    Notes: Abstract A double indicator dilution technique for determining the mean cross-sectional area (CSA) of a blood vessel in vivo is presented. Analogous to the thermodilution method, dilution of hypertonic saline was measured by an electrical conductance technique. Because the change in conductance rather than absolute conductance was used to calculate CSA, pulsatile changes in shear rate of blood and conductance of surrounding tissues had no effect on the data. To calculate CSA from an ion mass balance, cardiac output was needed and estimated from the thermodilution curve using the same “cold” (hypertonic) saline injection. The mean CSA, obtained from this double indicator dilution method (CSAGD), was compared with the CSA obtained from the intravascular ultrasound method (IVUS) in 44 paired observations in six piglets. The regression line is close to the line of identity (CSAGD = −1.83 + 1.06⋅CSAIVUS, r = 0.96). The difference between both CSAs was independent of the diameter of the vessel, on average −0.99 mm2± 2.64 mm2 (mean CSAGD = 46.84 ± 8.21 mm2, mean CSAIVUS = 47.82 ± 9.08 mm2) and not significant. The results show that the double indicator dilution method is a reliable technique for estimating the CSA of blood vessels in vivo.

    Notes: Abstract A modified conductance method to determine the cross-sectional areas (CSAs) of arteries in piglets was evaluated in vivo. The method utilized a conductance catheter having four electrodes. Between the outer electrodes an alternating current was applied and between the inner electrodes the induced voltage difference was measured and converted into a conductance. CSA was determined from measured conductance minus parallel conductance, which is the conductance of the tissues surrounding the vessel times the length between the measuring electrodes of the conductance catheter divided by the conductivity of blood. The parallel conductance was determined by injecting hypertonic saline to change blood conductivity. The conductivity of blood was calculated from temperature and hematocrit and corrected for maximal deformation and changes in orientation of the erythrocytes under shear stress conditions. The equations to calculate the conductivity of blood were obtained from in vitro experiments. In vivo average aortic CSAs, determined with the conductance method CSA (G) in five piglets, were compared to those determined with the intravascular ultrasound method CSA(IVUS). The regression equation between both values was CSA (G) =−0.09+1.00·CSA(IVUS) r=0.97, n=53. The mean difference between the values was −0.29% · 5.57% (2 standard deviations). We conclude that the modified conductance method is a reliable technique to estimate the average cross-sectional areas of the aorta in piglets. © 1999 Biomedical Engineering Society.     Notes: The aim of this study was to investigate the bone response to three different types of titanium (Ti) alloys and hydroxyapatite (HA) coated titanium alloy by histological and histomorphometrical analysis. Therefore, implants made of these materials were inserted into the tibia of rabbits. Implantation times were 6 and 16 weeks. The histological evaluation included measurement of the amount of bone apposition and analysis of the bone reaction and interface characteristics around the implants. The results demonstrated no marked differences in bony reaction to the different implant materials. In addition, the HA coatings showed loss of thickness. © 1993 John Wiley & Sons, Inc.

    Notes: To evaluate the effect of surface treatment and surface microtexture on cellular behavior, smooth and microtextured silicone substrata were produced. The microtextured substrata possessed parallel surface grooves with a width and spacing of 2.0 (SilD02), 5.0 (SilD05), and 10 μm (SilD10). The groove depth was approximately 0.5 μm. Subsequently, these substrata were either left untreated (NT) or treated by ultraviolet irradiation (UV), radiofrequency glow discharge treatment (RFGD), or both (UVRFGD). After characterization of the substrata, rat dermal fibroblasts (RDF) were cultured on the UV, RFGD, and UVRFGD treated surfaces for 1, 3, 5, and 7 days. Comparison between the NT and UV substrata revealed that UV treatment did not influence the contact angles and surface energies of surfaces with a similar surface topography. However, the contact angles of the RFGD and UVRFGD substrata were significantly smaller than those of the UV and NT substrata. The dimension of the surface microevents did not influence the wettability characteristics. Cell culture experiments revealed that RDF cell growth on UV-treated surfaces was lower than on the RFGD and UVRFGD substrata. SEM examination demonstrated that the parallel surface grooves on the SilD02 and SilD05 substrata were able to induce stronger cell orientation and alignment than the events on SilD10 surfaces. By combining all of our findings, the most important conclusion was that physicochemical parameters such as wettability and surface free energy influence cell growth but play no measurable role in the shape and orientation of cells on microtextured surfaces. © 1995 John Wiley & Sons, Inc.

