Notes: Recent developments in the use of negative ion detection in GC-MS have indicated the potential for highly sensitive analyses for certain compounds. This paper describes the application of the techniques to the assay of Nitroglycerin and its metabolites yielding sensitivity to two femtograms. Because of the low levels of nitroglycerin in biological media such sensitivity is highly desirable if not essential for meaningful measurements. In addition to sensitivity the method is shown to yield extremely good selectivity contributing further to the credibility of quantitative results.

Notes: An optimized HPLC method for the quantification of metoclopramide (MCP) in human plasma and urine is described. MCP and internal standard are extracted from alkalinized substrate into diethyl ether and back-extracted into dilute acid. The analytes are separated with a ternary mobile phase at cyanopropyl-silica and detected at 312 nm (UV detection). The lower limit of quantification is 0.5 ng/ml in plasma and 50 ng/ml in urine. Optimization of extraction, chromatography, and detection is discussed. The method is selective to numerous common drug substances with excellent accuracy and precision data. After validation, the method is applied to the samples of a pharmacokinetic study. Pharmacokinetic parameters indicate the need for a sophisticated method as tool for optimization of metoclopramide formulations.

Description / Table of Contents: Surface Treatment of Carbon Fibres and Resulting Composite PropertiesIn composites carbon fibres are used as reinforcing fibres with thermosetting and thermoplastic resins as martices. These carbon fibres differ strong in their micro-structure and therefrom in fibre properties. To achieve sufficiant composite properties special carbon fibre surface treatment methods are necessary. This paper describes a systematic study on oxidative surface treatment of carbon fibres by wet-, dry- and anodic oxidation. Further investigations by matrix variation show us the influence of matrix strength on the mechanical composite properties. Finally it is shown that in case of impact load composite fracture behaviour is controlled only by the fibre itself.

Notes: A method is described for the determination of acetylsalicylic acid and salicylic acid in human whole blood and plasma which uses liquid chromatography with UV detection. The enzymatic degradation of acetylsalicyclic acid in blood and plasma is examined. Addition of potassium fluoride as enzyme inhibitor and deep freezing after plasma preparation allows storage of plasma for one week. A mixture of acetonitrile and diluted acetic acid as mobile phase give the best peak shape. Traces of iron lead to slight peak broadening; addition of EDTA results in excessive tailing. The detection limit for acetylsalicylic acid and salicylic acid is about 250 ng/ml for a 1.0 ml primary sample. The method is sensitive enough for the monitoring of both drugs in bioavailability studies.

Notes: An automated specific and sensitive method for the simultaneous determination of isosorbide dinitrate and its metabolites isosorbide-2-mononitrate, isosorbide-5-mononitrate, isomannide mononitrate, and isoidide mononitrate at concentrations down to 0.5, 1, 3, 2, and 2 ng/ml, respectively, in human plasma is described. The procedure involves the extraction of the drug and its metabolites together with their internal standards isomannide dinitrate and o-nitrobenzyl alcohol from plasma with dicholromethane. The analysis is carried out by fused silica capillary gas chromatography using a 63Ni-electron capture detector. The method is applied to the pharmacokinetics of isosorbide dinitrate in humans.
