Notizen: Abstract The Mediterranean fruit fly, Ceratitis capitata, is a major pest of fruits and vegetables in the world. With the help of an extensive trapping program, the continental United States has remained free of established medfly populations. In an effort to increase trapping efficiency, controlled-release polymeric panels with trimedlure and ceralure were developed in conjunction with the development of a new trap, the C&C trap. This study contrasts the effectiveness of different panel formulations with the standard trimedlure polymeric plug and describes the use of the panels in the C&C trap. Field tests in Hawaii using released sterile flies showed consistently higher male medfly catches with panels in C&C traps than with the standard trimedlure polymeric plug in Jackson traps.

Notizen: Abstract Ceralure or trimedlure polymeric TNO panels in C&C traps were compared for attractancy by the capture of male Mediterranean fruit flies, Ceratitis capitata (Wiedemann). This study contrasts the effectiveness of ceralure with trimedlure, the former of which is an iodo-analog of trimedlure. Field tests in Hawaii with released flies showed that the active ceralure B1 isomer in a commercial mixture of ceralure isomers consistently caught as many male flies as active trimedlure C isomer in a commercial mixture of trimedlure isomers at one-half the molar quantity of trimedlure C. These panels caught 6.4 and 5.2 times, for ceralure and trimedlure, respectively, more than the standard 2 g trimedlure AgriSense aged plug over a 26-week period. The persistence of ceralure is demonstrated by residual analyses of aged panels that revealed the presence of 2.75 times more ceralure than trimedlure in panels after 130 days of field exposure.

Notizen: A quantitative method for loading nanogram samples onto field desorption emitter wires is described. Sample consumption is reduced and problems with emitter wettability are overcome by freezing microliter droplets onto the emitters. Using this technique, good spectral data are obtained from 10 to 50 ng of material. Field desorption sensitivity measurements are reported for neomycin B, adenosine and cholesterol. Evaluation of the nonquantitative dipping technique previously employed reveals poor and nonreproducible sample loading efficiency.

Notizen: Because of poor field ionization sensitivity or an insufficient number of peaks, conventional reference materials are inadequate for field desorption mass spectrometry. Hexakis (multifluoroalkoxy)cyclotriphosphazenes satisfy a growing need for high molecular weight reference compounds amenable to either electron impact or field ionization. In comparison with tris(perfluoroalkyl)-s-triazine standards, the substituted phosphazenes offer greater volatility, easier synthesis and better mass spetral characteristics. Phosphazene mixtures, in particular, cover a broad mass range in either ionization mode, creating new measurement capabilities for ions at high masses.

Notizen: Isolation of a pentaene macrolide antibiotic (NSC-277813) from Streptomyces griseus (FCRC-21) fermentation broth is described. Using field desorption mass spectrometry and high resolution field desorption mass spectrometry on the intact and derivatized antibiotic and degradation products, the antibiotic was identified as fungichromin. The application of field desorption mass spectrometry in the identification of polyene antibiotics is discussed.

Notizen: In phototrophic culture of Euglena gracilis, good synchrony was found only under rather restricted programs of light-dark cycles, and rather narrow ranges of temperature and light intensity, when cultures were flushed with air fortified with adequate amounts of CO2. When flushed with air alone, CO2 was found to be limiting, and while cell divisions were rhythmic, less than a doubling of cell number occurred in division bursts. With air as gas phase, rhythmic division activity was maintained over wide ranges of temperature, light intensity, and the ratio of light:dark in a given program; all these factors affected the amplitude of the division burst, however.

Notizen: Growth characteristics of Euglena gracilis Z as functions of culture pH, CO2 tension, temperature, and lighting regime were investigated. The results are consistent with the possibility that cell division is preceded by a lowered intracellular pH. Also consistent with this possibility is the finding that division rhythmicity can be induced by periodic changes in CO2 tension. It is suggested that the rhythmicity is induced by changes in intracellular pH produced by carbonic acid.

Notizen: Visible light of moderate intensity causes two and perhaps three types of division inhibition in Euglena gracilis are related cells. Fluorescent light causes a general inhibition of growth and division which is temperature-dependent. Pigmentation or complex organic media partially lifts this inhibition. A second type of inhibition, which is transient, can be caused by either fluorescent or incandescent light, and is found with an irreversibly bleached strain of Euglena grown on a limiting concentration of acetate; this inhibition could not be demonstrated in cells grown on limiting concentrations of glucose.

Notizen: In the dark, growth rates of Euglena gracilis were independent of culture pH between the limits of three to eight. Visible light of moderate intensity inhibited growth rates, with the degree of inhibition being markedly pH dependent. The most severe inhibition was observed at pH 4 to 5, with little or no inhibition at pH 3 or above pH 6.8 at the light intensity used (400 foot-candles).

Notizen: Visible light of moderate intensity (1200 ft-cd) can severely inhibit cell division of a non-photosynthetic mutant of Euglena gracilis when growth is supported by butanol, ethanol, or fumarate as sole carbon source. The degree of inhibition is pH dependent, being greatest at pH 4 to 5. A wide variety of other carbon sources permitted growth in the light without inhibition.
