Description / Table of Contents: Summary The correlation between renal diseases and the argentaffine system of the gastrointestinal tract was investigated. In 5 patients with renal lesions but without uremia (solitary or multiple cysts, hypernephroid renal carcinoma, and renal infarcts) the number of argentaffine cells was found to be significantly higher than in a control group of 30 patients without any renal lesions. Two further patients with hypernephroid renal carcinomas had also a carcinoid tumor. At least 4 of 12 patients with a malignant carcinoid syndrome showed multiple or solitary cysts of the kidneys. This combination is estimated to be more than accidental. Previous results of studies of the action of serotonin on the kidney disclose a change in the renal blood flow (vasoconstriction). Whether the renal changes and the increase of argentaffine cells (diffuse hyperplasia or carcinoid as „nodular hyperplasia”) can be correlated with serotonin-metabolism cannot be decided at present from the scanty material studied, and from the paucity of parallel studies on the metabolism of serotonin.

Description / Table of Contents: Summary A report is given on the number of argentaffine cells in an average of 20 specimens per case of the entire gastrointestinal tract (in the stomach per 100 visual areas, in the intestine per 100 longitudinal crypts) of 70 patients, who died of various diseases or accidents. Since autolysis causes rapid loss of the argentaffine granules, it is important to use only tissue fixed within 3 hours after death. The average value for the control group (14 accident cases and 25 patients with cardiovascular diseases) came to 100 argentaffine cells per 100 crypts or visual areas. High values were found in the oral parts, as in the juxtapyloric part of the stomach and duodenum, and in the rectum. Within a fairly broad range of distribution of ±40%, the number of argentaffine cells in various pathological situations was strikingly constant. Thus, no significant differences could be found in patients with liver cirrhosis, primary malignant tumors of gastrointestinal tract, and kidney diseases leading to uremia when compared with the control cases. A significant increase in the number of argentaffine cells (by about 50%) could be demonstrated in patients with kidney diseases without uremia, and extremely high rates were found in two cases with prolonged fasting. There was a significant decrease of about 50% in patients with extra-abdominal primary tumors. The main cause of this decrease was probably a non-specific effect on the intestinal mucosa.

Notes: Summary In 12 selected testicular biopsies of 12 boys 3 to 14 years old, with cryptorchid, abdominal, inguinal or ectopic testes, the shape of the deformed segments of the testicular tubules with intratubular bodies, the so called “ring-like tubules” was controlled in serial sections. These ring-like tubular formations are deformed parts of otherwise normal seminiferous tubules. They represent a hollow sphere with a central globular body. These spheres can be found at the blind end of a seminiferous tubule, at any place within the course of the tubules or at the branch-point of the tubules. Combinations of these malformations are rare. Structure, special forms and nomenclature of these spheric tubular formations are discussed.

Notes: Summary In 28 selected autopsy cases of male subjects 15 to 33 years old who died by accidents, suicide or severe systemic diseases, the so called hypoplastic zones present in one or both of the normally descended testes were examined. Four hypoplastic tubules in the testes of two different cases could be controlled in serial sections. The hypoplastic zones are parts of the normally developed seminiferous tubules. The diameter and the shape of the tubule formed by the tunica propria changes only little at the transition point into the hypoplastic zone. Within the hypoplastic tubule itself the seminiferous epithelium is replaced by a small solid cord with dense nuclei. This cord is coiled up within the tunica propria. Often the cord swells up and contains small concrements.

Notes: Summary Hearts of rabbits were isolated and perfused with Tyrode solution containing 10 ng/ml (−)-α-methyl noradrenaline. The uptake by the heart ofα-methyl-noradrenaline was calculated by estimating the amount of amine removed from the perfusion fluid while passing through the heart. In control experiments the mean removal ofα-methyl noradrenaline was 43,7 ± 1,6% of the amount infused. The rate of removal remained constant throughout the time period of the experiment, i.e. 20 min. Sympathomimetic amines were found to inhibit the uptake of α-methyl noradrenaline. In order to investigate structure-activity relationships eight different amines were added to the perfusing medium at various concentrations. Dose-response curves representing the inhibition of α-methyl noradrenaline uptake were plotted as regression lines; the drug concentration which produced a 50% inhibition of the control uptake was calculated. From the results the following conclusions were drawn. 1. Introduction of a phenolic hydroxyl group in meta-position increases the inhibitory action. 2. Hydroxylation of theβ-carbon atom of the side-chain decreases the inhibitory action. 3. Methylation of theα-carbon atom of the side chain increases the inhibitory action. 4. Differences in the absolute configuration of a molecule affect the inhibitory action. Among sympathomimetic amines, the structural requirements of an uptake inhibition ofα-methyl noradrenaline appear to be similar to the structural requirements which, according to other authors, have to be satisfied for a comparable inhibition of noradrenaline uptake.

Notes: Summary Isolated rabbit hearts were perfused with Tyrode solution containing 10 ng/ml (-)-noradrenaline for 20 min. The uptake by the heart of noradrenaline was calculated by estimating the percentage of amine removed from the perfusion fluid during a single passage through the heart. Previous experiments have shown that the amount of noradrenaline removed from the perfusion fluid corresponds well to the amount retained by the heart. In control experiments, the mean removal (elimination) of noradrenaline was 50±1.7% of the amount infused as measured biologically and 46±2.2% as measured fluorimetrically. Ouabain was added to the perfusion fluid at concentrations ranging from 2.74·10−7 to 2.0·10−6 mol/l. The lowest dose caused a positive inotropic effect. With higher concentrations of ouabain there was a transitory increase in contractile force which was followed by a negative inotropic action, atrioventricular block and contracture of the myocardium. The noradrenaline uptake by the heart was neither affected by “therapeutic” nor by “toxic” concentrations of ouabain. On the other hand, almost complete inhibition of noradrenaline uptake was found after administration of 3·10−5 mol/l cocaine. It is concluded that in the isolated heart ouabain does not interfere with noradrenaline transport across the cell membrane although the concentrations of ouabain employed are known to inhibit active transport of sodium and potassium.
