Summary A dialysable substance has been cleaved from bovine retinal gangliosides by the action of receptor-destroying enzyme from V. cholerae. Crystallisation and paper chromatography showed that the substance is N-acetylneuraminic acid. Susceptibility to enzymatic hydrolysis of the gangliosides was dependent upon methods of isolation and degree of purity. The action of mild acid hydrolysis produced an additional derivative of neuraminic acid; i.e., N-glycolylneuraminic acid. The identification of the derivative was based on its chromatographicR f value and its reaction with 2,7-dihydroxynaphthaline.

Zusammenfassung Aus Netzhaut-Gangliosiden des Rindes konnte durch Einwirkung von „Receptor-Destroying-Enzyme” aus Vibrio cholerae eine dialysierbare Substanz abgespalten werden, die nach der Kristallisationsform und der ehromatographischen Wanderungsgeschwindigkeit sich als N-Acetyl-neuraminsäure erwies. Reinigungsverfahren und Reinheitsgrad der Gangliosidfraktion beeinflußten in hohem Maße die Spaltbarkeit. Nach milder Säurehydrolyse ließ sich außerdem ein Neuraminsäure-Derivat nachweisen, das nach der Reaktion mit 2,7-Dihydroxy-naphthalin und der ehromatographischen Wanderungsgeschwindigkeit N-Glykolyl-neuraminsäure entspricht.
