcolchicine/kg i.p., a strong inhibitor of PMN chemotaxis, and observed the influence of reduced PMN migration in experimental cutaneous candidiasis with special interest in the elimination course of the organisms. There was a significant delay in the clearance of the organisms in the colchicine-treated group with decreased epidermal PMN infiltration and prolonged visible scaling process. Our data suggest that the delay of epidermal PMN migration parallels the disappearance of the organisms from the infected skin and that PMNs play an important part in defense against candida infections especially in the elimination process.
