    Notizen: SUMMARY: During the aging of beef, the external loading required to stretch sterno- mandibularis muscle to its fullest extent declines 5-10-fold from the maximum of 2 Kg/cm2 muscle attained at rigor onset. This loss of tensile strength is due to a weakening of the myofibrillarstructures at the junction of the I filaments with the Z discs of the sarcomeres. The Z discs undergo progressive changes and lose ground substance as aging proceeds. There is no obvious structural change in the I filaments during aging although a weakening in the association of the constituent proteins of these structures apparently occurs.

    Notizen: In an attempt to explain the paradoxical tenderizing, observed when muscle shortening exceeds about 40% of the initial excised length, the fine structure of highly cold-shortened muscle has been studied by light and electron microscopy. Tissue at the ‘peak’ of toughness is uniformly shortened and shows no sign of structural weakness or damage, but with further shortening super-contraction and node formation take place in an increasing proportion of the sarcomeres. Widespread fibre fracturing is seen in the internodal zones when the length change has exceeded about 50%. This disruption appears sufficient to account for the declining toughness which accompanies supra-physiological shortening.

    Notizen: Abstract The ability of Flexibacter BH3 to adhere to solid surfaces and to overcome the horizontal drag involved in gliding across the surfaces was considered in terms of the Stefan adhesion principle. The extracellular slime produced by Flexibacter BH3 was suitable as a Stefan adhesive because it exhibited viscous properties characteristic of a linear colloid, increasing the adhesiveness of the bacterium but allowing translational motion across the surface. The water-soluble slime was a glycoprotein, containing glucose, fucose, galactose and some uronic acid. Vesicles and tubules on the outer surface of Flexibacter BH3 possessed trilaminar membranes, contained 2-keto-3-deoxyoctonate (KDO), and showed identity with phenol-extracted lipopolysaccharide (LPS) in gel-diffusion tests. Sections of Flexibacter BH3 gliding on a gold film overlaying an agar medium reveraled a highly convuluted cell envelope outer membrane, portions of which closely conformed to the microcontours of the gold surface. Possible mechanisms of gliding are discussed in relation to this close association with solid surface features, to the finding that flexibility and spiral motion are not essential for gliding, and to evidence revealing the extrusion of slime in advance of “pathfinder” bacteria.

    Notizen: Abstract A technique for observation of surface microlayer bacteria (bacterioneuston) is described, utilizing direct sampling of the air-water interface with carbon-stabilized electron microscope specimen support grids, followed by negative staining and transmission electron microscopy. The method resulted in excellent preservation of forms of microcolonial association, regular surface arrays, surface appendages, and prosthecae in the bacterioneuston of a freshwater pond.

    Notizen: According to Le Chatelier's principle, dehydration and the associated decomposition of hydroxyapatite (HAP) to biodegradable unhydrated calcium phosphates during sintering may be suppressed under a moist sintering atmosphere (thermodynamic effect), or possibly under a pressurized sintering atmosphere (physical effect), by opposing the release of water. The present study explored this possibility. High-purity powdered additives were used to minimize impurity and morphological effects. Al2O3, C, SiC, SiO2, ZrO2, and 316L stainless steel were all trialled at an addition level of 20 vol%. Heat treatment was at 1200°C for 1 h under two experimental atmospheres and two corresponding control atmospheres: flowing H2O/O2 mix—ambient air as a control; pressurized (1 MPa) argon—ambient argon (0.1 MPa) as a control. Specimens were analysed for decomposition by X-ray diffraction (XRD), for densification by porosity measurement, and for microstructural uniformity by energy dispersive spectroscopy (EDS) and image analysis. Significant decomposition occurred under all atmospheres with the exception of flowing H2O/O2 which eliminated decomposition in the HAP-Al2O3, HAP-ZrO2, and HAP-316L systems, and reduced the decomposition levels from near completion to ∼50% in the HAP-SiC and HAP-SiO2 systems. Moistureless pressurization had little effect. Microstructural uniformity was confirmed. No generalized atmosphere-densification interrelationships were observed.

    Notizen: Summary The periodic acid-thiocarbohydrazide-silver proteinate technique and α-amylase digestion were used to locate glycogen in the spermatids and sperms of the bivalve Bankia australis. Glycogen was found in the middle piece and around the acrosome in spermatozoa, apparently randomly scattered throughout the cytoplasm of young spermatids, and in a cytoplasmic bead in old spermatids.