    Notes: It has been suggested that during wound healing microtextured surfaces can alter events at the interface between implant surface surface and surrounding tissues. To investigate this phenomenon, smooth and microtextured silicone rubber implants were implanted subcutaneously in rabbits for 3, 7, 42, and 84 days. The textured implants possessed parallel surface microgrooves and ridges with a width of 2.0, 5.0, and 10.0 μm. All grooves had a depth of approximately 0.5 μm. SEM observation showed fibroblasts, erythrocytes, lymphocytes, macrophages, fibrin, and collagen on all implant surfaces after 3 and 7 days. After 42 and 84 days only little collagen, a small number of fibroblasts, but no inflammatory cells were seen on the implant surfaces. The fibroblasts were not oriented along the surface grooves on all textured surfaces. Three-dimensional reconstruction of CLSM images and LM images showed no significant differences between the thickness of the capsules surrounding the smooth and those surrounding the microgrooved implants. In contrast, LM did show a significantly lower number of inflammatory cells and a significantly higher number of blood vessels in the capsules surrounding the microgrooved implants. Differences between the 2.0, 5.0, and 10.0 μm grooved implants were not detected. Our results concerning the capsule thickness suggest that the depth of our grooves was not sufficient to facilitate mechanical interlocking, but the cause for the observed differences in inflammatory response and number of blood vessels remains unclear. © 1997 John Wiley & Sons, Inc.     Notes: The microfilaments and vinculin-containing attachment complexes of rat dermal fibroblasts (RDF) incubated on microtextured surfaces were investigated with confocal laser scanning microscopy (CLSM) and digital image analysis (DIA). In addition, depositions of bovine and endogenous fibronectin and vitronectin were studied. Smooth and microtextured silicone substrata were produced that possessed parallel surface grooves with a groove and ridge width of 2.0, 5.0, and 10.0 μm. The groove depth was approximately 0.5 μm. CLSM and DIA make it possible to visualize and analyze intracellular and extracellular proteins and the underlying surface simultaneously. It was observed that the microfilaments and vinculin aggregates of the RDFs on the 2.0 μm grooved substrata were oriented along the surface grooves after 1, 3, 5, and 7 days of incubation while these proteins were significantly less oriented on the 5.0 and 10.0 μm grooved surfaces. Vinculin was located mainly on the surface ridges on all textured surfaces. In contrast, bovine and endogenous fibronectin and vitronectin were oriented along the surface grooves on all textured surfaces. These proteins did not seem to be hindered by the surface grooves since many groove-spanning filaments were found on all the microgrooved surfaces. In conclusion, it can be said that microtextured surfaces influence the orientation of intracellular and extracellular proteins. Although results corroborate three earlier published hypotheses, they do not justify a specific choice of any one of these hypotheses. © 1998 John Wiley & Sons, Inc.     Notes: The aim of this study was to investigate the biological and mechanical response of bone to titanium plasma-sprayed implants of different roughnesses. Three types of titanium plasma-spray coating were applied to beam-shaped implants: Ti2, Ti3, and Ti4, with a Ra of 16.5, 21.4, and 37.9 μm, respectively. An Al2O3 grit-blasted implant (Ti-un) with a Ra of 4.7 μ was used as a control. In total, 72 implants were inserted in the tibial cortical bone of nine adult female goats. These implants were evaluated histologically and mechanically 3 months after implantation. At the end of the experiment, of the 72 inserted implants, two implants (one Ti2 and one Ti4) were lost. Histological evaluation of the other retrieved implants revealed a uniform bone reaction for all implants. The unloaded plasma-spray coatings showed no signs of delamination at the implant-coating interface. Occasionally, particles of the Ti4 coating broke free and were found near the implant. Histomorphometry revealed no difference in bone contact for the different implants (P 〉 0.05). Furthermore, the push-out test showed no significant difference (P 〉 0.05). Linear regression showed no interaction between the push-out values and the roughness values (r = 0.5). On the basis of these results, it may be concluded that the used surface roughnesses did not lead to differences in bone response or mechanical attachment strength in goat cortical bone. © 1998 John Wiley & Sons, Inc.     Notes: During this study, microtechnology and plasma etching were used to produce gratings 1.0 (TiD01), 2.0 (TiD02), 5.0 (TiD05), and 10.0 μm wide (TiD10) into commercially pure titanium wafers. After incubation of rat dermal fibroblast (RDFs) on these surfaces for 3 days, the cells were observed with scanning electron (SEM), transmission electron (TEM), and confocal laser scanning microscopy (CLSM). Results showed that the RDFs as a whole and their stress fibers oriented strictly parallel to the surface pattern on the TiD01 and TiD02 surfaces. On the TiD05 and TiD10 surfaces, this orientation was not observed. In addition, TEM and CLSM demonstrated that the focal adhesion points (FAP) were located mainly on the surface pattern ridges. TEM revealed that FAP were wrapped occasionally around the edges of the ridges. Only the RDFs on both the TiD05 and TiD10 surfaces protruded into the grooves and possessed FAP on the walls of the grooves. Attachment to the groove floor was observed only on the TiD10 textures. Comparison of these results with earlier observations on microtextured silicone rubber substrata suggests that material-specific properties do not influence the orientational effect of the surface texture on the observed RDF cellular behavior.     Notes: RF magnetron sputter deposition was used to deposit thin calcium phosphate (hydroxyapatite) layers on titanium alloy substrate material. We determined the precise amount of calcium, phosphorus, oxygen, and hydrogen in these films by combining two ion-beam analysis techniques: Rutherford backscattering spectrometry (RBS) and elastic recoil detection (ERD). Variables were power level, partial oxygen pressure, or additional water vapor. Also examined was the influence of additional bias power to the substrates during deposition and the effect of annealing on the final composition of the deposited layers. Measurements showed that the Ca/P ratio decreased with increasing oxygen pressure or decreasing sputtering power. In addition, the Ca/P ratio increased when a bias was applied to the substrates. The O/P ratio of the films decreased with additional oxygen pressure but increased when additional water vapor was applied during deposition. All as-deposited films showed a higher hydrogen content than stoichiometric HA. The hydrogen content in the films deposited with water vapor was more than 7 times higher than in the films deposited under pure argon conditions. After annealing, the hydrogen content decreased to about 3.5 at % whereas in stoichiometric HA the hydrogen content amounts to 4.5 at %. After annealing, the oxygen concentration in the film also decreased. We assume that hydrogen disappears out of the film as H2O during annealing. For the targets used in these experiments, sputter deposition at 400 W, with additional water vapor and annealing at 500°C, produced films with a stoichiometry closest to that of hydroxyapatite. © 1998 John Wiley & Sons, Inc.
