Notes: Summary In the outermost layer of porcine-developing enamel adjacent to the ameloblasts in the secretory stage, the activities of two proteinases having molecular masses of 76 and 78kDa were detected by enzymography using gelatin as a substrate. On the other hand, high activities of known 30 and 34kDa proteinases were localized in the inner layer of the enamel. The 76kDa proteinase cleaved the carboxylterminal peptide of porcine 25kDa amelogenin to convert it to 20kDa amelogenin. The 78kDa proteinase also acted on the 25kDa amelogenin similarly, but its activity was weak. The results indicate that the 25kDa amelogenin synthesized and secreted by ameloblasts is converted to 20kDa amelogenin by the action of proteinase localized in the outermost layer of the secretory enamel, and then further degraded by the proteinases in the inner layer of the enamel associated with the increase of mineralization.

Notes: Two patients became infested with Gnathostoma nipponicum after eating raw loach-fish they had caught in a rice field in central Japan. A fragment of Gnathostoma was found in a biopsy from one of them. The sera of both patients reacted with Gnathostoma antigen using indirect immunofluorescence. Scanning electron microscopy was performed on blocks of the paraffin-embedded parasite sample and the viscera of a fish from the same rice field. The risk of eating raw freshwater fish is discussed.

Notes: Abstract— A simple fluorometric method for the estimation of endogenous 3,4-dihydroxyphenylethylene glycol sulphate (DOPEG-SO4) in brain tissue is described. DOPEG-SO4 was extracted from brain tissue with 0.4 n-perchloric acid and purified by column chromatography on QAE-Sephadex A-25, Cl− form. The isolated DOPEG-SO4 was hydrolysed by incubation with a purified preparation of sulphatase from Helix pomatia. The free DOPEG thus formed was further purified on a column of aluminium oxide and determined fluorometrically after conversion to a fluorescent compound by condensation with ethylenediamine. The sensitivity of this method was about 15 ng per perchloric acid extract applied on the column (3 ml) and the reproducibility was excellent, with a variation coefficient of 1.6% (n = 5). The concentration of DOPEG-SO4 in whole brains of rats, determined by the present method, was 79.3 ± 4.7 ng/g (mean ± S.D., n = 10) in terms of free DOPEG. No appreciable DOPEG-SO, was detected in the brains of mice and guinea-pigs.

Notes: Abstract. The activity of piromidic acid (PA, 8-ethyl-5, 8-dihydro-5-oxo-2-prior lidinopyrido [2,3–d] pyrimidine-6-carboxylic acid) against bacteria in fish was evaluated in vitro and in vivo. In vitro, PA was effective against a wide range of pathogenic bacteria stocked in this laboratory and freshly isolated from fish. PA was active against organisms showing multiple drug-resistance and showed no cross-resistance to chloramphenicol, tetracycline and sulfamonomethoxine. In vivo, orally administered PA was effective against experimental infections with Aeromonas hydrophila and Vibrio spp. in goldfish and eels. The efficacy of PA against these infections was equal to or higher than that of chloramphenicol, and higher than that of tetracycline. Distribution of PA in the Ash was investigated by bioassay and autoradiography. In the bioassay, orally administered PA was easily absorbed and distributed in the blood and tissues of major organs within 1 h after the administration. Peak levels were attained 2 to 4 h post dosing, and no residue was detected in the blood and tissues 48 h post dosing. The results obtained from the autoradiographic investigation in goldfish were similar to those from the bioassay. Therapeutic levels of PA were well tolerated by fish when administered with feed.

Notes: Abstract. The pharmacokinetic and metabolic disposition of piromidic acid (PA) was studied in goldfish orally administered a single dose of 40 mg/kg of PA or 14C-PA with the following results. PA was rapidly absorbed from the alimentary tract and became widely distributed to give maximal organ and tissue levels mostly after 1 h post dosing. Organ and tissue levels of PA as measured by the radioactivity were 1.4–2.2 times as high as those obtained by the bioassay, though their distribution patterns were similar. PA was partly biotransformed to unconjugated antibacterial metabolites, i.e., α-OHPA and β-OHPA and partly to more hydrophilic glucuronides which were excreted into bile. Bioactive metabolites along with PA disappeared almost entirely from the fish within 48 h post dosing. The antibacterial activity of α-OHPA and β-OHPA against major pathogenic bacteria isolated from fish showed two to four-fold greater activity as compared with PA. This pharmacokinetic and metabolic disposition, together with the antibacterial activity of PA and its active metabolites, suggest a possible practical application of PA against bacterial fish diseases.

Notes: Abstract— A study has been made of the role of Prior Austenite Grains (PAG) in fatigue crack initiation and propagation in low carbon martensite. The occurrence of ferrite precipitation along prior austenite grain boundaries during rapid cooling will lead to the formation of intergranular cracking. This intergranular cracking has a close relationship with the reduction of the endurance limit for fine grained low carbon martensite.

Notes: Summary New compositional and optical data are reported for antimonian and antimonianbismuthian varieties of hemusite from epithermal Au-Ag-Cu deposits in Japan. The empirical formula for the antimonian variety, from the Iriki mine is: (Cu5.83Fe0.14Ag0.01)Σ5.98Mo1.03(Sn0.54Sb0.41Te0.03Bi0.02)Σ1.00(S7.85Se0.15)Σ8.00, and that of the Sb-Bi variety from the Kawazu mine is: (Cu5.84Fe0.14Ag0.01)Σ5.99Mo1.03(Sn0.82Sb0.11Bi0.l0Te0.04)Σ1.07(S7.80Se0.12)Σ7.92. The theoretical formula of hemusite is Cu+ 4Cu2+ 2MO4+Sn4+S8, whilst the most probable formula of the Iriki hemusite is Cu+ 4.5CU2+ 1.5Mo4+Sn4+ 0.5Sb5+ 0.5S8, with Sb5+ substituting for Sn4+ and forming (SbS4)3− tetrahedra as might be expected, given that the metal to sulphur ratio is 1, and given the sphalerite-like structure of the mineral. However Bi3+ cannot be so accommodated, resulting in a deficiency in (S + Se) for Kawazu hemusite. Reflectance spectra for both are compared with those of the tungsten analogue (compositional) of hemusite, kiddcreekite. The relationship between hemusitesensu stricto and these newly reported varieties is discussed in terms of simple and coupled chemical substitutions, and inferences are drawn on the valency of Sb, Bi, Mo and Cu in the hemusite structure.

Description / Table of Contents: Zusammenfassung Potosiit and Incait wurden in Erzen der Hoei-Mine, Oita Präfektur, Japan, nachgewiesen.
Notes: Summary Potosiite and incaite have been found at the Hoei mine, Oita Prefecture, Japan. Potosiite occurs as tabular crystals (3 cm x 2 cm in size), and is associated with arsenopyrite, pyrrhotite, stannite, jamesonite and pyrite, whereas incaite occurs as polycrystalline aggregates rimming galena or as hair-like masses in a matrix of kutnahorite. X-ray powder data reveal that these minerals are potosiite and incaite, respectively. Electron-microprobe analyses of potosiite and incaite gave the general formula (Pb, Sn)2+ 6+xFeSb2Sn4+ 2S14+x, with x 〈 2. Potosiite is homogeneous, whereas incaite, in contact with galena, is quite heterogeneous. Reflectance data for potosiite show that its bireflectance is one of the lowest among opaque minerals with a sheet-like structure.
