    Notes: Ovarian morphology and behavioral relationships were studied in a group of nine miniature pigs with a characteristically small litter size (∼5) and an average coefficient of inbreeding of 0.39. The first day of standing estrus was designated as day 0. Laparoscopy was used to evaluate and photograph ovarian activity on days 1, 5, 11, 17, and day 2 of the subsequent estrous cycle. Mean (±SEM) duration of estrus and the estrous cycle was 3.1±0.2 and 22.6±0.3 days, respectively. An average of 9.0±0.4 vesicular follicles developed/estrous period and 96% of the follicles showed morphologic evidence of ovulation resulting in a mean of 8.6±0.3 corpora lutea (CL) per animal. There was no significant correlation between the degree of inbreeding and number of vesicular follicles (r=0.27) or CL (r=0.28) developing/cycle within the experimental group. Mature preovulatory follicle and CL size ranged from 6-12 mm and 8-12 mm in diameter, respectively, and 22.5% of the newly formed CL contained distinct postovulatory stigmata. These data indicated that 1) temporal relationships of sexual behavior and gross ovarian morphology in the miniature pig were similar to descriptions previously reported for the standard pig, and 2) the reduced litter size characteristic to this particular strain is, at least in part, due to decreased number of vesicular follicles developing during the estrous cycle.

    Notes: Studies on isolated perfused tubules of the tiger salamander (Ambystoma tigrinum) have shown that the distal nephron is heterogeneous with respect to function (Stoner, 1977). In this study, the initial portion of the distal tubule (diluting segment) exhibited a voltage, positive in the lumen, and a net absorption of chloride. Since the chloride was transported against an electrochemical gradient, its transport was active. More distad, the junctional segment exhibited a lumen-negative voltage and sodium, rather than chloride, was transported actively. More recently Delaney and Stoner (1981) have demonstrated in vitro that the collecting duct of this species also has a lumen-negative voltage which is probably associated with active sodium reabsorption.The primary objective of the present paper was to correlate the morphology of the diluting and junctional segments of the Ambystoma distal tubules with the physiologic data from the same isolated perfused tubules. The results indicate that the morphological heterogeneity previously demonstrated in distal tubules of Necturus exists with respect to both structure and function in Ambystoma. The cell types found in the amphibian distal nephron appear to be homologous to those seen in the mammalian nephron.

    Notes: Buccopharyngeal epithelium covering gill arches and gill rakers of the fathead minnow was studied by light microscopic, scanning, and transmission electron microscopic techniques. Mature mucous cells in goblet pattern and nonmucus containing cells were in the apical one-third of the tissue. The latter cells contributed to a surface microridge system which overlapped apices of goblet cells. The bottom of the epithelium was comprised of a continuous row of darkly stained basal epithelial cells. In this region, two to three epithelial cells of similar staining characteristics were piled up forming apical columns which partially encircled nests of lightly stained cells. A basal lamina and thick basement lamella of about 20 plies of orthogonally arranged collagen supported the epithelium. Numerous taste buds were seen in gill arches and rakers. Taste bud cellular components included marginal cells, light receptor cells, dark receptor cells, and basal cells. These were identical in all taste buds. Taste bud surface morphology differed between gill arch and raker. Pores of the former were depressed, while those of the latter were raised. Thick microvilli of taste pores were apical extensions of light cells, while smaller, more numerous microvilli were projections from dark cells.

    Notes: Mouse liver cells in primary culture were evaluated by highresolution light microscopy (HRLM) and transmission electron microscopy (TEM). Cells after 2 hours of culture in L-15 medium supplemented with 10% fetal bovine serum were spherical in shape, and were either individual or in small clusters of up to ten cells. Following 1 day in culture, hepatocytes were flattened and usually found in groups. Bile canaliculus-like structures were apparent between hepatocytes. Tight junctions and desmosomes were also present along adjacent plasma membranes. Autophagic vacuoles were seen within the cytoplasm. After 2 days in culture, hepatocytes appeared more elongated and flattened than in earlier sampling periods. Both autophagic and clear vacuoles were seen in the cytoplasm. Mitochondria were present in a variety of shapes and sizes. Small bundles of microfilaments were frequently seen in the basal region of cross-sectioned cells. From the fourth until the eighth day in culture, hepatocytes displayed further progression of the morphologic changes seen after 2 days. Nuclear elongation and the projection of cytoplasmic pseudoinclusions into the nucleus were also evident after 4 days. Cytoplasmic and nuclear changes were eventually observed in all hepatocytes by the eight day of culture. DNA synthesis in the cells during culture was investigated by autoradiography. The percentage of S-phase labeled cells was 0.1% after 1 day of culture. The labeling index increased to 1.02%, 3.14%, and 5.88% after 2, 4, and 6 days of culture, respectively. Synthesis of albumin by the liver cells was also detectable during the first 8 days of primary culture. A gradual drop in albumin synthesis was noted with increased time in culture. The percentage of hepatocytes that histochemically stained for gamma glutamyl transpeptidase (GGT) progressively increased from 0.01% of the cells after 2 hours culture to 3.14% of the cells after 8 days of culture.

