    Notizen: Abstract Angiotensinogen (AGT) is a unique substrate of the renin-angiotensin system and fibronectin (FN) is an important component of the extracellular matrix. These play critical roles in the pathophysiological changes including cardiovascular remodeling and hypertrophy in response to hypertension. This study was performed to examine the regulation of AGT and FN gene in cardiac myocytes (CMs) and vascular smooth muscle cells (VSMCs) in response to mechanical stretch. Mechanical stretch significantly increased the AGT mRNA expression in CMs, while these stimuli did not affect FN mRNA levels. On the other hand, Mechanical stretch upregulated FN mRNA levels in VSMCs, whereas no increase in AGT mRNA levels was observed in response to stretch stimuli. An angiotensin II type 1 (AT1) receptor antagonist (CV11974) significantly decreased these stretch-mediated increases in mRNA level and promoter activity of the AGT and FN gene, whereas angiotensin II type 2 (AT2) receptor antagonist (PD123319) did not affect the induction. These results indicate that mechanical stretch activates transcription of the AGT and FN gene mainly via AT1 receptor-pathway in CMs and VSMCs. Furthermore, mechanisms regulating AGT and FN gene seem to be different between CMs and VSMCs.

    Notizen: Abstract A non-radioactive digoxigenin-labelled DNA method was used successfully to identify RFLP markers in 54 Triticum aestivum cv ‘Chinese Spring’ — Agropyron cristatum (2n=28, genome PPPP) P-genome monosomic addition lines. Southern analysis using a set of 14 DNA probes identifying each homoeologous chromosome arm, combined with two restriction enzymes HindIII and EcoRI, indicated that six A. cristatum chromosomes (1P, 2P, 3P, 4P, 5P and 6P) and five A. cristatum chromosome arms (2PS, 2PL, 5PL, 6PS and 6PL) have been individually added to the wheat genome. The added chromosomes of three lines were Agropyron translocated chromosomes. It was also found that two addition plants possessed an Agropyron-wheat translocation. These results showed that RFLP analysis using the set of assigned wheat probes was a powerful tool in detecting and establishing homoeology of alien A. cristatum chromosomes, or arms, added to wheat, as well as in screening the alien addition material. The creation of the monosomic addition lines should be useful for the transfer of disease-resistance genes from A. cristatum to wheat.

This paper presents an approach to extracting patient pharmacokinetic information from a set of longitudinal scintigraphic images and representing the kinetic information in parametric image form. In this approach, three or four pairs of simultaneously acquired anterior and posterior images, for internal dosimetry with iodine-131, are obtained at 24-h intervals. Each pair of anterior and posterior images is converted to a ”conjugate-view” image. The sequential conjugate-view images are then spatially registered to each other using a symmetric phase-only matched filter technique. With the registered images, the value at each pixel location, over time, is fitted to a monoexponential function. The resulting decay constant and intercept parameters are then represented as ”decay-constant” and ”intercept” images. Further analytical integration of the exponential function results in a ”cumulated-activity” image. Phantom studies demonstrated that the proposed method is accurate and reliable in the registration and kinetic data extraction of the longitudinal images. Clinical application of the approach to patients with thyroid carcinoma showed that regions of rapid uptake or clearance that would not otherwise be highlighted were easily identified and some regions with unexpected kinetics were observed. It is concluded that decay-constant, intercept, and cumulated-activity images provide a rapid and comprehensive visual assessment of patient pharmacokinetics. The technique may have a greater impact on the care of patients with extensive and distributed disease.
