    Notes: Abstract Two new members (Bsar1a and Bsar1b) of the Sar1 gene family have been identified from a flower bud cDNA library of Brassica campestris and their functional characteristics were analyzed. The two clones differ from each other at 14 positions of the 193 amino acid residues deduced from their coding region. The amino acid sequences of Bsar1a and Bsar1b are most closely related to the Sar1 family, genes that function early in the process of vesicle budding from the endoplasmic reticulum (ER). The sequences contain all the conserved motifs of the Ras superfamily (G1–G4 motifs) as well as the distinctive structural feature near the C-terminus that is Sar1 specific. Our phylogenetic analysis confirmed that these two clones can indeed be considered members of the Sar1 family and that they have a close relationship to the ARF family. The Bsar1 proteins, expressed in Escherichia coli, cross-reacted with a polyclonal antibody prepared against Saccharomyces cerevisiae Sar1 protein. It also exhibited GTP-binding activity. Genomic Southern blot analysis, using the 3'-gene-specific regions of the Bsar1 cDNAs as probes, revealed that the two cDNA clones are members of a B. campestris Sar1 family that consists of 2 to 3 genes. RNA blot analysis, using the same gene-specific probes, showed that both genes are expressed with similar patterns in most tissues of the plant, including leaf, stem, root, and flower buds. Furthermore, when we placed the two Bsar1 genes under the control of the yeast pGK1 promoter into the temperature-sensitive mutant yeast strain S. cerevisiae Sec12-1, they suppressed the mutation which consists of a defect in vesicle transport. The amino acid sequence similarity, the GTP-binding activity, and the functional suppression of the yeast mutation suggest that the Bsar1 proteins are functional homologues of the Sar1 protein in S. cerevisiae and that they may perform similar biological functions.

    Notes: A series of 2,3-O-carboxymethylcelluloses (CMC's), which are regioselectively substituted at the C-2 and C-3 position, were prepared and their water solubility was examined. It was found that the lower limit for the degree of substitution (DS) value of water-soluble 2,3-O-CMC is about 0.3. This value was almost the same as that of CMC prepared in a slurry of isopropyl alcohol/water with isopropyl chloroacetate and sodium hydroxide, showing that the uniform alkylation is rather important to convert cellulose into water-soluble derivatives.

    Notes: The reinforcement of a natural rubber compound by various surface-modified precipitated silicas was compared. Compound physical properties were determined for two silicas differing in surface area and were used as controls to evaluate these silicas after surface modification by using either a bifunctional organosilane coupling agent (γ-mercaptopropyl-trimethoxysilane) or a new surface modification process. This new process is based on the in situ polymerization of organic monomers solubilized inside surfactant bilayers that are adsorbed onto the silica surface to afford silicas modified with styrene-butadiene and styrene-isoprene copolymers. Both surface modification processes afford materials that dramatically increase the compound cure rate, thereby significantly reducing T90 cure times, while also improving tensile properties, tear strength, abrasion resistance, and compression set of the cured compound. The silane-modified silica gives a higher flex-cracking resistance than do the silicas modified by the in situ polymerization of organic monomers, whereas these latter silicas significantly increase rebound resilience and offer greater overall improvements in rubber compound performance. The rubber compound physical properties obtained using the modified, higher surface area Hi-Sil® 255 silica are generally improved relative to those obtained using the modified Hi-Sil® 233 silica. © 1996 John Wiley & Sons, Inc.

    Notes: The crystal structure of poly(aryl ether biphenyl ether ketone ketone) (PEDEKK) was determined to comprise a two-chain orthorhombic unit cell with dimensions a = 0.778 nm, b = 0.606 nm and c = 2.375 nm by using wide-angle X-ray diffraction (WAXD). According to the orthorhombic system, the 12 reflections of this polymer were indexed. The crystallite size increases with increasing the crystallization temperature. The results of the degree of crystallinity (Wc,x) calculated from WAXD were compatible with those from density (Wc,d) and calorimetry (Wc,h) measurements.

    Notes: Structures of poly(ether ether ketone ketone)-poly(ether biphenyl ether ketone ketone) copolymers were studied by using small angle X-ray scattering and the one-dimensional electron density correlation function method. The results revealed that structures of the aggregated state of the copolymers depend closely on the biphenyl content (nb). When nb = 0.35, invariant Q, long period L, average thickness of crystal lamellae d̄, electron density difference ηc - ηa and degree of crystallinity Wc, x assume minimum values.

    Notes: Vacuum line kinetic studies of the reaction of p-toluenesulfonyl chloride and benzene or toluene, using aluminum chloride as the catalyst and dichloromethane as the solvent were determined at 25°C by means of gas chromatography. The reaction is first-order in arene, tosyl chloride, and in AlCl3 as catalyst. Noncompetitive results are kT/kB=22±7 with a product sulfone isomer distribution: ortho, 14±1%; meta, 4.3±0.2%; and para 82±1%. With hexadeuteriobenzene kH/kD was determined to be 1.8±0.1. Rate constant ratios and product isomer distributions were also determined competitively: with AlCl3, kT/kB=30±2; % ortho, 13±1; % meta, 4.0±0.5; % para, 84±3; with SbCl5, kT/kB=40±4; % ortho 10.3±0.4; % meta, 4.7±0.2; and % para, 85.0±0.5. The kT/kB ratio for AlCl3 and the meta sulfone product percentages for both AlCl3 and SbCl5 are considerably higher than those reported in the literature. NMR and Raman studies suggest a molecular complex between p-tosyl chloride and AlCl3, with coordination through oxygen as the dominant species and the probable electrophile in CH2Cl2.
