    Notes: Abstract Hydroxylamine oxidation was measured in four recently isolated heterotrophic nitrate-reducing bacteria belonging to the genera Pseudomonas, Moraxella, Arthrobacter and Aeromonas. A hydroxylamine-cytochrome c oxidoreductase activity was detected in periplasmic fractions of the Pseudomonas and Aeromonas spp. and in total soluble fractions of the Arthrobacter sp. A monomeric 19-kDa non-haem iron hydroxylamine-cytochrome c oxidoreductase was purified from the Pseudomonas species and shown to be similar to hydroxylamine-cytochrome c oxidoreductase of Paracoccus denitrificans.

    Notes: Abstract A strain of Pseudomonas putida that can express a nitrate reductase that is located in the periplasmic compartment was isolated from freshwater. The enzyme was active in vivo during arginine fermentation and at the onset of oxygen limitation in batch cultures. The activity of the enzyme increased the yield of bacteria following fermentative growth under anoxic conditions with arginine, but nitrate reduction did not support growth on non-fermentable carbon substrates under anoxic conditions. Cells expressing the periplasmic nitrate reductase were capable of reducing nitrate in the presence of oxygen. Nitrate reduction under oxic conditions was clearly coupled to a respiratory electron transport chain because: (1) the process was sensitive to the respiratory inhibitors rotenone and 2-n-heptyl-4-hydroxyquinoline N-oxide, and (2) membrane-bound and periplasmic cytochromes were involved. This is the first report of the presence of a periplasmic nitrate reductase in a member of the γ proteobacteria.

    Notes: Objective To undertake a five year follow up of a cohort of women and children delivered by forceps or vacuum extractor in a randomised controlled study.Design Follow up of a randomised controlled trial.Setting District general hospital in the West Midlands.Population Follow up questionnaires were sent to 306 of the 313 women originally recruited at the North Staffordshire Hospital to a randomised controlled study comparing forceps and vacuum extractor for assisted delivery. Two hundred and twenty-eight women responded (74.5%) and all were included in the study; forceps (n= 115) and vacuum extractor (n= 113).Main outcome measures Bowel and urinary dysfunction, child vision assessment, and child development.Results Maternal adverse symptoms at long term follow up were relatively common. Urinary incontinence of various severity was reported by 47%, bowel habit urgency was reported by 44% (98/225), and loss of bowel control ‘sometimes’ or ‘frequently’ by 20% of women (46/226). No significant differences between instruments were found in terms of either bowel or urinary dysfunction. Overall, 13% (20/158) of children were noted to have visual problems. There was no significant difference in visual function between the two groups: ventouse 11/86 (12.8%), compared with forceps 9/72 (12.5%); odds ratio 0.97,95% CI 0.38–2.50. Of the 20 children with visual problems, a family history was known in 18, and 17/18 (94%) had a positive family history for visual problems. No significant differences in child development were found between the two groups.Conclusions There is no evidence to suggest that at five years after delivery use of the ventouse or forceps has specific maternal or child benefits or side effects.

    Notes: Abstract: Cytoplasmic inclusion bodies that are accumulations of neurofilaments are the pathological hallmark of many neurodegenerative diseases and have been produced in transgenic mice by overexpression of mouse (NF-L and NF-M; light and medium chains, respectively) and human (NF-M and NF-H; medium and heavy chains, respectively) neurofilament subunits. This report describes a neuronal culture model in which human NF-L was overexpressed to produce cytoplasmic accumulations of neurofilaments within cell bodies concomitant with the collapse of the endogenous neurofilament network. Electron microscopy showed that, within accumulations, neurofilaments retained a filamentous structure. The culture model thus provides a novel system in which the effect on neurofilament accumulations of manipulating protein phosphorylation can be studied. Treatment of cells containing neurofilament accumulations with bisindolylmaleimide, a specific protein kinase C inhibitor, resulted in regeneration of the filamentous network; this effect was not due to a change in the level of transfected NF-L expression. These findings lend support to the suggestion that an impairment in the regulation of protein phosphorylation may lead to the accumulation of neurofilaments seen in neurodegenerative disease.

    Notes: Abstract— A total of 25 human brain tumors and 4 specimens of human brain were rapidly frozen at the time of operation and analyzed for ATP, ADP, AMP, UTP, total nucleoside triphosphates, P-creatine, creatine, inorganic P, creatine kinase, lipid and glycogen. Analyses were made on submicrogram samples dissected from frozen dried sections in order to obtain material as free as possible from admixture with brain, necrotic tissue, blood, etc. A method was developed to estimate the original water content of the frozen dried samples. The brain specimens contained five times as much glycogen as small mammal brains, otherwise the values were similar. The tumors were in fair to excellent energy status. Within the areas chosen for assay, most of ATP and total adenylate were substantially higher than in brain in the case of 5 out of 15 gliomas, 3 of 5 meningiomas, and 1 of 4 schwannomas. UTP was almost invariably higher and other nucleotide triphosphates (besides ATP and UTP) lower than in brain. Glycogen was extremely variable, ranging among the gliomas from 0.05% to 6% of dry wt (4 times the level in the human brains). Creatine plus P-creatine, compared to cerebral cortex levels, ranged from 15 to 85% in gliomas, was about 25% in meningiomas and the only medulloblastoma, and varied between 6 and 8% in the schwannomas. P-Creatine varied more or less in keeping with the energy status. Creatine kinase was exceedingly variable. It was almost zero in the schwannomas, the medulloblastomas, 3 of 5 meningiomas, and 2 of 15 gliomas, whereas in some of the gliomas the activity approached that found in brain.

    Notes: We report an extremely rare complication of regional anaesthesia, a spinal subdural haematoma, which resulted in permanent neurological damage occurring 8 days after dural puncture at T12–L1. Although spinal subdural haematoma following spinal anaesthesia and lumbar puncture has been described before, this is the first report of this complication occurring after dural puncture using a 25G atraumatic pencil point (Whitacre) needle. Contributory factors might have been the perioperative intermittent low dose aspirin therapy and the fact that spinal anaesthesia was performed at the T12-L1 level.