    Notes: Glycoprotein components of epidermal mucous cells in control (pH 6.8) and acid-stressed (pH 5.8, 4.8, 4.0) brown bullhead catfish were studied by histochemical and autoradiographic methods. Following exposure to acid, increased numbers of epidermal mucous cells were noted in all tissue sites studied, particularry dorsal and ventral skin. Mucous cells of control fish and acid-stressed (pH 5.8) fish contained a mixture of neutral and acidic mucosubsf ances, the latter including both sialomucins and sulfomucins, with sialomucins predominating. An apparent shift toward increased sulfomucin production was observed in skin from fish exposed to pH 4.8 and 4.0. In addition, autoradiographic studies using 35S showed increased labeling of mucous cells in acid-exposed (pH 4.0) epidermis, offering further support for a shift toward sulfomucin production in acid-stressed fish. Absence of appreciable sulfomucin production in younger fish (〈1 year old) suggested that this shift may be partly age-dependent. The functional significance of increased sulfomucin production in acid-stressed epidermal mucous cells is uncertain but may be related to maintenance of ionic equilibrium.

    Notes: A 10,000-dalton calcium-binding protein (10-kd CaBP) has been described in the placentae and yolk sacs of rats and mice. This protein is similar or identical to vitamin D-dependent intestinal CaBP and these proteins have been implicated in the molecular mechanisms of intestinal calcium absorption and transplacental calcium transport. Using an antiserum to the purified 10-kd rat intestinal CaBP, the localization of CaBP in the 16-17-day mouse yolk sac and placenta was studied immunocytochemically with peroxidase-antiperoxidase labelling and quantified by radial immunodifussion. A high concentration of immunolabelling was observed in the endodermal cells of the intraplacental yolk sac lining the sinuses of Duval. The columnar endodermal cells lining one side of the endodermal sinuses adjacent to fetal vessels contained most of the immunoreactive label. Quantitation by radial immunodiffusion demonstrated a 5.5-fold higher concentration of CaBP in the portion of the placenta containing most of the intraplacental yolk sac than in the maternal half of the placenta. This demonstration of a 10-kd CaBP within the intraplacental yolk sac suggests this protein functions to facilitate placental calcium transport and is the first study which directly supports the hypothesis of a functional role for the sinuses of Duval in calcium transport.

    Notes: The third ventricular walls and floor of male and female mink (Mustela vison) were analyzed with comparative light microscopy, transmission and scanning electron microscopy. Distinct regional fine structural variations were noted in the appearance of the dorsal, middle third and lower portions of the ventricular wall and floor. These variations were characterized by an abrupt decrease in the density of ciliated ependymal surfaces in the middle one-third of the ventricular wall. The ventral walls and floor of the lateral and infundibular recesses displayed smoother contours with numerous bleb-like protrusions and blunt villiform processes. Occasionally multipolar neuron-like cells were observed to lie upon the floor of the infundibular recess. Speculation is raised relative to the differing functional capacities of these various ventricular areas.

    Notes: A method was developed to alter surgically the bursa of the bitch to allow sequential observation of the ovary by means of laparoscopy. Adult anesthetized female dogs of three breeds underwent laparoscopic examinations in which each natural bursa slit was lengthened 1.0-1.5 cm using laparoscopic hook scissors in conjunction with electrocautery. The ovary could be effectively and repeatedly exposed for laparoscopic viewing through the lengthened bursa slit. Neither Laparoscopic surgery nor frequent routine laparoscopic examinations of the reproductive organs had an effect on subsequent ovarian activity. All dogs subjected to the laparoscopic ovarian exposure technique exhibited estrus behavior, follicle development, ovulation and corpus luteum formation.Preovulatory follicular development was detected as early as 11 days before ovulation with the most rapid follicular maturation occurring two to three days before ovulation. Follicular collapse and extensive hemorrhage at the rupture site during ovulation did not occur. Release of the cumulus mass from a vesicular follicle was observed in one bitch. The cumulative mass adhered to the follicular wall for an indefinite time following ovulation.The structural size of the bitch corpus luteum was maximal four to eight days following ovulation. Corpora lutea remained visible on the ovarian surface for 16 weeks. No differences in cyclic ovarian morphology were observed among the three breeds of dogs studied.

    Notes: The effects of acid water on alcian blue-periodic acid Schiff's stainable mucosubstances of epidermis from theanterior abdominal wall of brown bullhead catfish, Ictalurus nebulosus, were studied. Standard morphometric techniques were used to determine mucous cell volume density (MCVv) and number density (MCNv) of control (pH 6.8-7.0) and acid-stressed (pH 5.7-6.15) fish. Mucous cell volume density was significantly increased (P 〈 0.005) in fish which had been exposed to acid-stress for five days. Since increase in volume density can arise from either an increase in the size of individual mucous cells (hypertrophy), an increase in the number of mucous cells per unit volume (hyperplasia), or a combination of the two, average cell data were computed. No significant difference in cell diameter or cell volume between treated and control groups was seen. However, the number of mucous cells in acid-stressed fish was nearly twice (P 〈 0.005) that of controls. These results demonstrated that acid-stress resulted in increased number but not volume of mucous cells (i.e., hyperplasia without hypertrophy).

    Notes: The present study correlates cell body and nucleolar sizes for dorsal root ganglion cells in the rat. To do this, we measured cell body areas and calculated their diameters and measured nucleolar diameters for 719 cells. These data indicate that there is a correlation in that increasing cell size is associated with increasing nucleolar size. However, there is considerable variability of cell body size for each nucleolar diameter and vice versa. Nevertheless, when nucleolar diameters are grouped, the function In D = 1.687 + 0.334 dr, where D is the diameter of the cell body and dr the rounded diameter of the nucleolus, produced an almost straight line. Thus this formula provides a good estimate of the relation of nucleolar and cell body sizes for dorsal root ganglion cells of the rat. In addition, estimates of the variances of cell body size at each nucleolar diameter are provided.
