    Notizen: An isolated rat brain preparation was perfused using glucose-free (=aglycemic) media. The high-energy phosphates, substrates of the glycolytic pathway, free atnino acids, acetylcholine as well as the intracellular distribution of hexokinase activity were determined in brain tissues. The EEG was evaluated visually. The levels of glycolytic substrates, glutamate, and glutamine in cortical tissue decreased after aglycemic perfusion, whereas the aspartate level increased and the GABA level remained unchanged. The high-energy phosphate content seemed to be unaffected for about 15 min of aglycemic perfusion and fell significantly after 20 min. The EEG of the isolated brain changed rapidly after starting aglycemic perfusion and became isoelectric after 12–15 min. Hyperglycemic perfusion (35 mmol glucose per liter perfusion medium) did not alter the energy metabolism of the isolated brain. The breakdown of cerebral energy metabolism and of EEG activity was postponed when thiopental was added to the perfusion medium. The soluble hexokinase activity measured in cortical tissue was reduced after aglycemic perfusion and was enhanced after thiopental. Hyperglycemic perfusion did not influence the intracellular hexokinase distribution. The acetylcholine level in the striatum of the isolated rat brain was significantly decreased by aglycemia and was increased in hypothalamus by thiopental. It was suggested that hexokinase bound to the mitochondrial membrane may play an important role in the relationship of energy metabolism and neuronal activity.

    Notizen: Human chorionic gonadotrophin was given every other day to 8 women during 11 luteal phases and their excretion of oestrone and pregnanediol compared with the excretion of these steroids during an untreated luteal phase. Some of these subjects were also given chorionic somatomammotrophin while being treated with chorionic gonadotrophin. It was found that treatment with chorionic gonadotrophin resulted in an increase of ovarian hormone excretion and that the steroid levels remained elevated while the onset of menstruation was postponed. A decline in steroid excretion and menstruation eventually occurred in spite of continued treatment with chorionic gonadotrophin. There was no evidence that the administration of chorionic somatomammotrophin had any effect on ovarian steroidogenesis.

    Notizen: Abstract The formalism for fragment angular distributions in photofission with unpolarized and linearly polarized photons is summarized. First experiments on232Th using linearly polarized bremsstrahlung are described. The results for the analyzing power show for the first time in a model independent way that the low energy photofission can be explained byelectric dipole excitation. The possibilities of this new method are discussed, in particular when using monoenergetic tagged photons, which are available soon at new high duty cycle electron accelerators.

    Notizen: Summary The isolated perfused rat brain was used for a comparative study of the effects of promazine, imipramine, monodesmethyl promazine and desipramine on cerebral energy metabolism. After perfusion for 30 min or 1 h the brain levels of the following substrates and metabolites were estimated: P-creatine, creatine, ATP, ADP, AMP, glycogen, glucose, glucose-6-P, fructose diphosphate, dihydroxyacetone-P, pyruvate, lactate, α-ketoglutarate, and ammonia. Drug concentrations of 5·10−6 M and 10−5 M in the perfusion medium caused a significant decrease of glucose-6-P alone. When the drug concentration was raised to a toxic range (10−4 M), reflected in the EEG by the pattern of secondary discharges, an accumulation of P-creatine and glucose and a decrease of glycogen, glucose-6-P and ammonia occurred; the lactate/pyruvate ratios remained unchanged. As there were no qualitative differences between the effects of the investigated neuroleptics and antidepressants on cerebral metabolism, these effects might be unspecific and not correlated with the pharmacological action of the drugs.

    Notizen: Summary The isolated perfused rat brain was used for a comparative study of the effects of chloral hydrate and trichloroethanol on cerebral energy metabolism. After a perfusion period of 30 min the brain levels of the following substrates and metabolites were measured spectrophotometrically: P-creatine, creatine, ATP, ADP, AMP, glycogen, glucose, glucose-6-P, fructose diphosphate, α-glycero-P, dihydroxyacetone-P, pyruvate, lactate, glutamate, α-ketoglutarate and ammonia. Furthermore, the concentration of chloral hydrate and trichloroethanol in the isolated brain and in the perfusion medium was measured colorimetrically. Little more than 10% of chloral hydrate in the isolated brain and in the perfusion medium were reduced to trichloroethanol. In intact animals there were about 70% of chloral hydrate transformed. Chloral hydrate and trichloroethanol caused an accumulation of P-creatine, no change in the lactate/pyruvate ratio, an increase of the glucose concentration and a decrease of glucose-6-P level in the isolated brain. The rise of brain glucose level was more pronounced after trichloroethanol than after chloral hydrate. The effects of chloral hydrate and trichloroethanol on brain glucose and glucose-6-P levels suggest an inhibition of brain hexokinase activity by these drugs.

    Notizen: Abstract An isolated perfused brain preparation is regarded as offering some important advantages over intact animals or tissue slices for studying drug effects on the CNS. The rat is by far the most suitable laboratory animal for this technique because of low cost, ease of preparation and extensive literature available for comparative purposes. In this paper various preparation techniques and perfusion systems for an isolated rat brain are reported. Investigations are presented proving the viability of the isolated perfused rat brain for more than seven hours and its suitability for studies on cerebral metabolism. Until now this preparation has been successfully used for pharmacological investigations concerning the drug effects on cerebral energy metabolism, on the EEG and on the biogenic amines in the CNS. The results further demonstrate that the use of an isolated perfused rat brain affords the possibility of clarifying pharmacological problems which has not been possible using conventional methods. In addition, this preparation should become a very useful tool for studies on pharmacokinetics, on brain circulation or on brain oxygen consumption.
