    Notes: Abstract In the present report we studied the formation of severe gastric erosions produced in fasted rats by intragastric administration of piroxicam (PRX), an enolic acid-derived NSAID. The time course of gastric damage and the possible role of mucus secretion, endogenous sulphydryl compounds, changes of gastric vascular permeability and neutrophil infiltration in the development of PRX-induced gastric lesions were also investigated. PRX dose-dependently (1.25–20 mg/kg) caused acute gastric haemorrhagic erosion in the rat. The lesions increased with time until 9 hr after dosing. Mucus secretion did not change significantly with respect to the control group with 5, 10 and 20 mg/kg of PRX at different times (3 and 6 hours) of treatment. There was also no increase in the concentration of its components. In addition, oral pretreatment of the animals with PRX did not significantly change the amount of dye trapped in the stomach. In contrast, non-protein SH fraction was decreased after administration of PRX and MPO activity as an index of neutrophil infiltration was significantly increased. These results suggest that independently of the PRX dose, depletion of endogenous non-protein SH and neutrophil infiltration could play an important part in the pathogenesis of gastric mucosal injury induced by PRX.

    Notes: Abstract. Objective and Design: The aim of the present study was to examine the effects of cinitapride, a novel prokinetic benzamide-stimulating gastrointestinal motility agent, on gastric secretion and ulceration in rats and elucidate some possible vascular and anti-oxidant mechanisms of such protection.¶Material: Male Wistar rats.¶Treatment: Cinitapride (CNT, Lab. Almirall, S.A.) (0.25, 0.5, and 1 mg/kg) and 5-hydroxytryptamine (5-HT, Sigma Chemical Co., St. Louis, MO, USA) (10 mg/kg), administered intraperitoneally (i.p.).¶Methods: Gastric ulceration was induced by instillation of 1 mL/100 g animal of 50% (v/v) ethanol in distilled water and by pylorus-ligated rat model. Gastric microvascular changes, and the activity of myeloperoxidase (as a marker of neutrophil infiltration) and glutathione peroxidase (an important enzyme in scavenging of lipid peroxides) were determined. The results were compared with those of 5-HT. The data were evaluated using Student’s t-test for paired data and the non-parametric Mann-Whitney U-test.¶Results: In 4 h pyloric-ligated animals, i.p. CNT did not significantly reduce the incidence of gastric mucosal damage, and no significant differences were found in the values of total volume and acidity. However, CNT caused a marked and dose-dependent reduction of haemorrhagic lesions induced by 50% v/v ethanol. These protective effects were specifically related to a reduction of neutrophil infiltration. CNT at the dose of 1 mg/kg raised the decreased glutathione peroxidase activity to the control level. In contrast, pretreatment with 5-HT worsened the ethanol-induced erosions, but did not significantly induce any gastric microvascular changes. However, the myeloperoxidase activity rose markedly and the glutathione peroxidase levels decreased significantly in the mucosa injured by 50% v/v ethanol.¶Conclusions: This study demonstrates a new gastroprotective feature of CNT that could be partly explained not only through reduction of neutrophil toxicity but also by an increased synthesis of free-radical scavenging enzymes such as glutathione peroxidase. Furthermore, it is likely that serotonergic-dependent mechanisms are also involved via 5-HT2-receptor blockade and 5-HT1 receptor activation.

    Notes: Abstract  Pacharán is a traditional drink from Navarra (Spain). This liqueur is obtained by maceration of sloe berries (Prunus spinosa L.) with aqueous ethanol and subsequent addition of sugar and essential oils of anise (Pimpinella anisum L. or Illicium verum H.). The following volatile components of pacharán were identified and quantified: cis-anethol; trans-anethol; anisaldehyde; benzaldehyde; benzyl alcohol; γ-butyrolactone; 1,1-diethoxyethane; ethyl benzoate; ethyl lactate; diethyl malate (hydroxybutanedioic acid diethyl ester); estragol (methyl chavicol); eugenol [2-methoxy-4-(2-propenyl)-phenol]; 2-furaldehyde; linalool; cis-3-hexen-1-ol; (4-methoxyphenyl)propanone; 2-phenylethanol and vanillin.

    Description / Table of Contents: Zusammenfassung 67 Patienten mit 68 durch Streptokinasebehandlung eröffneten Arterien wurden bis zu sechs Jahren später nachuntersucht. In zehn der 68 Fälle (=15%) war ein Reverschluß noch während des stationären Aufenthaltes aufgetreten. Das verbleibende Patientenkollektiv mit definitiv eröffneten Gefäßen gliederte sich in drei Gruppen: Patienten, bei denen eine Beurteilung der Gefäßverhältnisse möglich war (A), Patienten, die in der Zwischenzeit verstorben waren (B) und Patienten, bei denen aus technischen Gründen eine Ermittlung des Gefäßstatus ausgeschlossen war (C). Folgende Ergebnisse wurden in der Gruppe A ermittelt: 1 In den ersten Jahren nach der Streptokinasebehandlung stieg die Reverschlußrate stetig bis auf 21% an und schwankte bis zum 6 Jahr um diesen Wert. 2 Die A. iliaca schnitt als eröffnetes Gefäßsegment am besten ab. Ihre Reverschlußtendenz war minimal und lag über die 6 Jahre Beobachtungszeit hinweg um 0 bis 14%. 3 Die A. femoralis erreichte eine Reverschlußinzidenz von 4/15 (=27%) nach 1 Jahr, von 2/11 (=18%) nach 2 Jahren, von 2/6 (=33%) nach 3 Jahren und von 3/6 (=50%) nach 5 und 6 Jahren. 4 Die Reverschlußtendenz der Aorta bewegte sich zahlenmäßig zwischen den für die A. iliaca und A. femoralis ermittelten Raten. 5 Die konsequente und therapeutisch gut eingestellte orale Antikoagulantienbehandlung setzte die Reverschlußrate signifikant herab. Die Rauchkarenz wirkte tendenziell in gleicher Richtung, ohne allerdings das Signifikanzniveau zu erreichen.
    Notes: Summary 67 patients in whom a total of 68 arteries had been made patent by streptokinase treatment were observed for periods of up to six years. In ten of the 68 cases (=15%) reocclusion occurred while patients were still in the hospital. The remaining case material with definitely cleared vessels was divided into three groups: (A) patients in whom evaluation of the vascular status was possible; (B) patients who had died in the meantime; and (C) patients in whom the state of the vessels could not be ascertained for technical reasons. Listed below are the results obtained in Group (A): 1 The reocclusion rate showed a steady rise of up to 21% during the first three years following streptokinase treatment, and fluctuated around that value up to the end of the sixth year. 2 The iliac artery permitted the best results among the vascular segments cleared. The reocclusion rate was very low at 0 to 14% throughout the six-year observation period. 3 The femoral artery reached a reocclusion rate of 4/15 (=27%) after 1 year, of 2/11 (=18%) after 2 years, of 2/6 (=33%) after 3 years and of 3/6 (=50%) after 4 and 5 years. 4 The aortic reocclusion figure was between the figures ascertained for the iliac and femoral arteries. 5 Regular and therapeutically effective oral treatment with anticoagulants produced a significant drop in the reocclusion rate. Abstinence from smoking showed a similar trend which, however, did not reach the significance level.
