    Notizen: Abstract Isolated squid stellate nerves and giant fiber lobes were incubated for 8 hr in Millipore filtered sea water containing [3H]uridine. The electrophoretic patterns of radioactive RNA purified from the axoplasm of the giant axon and from the giant fiber lobe (cell bodies of the giant axon) demonstrated the presence of RNA species with mobilities corresponding to tRNA and rRNA. The presence of labeled rRNAs was confirmed by the behavior of the large rRNA component (31S) which, in the squid, readily dissociates into its two constituent moyeties (17S and 20S). Comparable results were obtained with the axonal sheath and the stellate nerve. In all the electrophoretic patterns, additional species of radioactive RNA migrated between the 4S and the 20S markers, i.e. with mobilities corresponding to presumptive mRNAs. Chromatographic analysis of the purified RNAs on oligo(dT)cellulose indicated the presence of labeled poly(A)+ RNA in all tissue samples. Radioactive poly(A)+ RNA represented approximately 1% of the total labeled RNA in the axoplasm, axonal sheath and stellate nerve, but more than 2% in the giant fiber lobe. The labeled poly(A)+ RNAs of the giant fibre lobe showed a prevalence of larger species in comparison to the axonal sheath and stellate nerve. In conclusion, the axoplasmic RNAs synthesized by the isolated squid giant axon appear to include all the major classes of axoplasmic RNAs, that is rRNA, tRNA and mRNA.

    Notizen: Abstract Rats were long-term morphine-intoxicated by a fluid-diet model ensuring an equal nutrient intake in morphinized and control rats. Uptake of neurotransmitters and D-ala2-met5-enkephalinamide (Enk) was studied in the particulate fractions (obtained at 10,000g) from well defined brain regions of long-term intoxicated and morphine withdrawn rats. In control animals the accumulation of [3H]glutamate and [3H]γ-aminobutyric acid (GABA) reached the highest tissue/medium (T/M) ratio values, 30–120, in the regions studied while monoamine T/M values were 2–10. No active uptake of [3H]Enk could be demonstrated. Striatum showed the most evident modifications in neurotransmitter uptake. In this region the equilibrium T/M ratio for [3H]glutamate and [3H]GABA was significantly lower in intoxicated rats versus controls. Moreover, the T/M ratio for [3H]5-hydroxytryptamine (5-HT) was lower, while that for [3H]dopamine (DA) was higher in abstinent rats in comparison with the controls. [3H]glutamate and [3H]GABA uptakes were also significantly lower, respectively, in frontal cortex, hippocampus and brain stem in intoxicated rats, while [3H]5-HT uptake was significantly lower in hypothalamus after morphine withdrawal. The possible involvement of the endogenous opioid system in the etiology of the alterations is discussed.

    Notizen: Abstract The labeling of proteins in the particulate subcellular fractions and in the cell sap was studied in the cerebral cortex of rabbits 24 hours after a single electroconvulsive shock (ECS). To this end the animals were injected with [3H]valine subarachnoidally and sacrificed 30 minutes later. The incorporation of the labeled aminoacid into proteins was slightly increased in all subcellular fractions but the increase was significant only for soluble proteins. Referring to the type of soluble polypeptides found in the cortex at this post ECS phase, no modification was found in the SDS electrophoresis pattern of steady state proteins. The fractionation pattern of labeled soluble proteins showed in one third (3 out of 9) of the experiments a remarkable stimulation of the synthesis of two polypeptides (25,000 and 54,000 Dalton MW), which were not labeled in the controls.

    Notizen: Abstract The reduction to the micro-scale of a recently described electrophoretic method for the evaluation of GABA catabolism by GABA-T is presented. The micromethod involves the electrophoresis, in 1 mm diam. capillaries, of small samples of mixtures of [14C]GABA and its metabolites. By coupling this procedure to previously devised micromethods, it was possible to evaluate GABA-T attack to14C labeled GABA diffusing across a single microdissected neuronal membrane.

    Notizen: Abstract The effect of preincubation of brain homogenates on the subsequent uptake of [3H]GABA was studied in rat thalamus and corpus striatum. The results show that in both brain regions preincubation causes a decrease in the initial rate of [3H]GABA influx, the phenomenon being most evident in the thalamus.
