Die Strukturaufklärung gelang durch eine Kombination chemischer, radiochemischer und physikalischer Methoden mit Gaschromatographie und Massenspektrometrie. Im Organismus bilden sich die Prostaglandine aus essentiellen Fettsäuren. Die Prostaglandine stimulieren die glatte Muskulatur und wirken auf den Fettstoffwechsel ein.

    Notes: This paper describes the use of electron ionization mass spectrometry for structural characterization of a mixture of glycosphingolipids from a single human kidney and presents mass spectrometric evidence for the presence of an eleven-sugar glycolipid. A large amount of structural information is obtained from minor glycolipid constituents having long sugar chains by a fractional evaporation in the ion source, achieved by steadily increasing the ion-source temperature.

    Notes: Negative electrospray ionization mass spectrometry (NESI-MS) of a series of native gangliosides containing one, two or three sialic acid groups, sulphatide-I and cholesterol 3-sulphate was undertaken, using a magnetic sector instrument. The results show that NESI-MS of gangliosides leads to the formation of multiple-charged molecular ions of the type [M — H]-, [M — 2H]2- and [M — 3H]3-. The number of charges appears to be correlated to the number of sialic acid groups present. NESI-MS of sulphatide-I predominantly gave ions of type [M — H]- whilst cholesterol 3-sulphate gave a more complex ionization pattern. The sensitivity of the NESI-MS system was enhanced by using water+methanol, 9:1 (v/v) as solvent, containing 300 pmol/μL of NH4HCO3. The application of NESI-MS to these compounds is discussed.

    Notes: Mass spectrometry has been applied for a detailed structural characterization of glycosphingolipids. Methylated and methylated plus reduced glycolipids (amide groups of ceramide and amino sugars were reduced to the corresponding amines) were found the most useful derivatives. Molecular weight ions [M - 1] were not obtained for methylated lipids with more than four sugar units, but were easily obtained for reduced derivatives, so far with six sugars. Spectra of reduced derivatives showed very abundant ions containing the complete carbohydrate and the fatty acid, allowing a conclusion to be made concerning the carbohydrate composition (ratio of hexose and hexosamine and sialic acid). In addition, conclusive information was obtained for the carbohydrate sequence, including branching of the chain. By a combined use of both derivatives the long chain base and fatty acid composition was also evident. Spectra are discussed for a synthetic monoglycosylceramide, the Forssman glycolipid hapten (pentaglycosylceramide) and two gangliosides (a triglycosylceramide and a pentaglycosylceramide). The mass spectrum of the Forssman glycolipid provides the first conclusive evidence for two amino sugars in the terminal disaccharide of the molecule.

    Notes: The major mammalian retinal ganglioside was analysed by mass spectrometry. Three types of derivatives were used, the fully methylated, the methylated and reduced and the methylated, reduced and trimethylsilylated compound. The mass spectra unambiguously showed the ganglioside to be a sialyl-sialyl-dihexosyl-ceramide. This is the first conclusive proof that the two N-acetylneuraminic acids are bound to each other constituting the terminal disaccharide. The major species contained sphingosine and stearic acid.

They are C20-carboxylic acids contining one five-membered ring, one keto group, two or three hydroxy groups, and one to three C=C double bonds. Their structures were elucidated by chemical, radiochemical, and physical methods in combination with gas chromatography and mass spectrometry. The prostaglandins are biosynthesized from essential fatty acids. By their action smooth muscles are stimulated and lipid metabolism is affected.
