    Notizen: An experimental demonstration of the interference between two different resonant two-photon, two-color ionization processes from the A 2Σ+, v′=1 state of nitric oxide is presented. This interference effect does not require a well-defined phase relationship between the two ionizing lasers and should, in principle, allow the control of the overall ionization process by small detunings of either or both of the two resonant ionization pathways. This interference effect is quite general, and the potential of this scheme for the control of photoionization and photodissociation partial cross sections is discussed. © 1996 American Institute of Physics.

    Notizen: Several new electronic states in HI have been identified from resonance-enhanced multiphoton ionization (REMPI) spectra in the 69 600–73 600 cm−1 region. These results have been combined with previously unpublished single-photon absorption data and, in selected wavelength regions, with REMPI spectra of DI to provide a more complete description of the excited electronic structure of HI up to ∼9.2 eV above the ground state. © 1995 American Institute of Physics.

    Notizen: Two-color laser-induced grating spectroscopy (TC-LIGS) via the A 2Σ+, v'=0 state has been used to study the B 2Π, v=28 and L 2Π, v=8 valence states and the Q 2Π, v=0 5pπ Rydberg state of NO. Energies for previously unobserved or unresolved transitions to the perturbed L 2Π1/2,3/2, v=8 and Q 2Π1/2,3/2, v=0 states are reported for the first time. The interaction between these states is reconsidered in light of the new assignments. A comparison of different detection methods made for the B 2Π1/2,3/2, v=28←A 2Σ+, v'=0 transitions demonstrates the ability of TC-LIGS to detect states that are difficult to detect by ionization or fluorescence-dip detection. © 1995 American Institute of Physics.

    Notizen: New results on the competition between autoionization and predissociation in the high Rydberg states of nitric oxide are presented. These results provide insight into the earlier work of Park et al. [Phys. Rev. Lett. 76, 1591 (1996)] that shows evidence for substantial mixing between Rydberg series and ionization continua with even and odd orbital angular momenta (l). New data based on fluorescence-dip spectroscopy, detection of neutral predissociation products, and photoelectron spectroscopy suggest that the A′ 2Σ+ and I 2Σ+ dissociative valence states play an important role in this l mixing. Zeeman splittings observed in a magnetic bottle electron spectrometer also result in an improvement in the assignment of these high Rydberg states. © 1998 American Institute of Physics.

    Notizen: During the dry season (early May through September of 1994), following a fall 1993 wildfire, a survey of seedling nodulation was conducted at several sites in the Santa Monica Mountains of Southern California. Seedlings of Ceanothus spinosus, C. megacarpus, C. oliganthus, and C. cuneatus were manually excavated. During this period, only 12 of the 182 seedlings excavated were nodulated, and all of the nodulated seedlings were found in the relatively moist clay soils of a stream bank. No nodules were observed on the 170 seedlings excavated from the drier sites. An irrigation experiment was established in midsummer to assess whether water stress inhibits nodulation of post-fire Ceanothus seedlings. Four plots with numerous seedlings of C. cuneatus and C. spinosus were irrigated with distilled water and monitored over a 9-week period. There was a significant increase in nodulation frequency, water potential, stomatal conductance, transpiration, shoot elongation, and photosynthetic rate of irrigated seedlings compared with adjacent controls. Although these data support the hypothesis that water stress inhibits nodulation. it is unclear whether this is because of an effect of soil moisture on the nodulation capacity of the soils (i.e. on the size and physiological state of the soil Frankia population) or to a host plant response to drought which might prevent actinorhizal root infection and/or nodule development.

    Notizen: Our understanding of the actinorhizal symbiosis, in particular of the Frankia-Ceanothus association, has been hampered by the failure to isolate infective strains in pure culture. Recently, the polymerase chain reaction (PCR) has been utilized to amplify regions of the Frankia genome, allowing analysis of the microsymbiont genome without first isolating the microbe in pure culture. Root nodules were collected from six Ceanothus spp. common to the coastal regions of the Santa Monica Mountains of southern California. Individual lobes were surface-sterilized, total DNA was extracted and amplified using prokaryotic-specific primers. To assess the genetic diversity of Frankia endophytes in the population studied, the BOX primer was used to generate genomic fingerprints of prokaryotic nodule inhabitants using rep-PCR. Fingerprint patterns fell into twelve distinct groups indicating the occurrence of genetic diversity of Frankia in the nodules sampled. DNA extracts of individual lobes that gave distinct BOX-PCR fingerprints were also amplified by PCR using primers directed against conserved regions of the 16S ribosomal RNA gene. The nucleotide sequences of the PCR products were determined and aligned with the corresponding region from other taxa for phylogenetic analysis. The sequences from Ceanothus nodules share a common ancestor to that of the Elaeagnus–infective strains.

    Notizen: X-ray fluorescence microscopy and microspectroscopy with micrometre spatial resolution and unprecedented capabilities for the study of biological and environmental samples are reported. These new capabilities are a result of both the combination of high-brilliance synchrotron radiation and high-performance X-ray microfocusing optics and the intrinsic advantages of X-rays for elemental mapping and chemical-state imaging. In this paper, these capabilities are illustrated by experimental results on hard X-ray phase-contrast imaging, X-ray fluorescence (XRF) imaging and microspectroscopy of mycorrhizal plant roots and fungi in their natural hydrated state. The XRF microprobe is demonstrated by the simultaneous mapping of the elemental distributions of P, S, K, Ca, Mn, Fe, Ni, Cu and Zn with a spatial resolution of approximately 1 × 3 µm and with an elemental sensitivity of approximately 500 p.p.b. Microspectroscopy with the same spatial resolution is demonstrated by recording near-edge X-ray absorption (XANES) spectra of Mn at a concentration of approximately 3 p.p.m.

    Notizen: Abstract Human cytomegalovirus (CMV) encodes a unique serine proteinase that is required in the maturation of the viral capsid. The CMV proteinase can undergo autocatalytic activation and is subject to proteolytic self-inactivation. Mutant enzyme forms were prepared to eliminate the initial autoprocessing site and thus form an active single-chain protein for structure-function studies. Two mutants of CMV proteinase were cloned and expressed inEscherichia coli. The A143V mutant was a conservative substitution at the first internal cleavage site. The S132A mutant modified one of the triad of residues responsible for catalytic activity. Through the use of computer-controlled high-cell-density fermentations the mutant proteins were expressed inE. coli at ∼170mg/L as both soluble (∼40% of total) and inclusion-body forms (∼60% of total). The soluble enzyme was purified by standard methods; inclusion-body protein was isolated by standard methods after refolding and solubilization in guanidine or urea. Sedimentation equilibrium and sedimentation velocity analyses reveal that the enzyme undergoes concentration-dependent aggregation. It exhibits a monomer⇔dimer equilibrium (K d =1ΜM) at low concentrations and remains dimeric at high concentrations (28 mg/ml). Differential scanning calorimetry data for protein thermal unfolding fit best to a non-two-state model with two components (T m =52.3 and 55.3
